Gloeobacter violaceus, the earliest diverging oxyphotobacterium (cyanobacterium) on the 16S ribosomal RNA tree, has five copies of the photosystem II psbA gene encoding the D1 reaction center protein subunit. These copies are widely distributed throughout the 4.6 Mbp genome with only one copy colocalizing with other PSII subunits, in marked contrast to all other psbA genes in all publicly available sequenced genomes. A clustering of two other psb genes around psbA3 (glr2322) is unique to Gloeobacter. We provide experimental proof for the transcription of a psbA3DC operon, encoding three of the five reaction center core subunits (D1, D2, and CP43). …

Subordinate males, which are excluded from reproduction often save energy by reducing their investment in sperm production. However, if their position in a dominance hierarchy changes suddenly they should also rapidly attain fertilization capability. Here, we asked how social suppression and ascension to dominance influences sperm quality, spermatogenesis and reproductive competence in the cichlid Astatotilapia burtoni, where reproduction is tightly coupled to social status. Dominant territorial (T) males are reproductively active while subordinate non-territorial (NT) males are suppressed, but given the opportunity, NT males will perform dominance behaviours within minutes and attain T male testes size within days. Using the …
The Phylogenetic Distribution And Ecological Role Of Carbon Monoxide Oxidation In The Genus Burkholderia, Carolyn F. Weber, Gary M. King
The Phylogenetic Distribution And Ecological Role Of Carbon Monoxide Oxidation In The Genus Burkholderia, Carolyn F. Weber, Gary M. King
Faculty Publications
Burkholderia is a physiologically and ecologically diverse genus that occurs commonly in assemblages of soil and rhizosphere bacteria. Although Burkholderia is known for its heterotrophic versatility, we demonstrate that 14 distinct environmental isolates oxidized carbon monoxide (CO) and possessed the gene encoding the catalytic subunit of form I CO dehydrogenase (coxL). DNA from a Burkholderia isolate obtained from a passalid beetle also contained coxL as do the genomic sequences of species H160 and Ch1-1. Isolates were able to consume CO at concentrations ranging from 100 ppm (vol/vol) to sub-ambient (< 60 ppb (vol/vol)). High concentrations of pyruvate inhibited CO uptake (> 2.5 mM), but mixotrophic consumption of CO and pyruvate occurred …

Limited Potential For Adaptation To Climate Change In A Broadly Distributed Marine Crustacean, Morgan W. Kelly, Eric Sanford, Richard K. Grosberg

The extent to which acclimation and genetic adaptation might buffer natural populations against climate change is largely unknown. Most models predicting biological responses to environmental change assume that species' climatic envelopes are homogeneous both in space and time. Although recent discussions have questioned this assumption, few empirical studies have characterized intraspecific patterns of genetic variation in traits directly related to environmental tolerance limits. We test the extent of such variation in the broadly distributed tidepool copepod Tigriopus californicus using laboratory rearing and selection experiments to quantify thermal tolerance and scope for adaptation in eight populations spanning more than 178 of …

Recruitment And Biological Consequences Of Histone Modification Of H3k27me3 And H3k9me3., Joomyeong Kim, Hana Kim
Faculty Publications
Two histone marks, H3K27me3 and H3K9me3, are well known for their repressive roles in the genic and nongenic regions of metazoan genomes. Several protein complexes are known to be responsible for generating these marks, including polycomb repression complex 2 and several H3K9 methylases. Recent studies have shown that the targeting of these histone-modifying complexes within mammalian genomes may be mediated through several DNA-binding proteins, including AEBP2, JARID2, and YY1. In this review, we discuss the potential targeting mechanisms in light of the recent results that have been derived from genome-wide chromatin immunoprecipitation sequencing data and the in vivo functions of …

Oyun: A New, Free Program For Iterated Prisoner’S Dilemma Tournaments In The Classroom, Charles H. Pence, Lara Buchak
Faculty Publications
Evolutionary applications of game theory present one of the most pedagogically accessible varieties of genuine, contemporary theoretical biology. We present here Oyun (oy-oon, http://charlespence.net/oyun), a program designed to run iterated prisoner's dilemma tournaments, competitions between prisoner's dilemma strategies developed by the students themselves. Using this software, students are able to readily design and tweak their own strategies, and to see how they fare both in round-robin tournaments and in “evolutionary” tournaments, where the scores in a given “generation” directly determine contribution to the population in the next generation. Oyun is freely available, runs on Windows, Mac, and Linux computers, …

In this review we examine the structure and function of the extrinsic proteins of Photosystem II. These proteins include PsbO, present in all oxygenic organisms, the PsbP and PsbQ proteins, which are found in higher plants and eukaryotic algae, and the PsbU, PsbV, CyanoQ, and CyanoP proteins, which are found in the cyanobacteria. These proteins serve to optimize oxygen evolution at physiological calcium and chloride concentrations. They also shield the Mn 4CaO 5 cluster from exogenous reductants. Numerous biochemical, genetic and structural studies have been used to probe the structure and function of these proteins within the photosystem. We will …

Xiphophorus fishes are well-established models for biomedical research of spontaneous or induced tumors, and their use in research dates back to the 1930s. Currently, 58 well-pedigreed lines exist among 24 Xiphophorus species housed as live animals at the Xiphophorus Genetic Stock Center. The technique of sperm cryopreservation has been applied to preserve these valuable genetic resources, and production of offspring has been reported with cryopreserved sperm in two species (X. helleri and X. couchianus). The goal of this research was to establish protocols for sperm cryopreservation and artificial insemination that yield live young in X. maculatus, a widely used research …

