Artemisinin-based combination therapies are the most effective drugs to treat Plasmodium falciparum malaria. Reduced sensitivity to artemisinin monotherapy, coupled with the emergence of parasite resistance to all partner drugs, threaten to place millions of patients at risk of inadequate treatment of malaria. Recognizing the significance and immediacy of this possibility, the Fogarty International Center and the National Institute of Allergy and Infectious Diseases of the U.S. National Institutes of Health convened a conference in November 2010 to bring together the diverse array of stakeholders responding to the growing threat of artemisinin resistance, including scientists from malarious countries in peril. This …

A Multi-Beach Study Of Staphylococcus Aureus, Mrsa, And Enterococci In Seawater And Beach Sand, Kelly D. Goodwin, Melody Mcnay, Yiping Cao, Darcy Ebentier, Melissa Madison, John F. Griffith

United States Department of Commerce: Staff Publications

Incidences of Staphylococcus aureus and methicillin resistant S. aureus (MRSA) have risen worldwide prompting a need to better understand routes of human exposure and whether standard bacterial water quality monitoring practices adequately account for this potential threat. Beach water and sand samples were analyzed during summer months for S. aureus, enterococci, and MRSA at three southern California beaches (Avalon, Doheny, Malibu Surfrider). S. aureus frequently was detected in samples of seawater (59%, n = 328) and beach sand (53%, n = 358). MRSA sometimes was detected in seawater (1.6%, n = 366) and sand (2.7%, n = 366) at …

Use Of Real-Time Pcr To Detect Canine Parvovirus In Feces Of Free-Ranging Wolves, L. David Mech, Emily S. Almberg, Douglas Smith, Sagar Goyal, Randall S. Singer

United States Geological Survey, Northern Prairie Wildlife Research Center: Publications

Using real-time PCR, we tested 15 wolf (Canis lupus) feces from the Superior National Forest (SNF), Minnesota, USA, and 191 from Yellowstone National Park (YNP), USA, collected during summer and 13 during winter for canine parvovirus (CPV)-2 DNA. We also tested 20 dog feces for CPV-2 DNA. The PCR assay was 100%sensitive and specific with a minimum detection threshold of 104 50% tissue culture infective dose. Virus was detected in two winter specimens but none of the summer specimens. We suggest applying the technique more broadly especially with winter feces.

From Protozoan Infection In Monarch Butterflies To Colony Collapse Disorder In Bees Are Emerging Infectious Diseases Proliferating In The Insect World?, Rebecca Bartel, Sonia Altizer

United States Fish and Wildlife Service: Publications

In the late 1990S, scientists first noticed mysterious declines in several wild bumble bee species (Bombus spp.) in both eastern and western North America, with one species now possibly extinct (Colla and Packer 2008). These bumble bee losses mirrored declines in the abundance of many other native pollinators (National Research Council 2007). Interestingly, the timing of bumble bee declines in the United States coincided with reports of disease outbreaks in commercial- reared bumble bees sold for use in the production of greenhouse tomatoes and peppers (Evans et al. 2008). This observation, together with reports of a higher incidence of …

Marine Biotoxins: Emergence Of Harmful Algal Blooms As Health Threats To Marine Wildlife, Spencer E. Fire, Frances M. Van Dolah Jan 2012

Marine Biotoxins: Emergence Of Harmful Algal Blooms As Health Threats To Marine Wildlife, Spencer E. Fire, Frances M. Van Dolah

United States Department of Commerce: Staff Publications

Harmful algal blooms (HABs) affect aquatic ecosystems around the world, adversely affecting marine animal and human health, coastal ecosystem integrity, and economies that depend on coastal resources. Shellfish poisoning events involving humans who had ingested bivalves contaminated with HAB toxins primarily drove early scientific and social interest in HABs. More recently, research efforts have shown that HABs are often temporally and spatially correlated with the occurrence of acute morbidity or mortality of marine animals (Landsberg et al. 2005), and to date at least four classes of algal toxins have been associated with such events. Although fish, seabirds, and many other …

