BACKGROUND: Salmonella enterica serovar Typhi (S. Typhi) is strictly a human intracellular pathogen. It causes acute systemic (typhoid fever) and chronic infections that result in long-term asymptomatic human carriage. S. Typhi displays diverse disease manifestations in human infection and exhibits high clonality. The principal factors underlying the unique lifestyle of S. Typhi in its human host during acute and chronic infections remain largely unknown and are therefore the main objective of this study. METHODOLOGY/PRINCIPAL FINDINGS: To obtain insight into the intracellular lifestyle of S. Typhi, a high-throughput phenotypic microarray was employed to characterise the catabolic capacity of 190 carbon sources …

In recent years, the removal of nitrate as a global soil and water contaminant has been increasingly considered. Various methods have been applied to remove this inorganic pollutant from water and wastewater. Among them is an integrated " bio-electrochemical reactor" system, which is a novel method for water and wastewater denitrification. The novelty of this system is improved biological denitrification by immobilizing autohydrogenotrophic bacteria directly on the surface of a cathode, providing easy access to NO 3 - and H 2 as the electron acceptor and electron donor, respectively. The system effectiveness depends on the configuration of reactor, and the …

Phosphomannose isomerase (pmi) gene isolated from Escherichia coli allows transgenic plants carrying it to convert mannose-6- phosphate (from mannose), a carbon source that could not be naturally utilized by plants into fructose-6-phosphate which can be utilized by plants as a carbon source. This conversion ability provides energy source to allow the transformed cells to survive on the medium containing mannose. In this study, four transformation vectors carrying the pmi gene alone or in combination with the beta-glucuronidase (gusA) gene were constructed and driven by either the maize ubiquitin (Ubi1) or the cauliflower mosaic virus (CaMV35S) promoter. Restriction digestion, PCR amplification …

Among enteric pathogens, Salmonella enterica serovar Typhi is responsible for the largest number of food-borne outbreaks and fatalities. The ability of the pathogen to cause systemic infection for extended durations leads to a high cost of disease control. Chronic carriers play important roles in the evolution of Salmonella Typhi; therefore, identification and in-depth characterization of isolates from clinical cases and carriers, especially those from zones of endemicity where the pathogen has not been extensively studied, are necessary. Here, we describe the genome sequence of the highly virulent Salmonella Typhi strain BL196/05 isolated during the outbreak of typhoid in Kelantan, Malaysia, …

Factor VIII (FVIII) functions as a cofactor for factor IXa in the contact coagulation pathway and circulates in a protective complex with von Willebrand factor (VWF). Plasma FVIII activity is strongly influenced by environmental and genetic factors through VWF-dependent and independent mechanisms. Single nucleotide polymorphisms (SNPs) of the coding and promoter sequence in the FVIII gene have been extensively studied for effects on FVIII synthesis, secretion, and activity, but impacts of non–disease-causing intronic SNPs remain largely unknown. We analyzed FVIII SNPs and FVIII activity in 10 434 healthy Americans of European (EA) or African (AA) descent in the Atherosclerosis Risk …

Small- and intermediate-conductance Ca(2+)-activated potassium channels, activated by Ca(2+)-bound calmodulin, have an important role in regulating membrane excitability. These channels are also linked to clinical abnormalities. A tremendous amount of effort has been devoted to developing small molecule compounds targeting these channels. However, these compounds often suffer from low potency and lack of selectivity, hindering their potential for clinical use. A key contributing factor is the lack of knowledge of the binding site(s) for these compounds. Here we demonstrate by X-ray crystallography that the binding pocket for the compounds of the 1-ethyl-2-benzimidazolinone (1-EBIO) class is located at the calmodulin-channel interface. …

Currently more than 40 million people worldwide are suffering from HIV/AIDS. Despite the billions of dollars that have been put into research and development of HIV drugs, a cure has not been discovered. Protease inhibitors are a class of drugs that slow the progression of the disease. This research aims to create five boronated amino acid analogs that will be used in future synthesis of potential HIV-1 protease inhibitors. The purpose of including these amino acid analogs in potential inhibitors is to increase drug bioavailability by mimicking an amino acid structure. The amino acid analog also may help to create …

The goal of this research was to find compounds with the potential for use as medicines. Many microorganisms use a mechanism known as active efflux to move antibiotics out of cells. As a result, the effectiveness of antibiotics may be partially or completely reduced allowing the pathogen to survive. A major theme of this research was to look for compounds in plants that inhibit active efflux processes and that are related to multi-drug resistance. After the diffusion of an antibiotic into a bacterial cell, an efflux pump inhibitor may prevent the antibiotic from being expelled from the organism, leading to …

We have discovered an Aleuria Aurantia Lectin (AAL)-reactive immunoglobulin G (IgG) that naturally occurs in the circulation of rabbits and mice, following immune responses induced by various foreign antigens. AAL can specifically bind to fucose moieties on glycoproteins. However, most serum IgGs are poorly bound by AAL unless they are denatured or treated with glycosidase. In this study, using an immunogen-independent AAL-antibody microarray assay that we developed, we detected AAL-reactive IgG in the sera of all animals that had been immunized 1-2 weeks previously with various immunogens with and without adjuvants and developed immunogen-specific responses. All of these animals subsequently …

B-cell lymphoma 2 (Bcl-2) homology 3 (BH3)-only proteins represent a class of pro-apoptotic factors that neutralize pro-survival Bcl-2 proteins, and, in some cases, directly activate Bax. The mechanisms of control and the role of BH3-only proteins, such as Bcl-2 like protein 11 extra large and Bad are well studied. By contrast, relatively little is known about the regulation and role of Bcl-2 modifying factor (Bmf). The B-RAF oncogene is mutated in ∼8% of human tumors. We have previously shown that Bmf is upregulated at the transcript level and is required for apoptosis induced by targeting B-RAF signaling in tumor cells …

Gesture Use By Free-Living Chimpanzees (Pan Troglodytes) Related To Partner Attentional State, Glee Larsen
Undergraduate Honors Theses
Both in the wild and in captivity, chimpanzees (Pan troglodytes) use gesture to communicate with each other. The three modalities of gesture that chimpanzees use are auditory, tactile, and visual. Captive chimpanzees use gesture modalities appropriately according to the communicative partner's attentional state. A communicative partner is the target of the communicator's actions. A partner may be either paying attention (attending) or not paying attention (inattentive) to the chimpanzee who is attempting to communicate with them. The present study investigated whether a free-living population of chimpanzees use gesture modality appropriately to the partner's attentional state. Visual gestures depend …