We report the presence of arbuscular mycorrhizae (AM) in Spartina alterniflora Loisel roots. Roots were sampled for AM in field-collected and greenhouse-maintained Spartina patens (Aiton) Muhl. and S. alterniflora, the dominant species of Louisiana's brackish and saline marshes, respectively. Previous reports of AM association in these Spartina sp. are limited and conflicting. Field-collected 5. alterniflora had minimal AM (2.4%), whereas 39.5% of the 5. patens roots examined were AM colonized. Greenhouse conditions of reduced salinity [3 parts per thousand (ppt)] appeared to increase AM association for 5. patens compared with field samples. AM occurrence varied significantly among the three sample …

This study characterized lateral root (LR) development attributes during the onset of storage root (SR) initiation stage in 'Beauregard' sweetpotato. SR initiation has been defined as the appearance of cambia around the protoxylem and secondary xylem elements. Our results showed that 20-day-old adventitious roots (ARs) classified as SRs had 53% and 85% greater mean LR count than pencil roots (PRs) and lignified roots (LGs), respectively. SRs had 53% and 78% greater mean LR density relative to PRs and LGs, respectively. SRs had 66% and 130% greater mean total LR length than PRs and LGs, respectively. SRs had lower mean main …

Host Interferon-Γ Inducible Protein Contributes To Brucella Survival., Jennifer A. Ritchie, Adam Rupper, James A. Cardelli, Bryan H. Bellaire
Faculty Publications
Brucella spp. are highly adapted intracellular pathogens of mammals that cause chronic infections while surving and replicating in host monocytes and macrophages. Although monocytes are normally susceptible to infection, pretreatment with pro-inflammatory cytokine interferon-γ (IFN-γ) activates cellular defense mechanisms that increase intracellular killing of Brucella and prevents bacterial replication. We examined the contribution of the IFN-γ inducible GTPase, LRG-47, to B. abortus 2308 infection in in vitro and in vivo murine models. Infecting non-activated macrophages from LRG-47(-/-) mice revealed that loss of this host protein negatively effected the intracellular survival and replication of IgG opsonized B. abortus. In contrast, survival …

Rice yield is greatly affected by environmental stresses such as drought and salinity. In response to the challenge of producing rice plants tolerant to these stresses, we introduced cDNA encoding the transcription factors DREB1A and DREB1B under the control of the stress inducible rd29 promoter. Two different indica rice cultivars were used, BR29, an improved commercially cultivated variety from Bangladesh and IR68899B, an IRRI bred maintainer line for hybrid rice. Agrobacterium mediated transformation of BR29 was done independently with DREB1A isolated from rice and Arabidopsis and DREB1B isolated from rice, whereas biolistic transformation was done with rice- DREB1B in the …

This study corroborated the findings of Lee et al. (2008) and relied on culture-independent techniques, particularly 16SrDNA sequencing. Results from the study demonstrated that ants in captivity completely change their gut bacterial diversity strongly supports the conclusion that fire ant gut bacteria are environmental in origin. Bacteria in the fourth-instar larvae were present because they were consumed with food sources. From molecular data, a differing list of bacteria from each colony with different branching and clustering among bacterial clades in the phylogenetic trees were found. Different bacterial diversity and evenness was found in all colonies, which provided evidence against the …

Antimicrobial Activity, Cytotoxicity And Phytochemical Analyses Of Rhus Aromatica, Rhus Glabra And Sanguinaria Canadensis Native To South Dakota, Gitanjali Nandakafle
Electronic Theses and Dissertations
A total of twenty five South Dakota native medicinal plants were screened against enterotoxigenic Escherichia coli using a disk diffusion assay. Out of these 25 plants, three plants Rhus aromatica, Rhus glabra and Sanguinaria canadensis demonstrated the highest bacterial inhibition. The minimum inhibitory concentrations (MIC) of these plant extracts were determined using a bacterial enumeration assay. Alamar Blue in vitro cytotoxicity test showed that all extracts have some toxic effect to porcine intestinal epithelial cell lines. Phytochemical analysis of these plant extracts indicated the presence high concentration of flavonoids and condensed tannins in R. aromatica and that R. glabra contained …

Beyond Priapus: A Call For A Feminist And/Or Queer Theory Archaeology Of Roman Masculinity And Phallic Iconography, Ashley J. Barnett
Beyond Priapus: A Call For A Feminist And/Or Queer Theory Archaeology Of Roman Masculinity And Phallic Iconography, Ashley J. Barnett
Nebraska Anthropologist
Phallic iconography is ubiquitous throughout the archaeology of the Roman world and has raised questions about sexuality in ancient Rome. Dominant modern Western discourses privilege heterosexual male frameworks which many not adequately correspond to ancient Roman culture. Thus, the application of Feminist and/or Queer theory may expand academic insight into not only Roman sexuality, but also into the power and politics of the Republican Period.
Justice In Context: Assessing Contextualism As An Approach To Justice, Michael Buckley
Justice In Context: Assessing Contextualism As An Approach To Justice, Michael Buckley
Publications and Research
Moral and political philosophers are increasingly using empirical data to inform their normative theories. This has sparked renewed interest into questions concerning the relationship between facts and principles. A recent attempt to frame these questions within a broader approach to normative theory comes from David Miller, who has on several occasions defended ‘contextualism’ as the best approach to justice. Miller argues that the context of distribution itself brings one or another political principle into play. This paper examines this claim. It considers several plausible strategies for carrying out Miller’s general project and argues that each strategy fails. Nevertheless, the author …
