The Molecular Machinery Critical To The Degradation Of Cellular Rna, Brad J. Schmier Mar 2012

The Molecular Machinery Critical To The Degradation Of Cellular Rna, Brad J. Schmier

Open Access Dissertations

Exoribonucleases are indispensable for cellular RNA metabolism. RNA processing, end-turnover, and degradation all require the concerted action of exoribonucleases. In this thesis, two families of exoribonucleases that act in the final steps of RNA decay pathways are explored. The first of these is the RNR superfamily of processive 3’→5’ RNases with major roles in both mRNA and stable RNA degradation. The initial focus of this work is the structural and enzymatic characterization of an unusual RNR family enzyme from the radiation-resistant bacterium Deinococcus radiodurans. This enzyme is demonstrated biochemically to be an RNase II-type enzyme (DrII), based on its sensitivity …

A Pathway Idea For Model Building, A.M. Mathai, Panagis Moschopoulos

Journal of Statistics Applications & Probability

Models, mathematical or stochastic, which move from one functional form to another through pathway parameters, so that in between stages can be captured, are examined in this article. Models which move from generalized type-1 beta family to type-2 beta family, to generalized gamma family to generalized Mittag-Leffler family to Levy distributions are examined here. It is known ´ that one can likely find an approximate model for the data at hand whether the data are coming from biological, physical, engineering, social sciences or other areas. Different families of functions are connected through the pathway parameters and hence one will find …

A Compound Class Of Poisson And Lifetime Distributions, Said Alkarni, Aiman Oraby

Journal of Statistics Applications & Probability

A new lifetime class with decreasing failure ratewhich is obtained by compounding truncated Poisson distribution and a lifetime distribution, where the compounding procedure follows same way that was previously carried out by Adamidis and Loukas(1998).A general form of probability, distribution, survival and hazard rate functions as well as its properties will be presented for such a class. Thisnew class of distributions generalizes several distributions which have been introduced and studied in the literature.

Phosphatidylserine Colocalizes With Epichromatin In Interphase Nuclei And Mitotic Chromosomes, Igor Prudovsky, Calvin P.H. Vary, Yolanda Markaki, Ada L. Olins, Donald E. Olins Mar 2012

Phosphatidylserine Colocalizes With Epichromatin In Interphase Nuclei And Mitotic Chromosomes, Igor Prudovsky, Calvin P.H. Vary, Yolanda Markaki, Ada L. Olins, Donald E. Olins

Pharmaceutical Sciences Faculty Publications

Cycling eukaryotic cells rapidly re-establish the nuclear envelope and internal architecture following mitosis. Studies with a specific anti-nucleosome antibody recently demonstrated that the surface (“epichromatin”) of interphase and mitotic chromatin possesses a unique and conserved conformation, suggesting a role in postmitotic nuclear reformation. Here we present evidence showing that the anionic glycerophospholipid phosphatidylserine is specifically located in epichromatin throughout the cell cycle and is associated with nucleosome core histones. This suggests that chromatin bound phosphatidylserine may function as a nucleation site for the binding of ER and re-establishment of the nuclear envelope.

Toward A More Phronetic Leisure Science, Daniel Dustin, Keri Schwab, Jeff Rose Mar 2012

Toward A More Phronetic Leisure Science, Daniel Dustin, Keri Schwab, Jeff Rose

Recreation, Parks and Tourism Administration

In this essay, we examine the assumptions underlying natural science, social science, and the humanities. More specifically, we suggest that social science in general and leisure science in particular be guided by a different set of assumptions than those guiding natural science and the humanities. Drawing on the Aristotelian idea of phronesis, we propose that value rationality more so than instrumental rationality guide social scientific inquiry, and that social science in general, and leisure science in particular, be viewed as a bridge between natural science and the humanities.

