Psychiatric symptoms including psychosis and anxiety-like states have been linked to methamphetamine (MA) use, with severity of psychiatric symptoms has been correlated to duration of use. Prolonged psychiatric residual symptoms have been observed and reported, but there have been no studies that have examined residual effects beyond 12 months of abstinence. There are no studies to date which focus on anger, anxiety, and perception of stress as residual effects following 12 months of abstinence. Arsenic and cadmium (Cd2+) are environmental carcinogens that have been implicated in the development of bladder cancer. However, there are no good in vitro model systems to study the development of bladder cancer caused by these carcinogens. Previous studies from this laboratory have shown that arsenite (As3+) and Cd2+ exposure can cause direct malignant transformation of an immortalized human urothelial cell line, the UROtsa. In the present study the repeatability of the transformation process was examined by isolating and characterizing 6 additional Cd2+- transformed lines and 5 additional As3+- transformed cell lines. Alzheimer Disease (AD) is a progressive, debilitating and the most prevalent neurodegenerative disorder affecting 5.5 million people and the sixth leading cause of fatality in the United States. The etiology of AD is not understood and clinically FDA-approved drugs offer only symptomatic relief for a short period of time and in only a subset of AD patients. AD is histopathologically characterized by the presence of extracellular deposits of aggregated Amyloid β (Aβ) peptide as plaques and the intracellular deposition of aggregated hyperphosphorylated tau protein as neurofibrillary tangles (NFT). The dopamine transporter (DAT) is an integral membrane phosphoprotein found on dopaminergic neurons that mediates the presynaptic reuptake of dopamine and is a site of action for psychostimulants. Numerous DAT functions including efflux, uptake activity, and surface expression are regulated by protein kinase C (PKC) through unknown mechanisms. Basal and PKC stimulated phosphorylation of DAT occurs on multiple serines in the distal N-terminus, but the exact sites modified and the kinases involved are unknown. To address these issues, we examined in vitro phosphorylation of a recombinant DAT N-terminal tail peptide (NDAT). We found that NDAT is phosphorylated at multiple sites …

Aminophosphines, diphosphines and related chelating P-containing bidentate ligands, due to their versatile structural features, are considered promising enantioselective catalyst alternatives to symmetric and monodentate phosphine ligands. There are several methods for preparation of such P,N-ligands; however, they are mostly limited to specific cases. In this dissertation, the development of a new general method for synthesis of P-containing bidentate ligands is described. The approach studied is based on using reactions of cyclopalladated complexes (CPCs) with LiPPh2, KPPh2 and HPPh2. This study is the first example of a systematic investigation of reactions of CPCs at a C–Pd bond with phosphorus nucleophiles. A …

We used X-ray fluorescence (XRF) to compare the heavy metal chemistry of sediments in caves in rural and urban areas to the chemistry of sediments from a control cave in a relatively undisturbed watershed in the Springfield (MO) Plateau. Sediment from Smallin Cave near Ozark, MO, the control cave, has the smallest peak sizes for Zn and Mn and a moderately-sized Pb peak. Sediment from the rural cave exhibited larger peaks of Zn and Mn and a smaller Pb peak. Sediment from the urban cave had the largest Zn, Mn and Pb peaks. Excess fine sediment from human activity is a major pollutant to streams across the U.S.; however, distinguishing human-induced sedimentation from natural fine sediment is complex. The U.S. Environmental Protection Agency recently implemented a protocol for the quantitative field assessment of human-induced sedimentation using measurements of stream geomorphology. Macroinvertebrate community composition, streambed sediment stability, and sediment composition were studied at 49 sites in the James River watershed in central Virginia. Sediment composition was found to be a stronger driver of community composition than sediment stability. Although I was not able to show that macroinvertebrate metrics were related to sediment stability independently …

Using TLR pathways, primary human cytomegalovirus (HCMV) induces innate responses including the production of inflammatory cytokines. Mounting evidence suggests that LPS recognition by TLR4/MD2/CD14 results in differential utilization of TIRAP-TRAF6 and TRAM-TRIF signaling, thereby leading to transcriptional activation of various cytokine genes. However, relative roles of the TLR4/MD2/CD14 complex and its adaptor proteins TIRAP and TRAM involved in regulating monocyte responses to HCMV are incomplete. Here, we provided evidence supporting the notion that the TLR4/MD2/CD14 complex contributes notably to HCMV-induced signaling and subsequent cytokine production in monocytes. Eukaryotic cells make many types of primary and processed RNAs that are found either in specific subcellular compartments or throughout the cells. A complete catalogue of these RNAs is not yet available and their characteristic subcellular localizations are also poorly understood. Because RNA represents the direct output of the genetic information encoded by genomes and a significant proportion of a cell’s regulatory capabilities are focused on its synthesis, processing, transport, modification and translation, the generation of such a catalogue is crucial for understanding genome function. During nervous system development, various diffusing biomolecules play a critical role in neurite outgrowth and guidance, resulting in the formation of a complex circuitry. However, the precise mechanism by which these molecules are spatio-temporally delivered to the growing tip of the neurites is unclear. Overall, the results suggest to the utility of this device for studying neurobiology, tissue engineering and cell migration.

Glycans, especially, cell surface glycans acting as receptors, are involved in a wide range of biological processes. Analysis of the cell surface glycans provides a basis for understanding the molecular mechanism of glycan-mediated biological process. The present method can be applied in derivatizing sugars from natural sources for glycan sensor and glyco-functionalization applications.

Cyclooxygenase-2 (COX-2) inhibitor nimesulide inhibits the proliferation of various types of cancer cells mainly via COX-2 independent mechanisms, which makes it a good lead compound for anti-cancer drug development. A series of new nimesulide analogs were evaluated with cell proliferation assay based on a non-small lung cancer cell line H292. The results showed that several derivatives were very active to against H292 cell growth with IC50s of sub nano mole. These results suggest the possibility of using these nimesulide derivatives as chemo preventive agents.

Meiosis is one type of cell division that produces germ cells. In Meiosis I, the DNA in a diploid cell is doubled and homologous chromosomes separate. In Meiosis II, each pair of sister chromatids are separated, resulting in four haploid daughter cells. The nine intervals were analyzed under several conditions and the number, timing, and distribution of crossovers was determined. Results and their compatibility with current models of synaptonemal complex functions will be discussed.

In human, the majority of protein coding genes are interrupted by dispensable intervening sequences (introns). These introns are removed by nuclear precursor (pre) mRNA splicing process to produce a mature mRNA needed for productive protein production in the cell. We are studying the splicing of minor class or U12-type introns which are spliced by U11, U12, U4atac, U5 and U6atac snRNAs. U11 snRNA binds to the 5’ end or splice site of the intron by RNA-RNA base-pairing to initiate the splicing process. The primary objective of our research was to assess population size and range of Ligumia nasuta, the Eastern Pond mussel in the Cuyahoga River watershed. We conclude that the Eastern Pond mussel may be all but gone from the East Branch and West Branch Cuyahoga Rivers, but may persist in better numbers in Portage County. Further studies are required before the sustainability of this species can be fully assessed.