Evaluation Of Urinary Pesticide Biomarkers Among A Sample Of The Population In The United States, Alex Lance Lebeau Mar 2012

Evaluation Of Urinary Pesticide Biomarkers Among A Sample Of The Population In The United States, Alex Lance Lebeau

USF Tampa Graduate Theses and Dissertations

Pesticide use in the United States continues to attract negative public attention. In recent years, this attention has focused on the effects that chronic, low-level pesticides may have, especially on children and various sub-populations. Over the past decade, studies have attempted to correlate negative health effects with detections of pesticide biomarkers in biological media. The current research investigates biomarker of exposure levels in a sample of the United States population. Data from the 2001-2002 NHANES dataset (n=11,039) was evaluated. The detection frequency of urinary biomarkers of exposure and the geometric mean from the NHANES pesticide dataset (n=3,152) were determined. Of …

Poplar Gtl1 Is A Ca2+/Calmodulin-Binding Transcription Factor That Functions In Plant Water Use Efficiency And Drought Tolerance., Hua Weng, Chan Yul Yoo, Michael J. Gosney, Paul M. Hasegawa, Michael V. Mickelbart

Department of Horticulture & Landscape Architecture Faculty Publications

Diminishing global fresh water availability has focused research to elucidate mechanisms of water use in poplar, an economically important species. A GT-2 family trihelix transcription factor that is a determinant of water use efficiency (WUE), PtaGTL1 (GT-2 like 1), was identified in Populus tremula × P. alba (clone 717-IB4). Like other GT-2 family members, PtaGTL1 contains both N- and C-terminal trihelix DNA binding domains. PtaGTL1 expression, driven by the Arabidopsis thaliana AtGTL1 promoter, suppressed the higher WUE and drought tolerance phenotypes of an Arabidopsis GTL1 loss-of-function mutation (gtl1-4). Genetic suppression ofgtl1-4 was associated with increased stomatal density …

Structural Insights Into Initial And Intermediate Steps Of The Ribosome-Recycling Process, Takeshi Yokoyama, Tanvir R. Shaikh, Nobuhiro Iwakura, Hideko Kaji, Akira Kaji, Rajendra K. Agrawal Mar 2012

Structural Insights Into Initial And Intermediate Steps Of The Ribosome-Recycling Process, Takeshi Yokoyama, Tanvir R. Shaikh, Nobuhiro Iwakura, Hideko Kaji, Akira Kaji, Rajendra K. Agrawal

Department of Biochemistry and Molecular Biology Faculty Papers

The ribosome recycling factor (RRF) and elongation factor G (EF-G) disassemble the 70S post-termination complex (PoTC) into mRNA, tRNA, and two ribosomal subunits. We have determined cryo-EM structures of the PoTC•RRF complex, with and without EF-G. We find that domain II of RRF initially interacts with universally conserved residues of the 23S rRNA helices 43 and 95, and protein L11 within the 50S ribosomal subunit. Upon EF-G binding, both RRF and tRNA are driven toward the tRNA-exit (E) site, with a large rotational movement of domain II of RRF toward the 30S ribosomal subunit. During this intermediate step of the …

Oxidative stress-related damage to the DNA macromolecule produces lesions that are implicated in various diseases. To understand damage to DNA, it is important to study the free radical reactions causing the damage. Measurement of DNA damage has been a matter of debate as most of the available methods measure the end product of a sequence of events and provide limited information on the initial free radical formation. We report a measurement of free radical damage in DNA induced by a Cu(II)-H2O2 oxidizing system using immuno-spin trapping supplemented with electron paramagnetic resonance. In this investigation, the short-lived radical generated is trapped …

Adolescent Girls' Experience Of Binge And Loss Of Control Eating, Allison Palmberg Mar 2012

Adolescent Girls' Experience Of Binge And Loss Of Control Eating, Allison Palmberg

Theses and Dissertations

The current investigation used qualitative methodology to examine adolescent girls’ perceptions of control over their eating, as well as triggers, and consequences of binge and related eating behaviors. Focus groups were completed with 19 adolescent girls (aged 13-17, 58% African American, 41% White) who endorsed the behaviors. Responses to focus group questions were qualitatively analyzed using a grounded theory approach and constant comparison coding. Results reflected a fundamental lack of awareness of the loss of control (LOC) eating behaviors. Yet, the data did reflect a central theme of the need to affirm independence and autonomy through eating behaviors via three …