Nebraska Center for Virology: Faculty Publications

Understanding the evolution of the human immunodeficiency virus type 1 (HIV-1) envelope during disease progression can provide tremendous insights for vaccine development, and simian-human immunodeficiency virus (SHIV) infection of nonhuman primate provides an ideal platform for such studies. A newly developed clade C SHIV, SHIV-1157ipd3N4, which was able to infect rhesus macaques, closely resembled primary HIV-1 in transmission and pathogenesis, was used to infect several pig-tailed macaques. One of the infected animals subsequently progressed to AIDS, whereas one remained a nonprogressor. The viral envelope evolution in the infected animals during disease progression was analyzed by a bioinformatics approach using ultra-deep …

Jay Reddy Publications

Enteroviruses like coxsackievirus B3 (CVB3) are common suspects in myocarditis/dilated cardiomyopathy patients. Autoimmunity has been proposed as an underlying mechanism, but direct evidence of its role is lacking. To delineate autoimmune response in CVB3 myocarditis, we used IAk dextramers for cardiac myosin heavy chain (Myhc)-α 334–352. We have demonstrated that myocarditis-susceptible A/J mice infected with CVB3 generate Myhc-α-reactive CD4 T cells and such a repertoire was absent in naïve mice as measured by proliferative response to Myhc-α 334–352 and IAk dextramer staining. We also detected Myhc-α 334–352 dextramer+ cells in the hearts of CVB3-infected mice. The autoreactive …

Functional Studies Of Novel Bioactives From Complex Host-Microbiomes As Drug Leads For Cancer, Infectious Disease, Depression And Pain, John Jason Bowling Jan 2012

Functional Studies Of Novel Bioactives From Complex Host-Microbiomes As Drug Leads For Cancer, Infectious Disease, Depression And Pain, John Jason Bowling

Electronic Theses and Dissertations

The drug discovery process has become increasingly complex in comparison to early efforts particularly those involving diseases or conditions that affect large numbers of the human population. It is difficult to point directly to one factor leading evolution of the process but the definition of a mechanism of pharmacological action for a drug lead has noticeably become a higher priority. This evolution has helped diminish the stigma of natural products and compound supply issues. The issue of compound supply has also been addressed to some extent with the emergence of recombinant technologies and new synthetic methodology. With approximately 60 years …

The Role Of The Intestinal Microbiota In Inflammatory Bowel Disease, Seth Bloom Jan 2012

The Role Of The Intestinal Microbiota In Inflammatory Bowel Disease, Seth Bloom

All Theses and Dissertations (ETDs)

Inflammatory bowel disease: IBD) arises from complex interactions of genetic, environmental, and microbial factors. The intestinal microbiota is crucial for IBD induction and complex shifts in microbiota composition occur in IBD, but disease has not been consistently associated with presence or absence of a specific microbe. It is thus controversial whether fulfilling Koch's postulates for individual bacterial species is relevant to IBD and whether disease-associated alterations in microbial colonization are predictive of underlying etiology. Resolving these controversies has been challenging due to paucity of animal models with rapid disease onset, experimental reliance on gnotobiotic animals, and difficulty specifically isolating many …

Pseudomonas Necrotizing Fasciitis In An Otherwise Healthy Infant, Shakeel Ahmed, Syed Rehan Ali, Zahra Aziz Samani Jan 2012

Pseudomonas Necrotizing Fasciitis In An Otherwise Healthy Infant, Shakeel Ahmed, Syed Rehan Ali, Zahra Aziz Samani

Department of Paediatrics and Child Health

Necrotizing fasciitis is an uncommon rapidly progressing infection of soft tissue characterized by a severe inflammation of the fascia and soft tissue. The disease is associated with necrosis and subcutaneous gangrene of the inflamed tissue with systemic toxicity that carries a significant mortality unless timely diagnosed and aggressively treated. Monomicrobial necrotizing fasciitis caused by Pseudomonas aeruginosa is an exceptionally uncommon condition with only few cases reported in the literature so far. We are reporting a six-month-old female infant who was previously healthy and who presented with necrotizing fasciitis and isolates Pseudomonas aeruginosa both from the blood and tissue. The child …

Evading Emergency: Strengthening Emergency Response Through Integrated Pluralistic Governance, Lance A. Gable Jan 2012

Evading Emergency: Strengthening Emergency Response Through Integrated Pluralistic Governance, Lance A. Gable

Law Faculty Research Publications

This Article examines the significant governance challenges that arise during responses to public health emergencies and proposes a new multifaceted strategy-integrated pluralistic governance-to address these challenges. Emergency preparedness is an inherently complex problem that entails the integration ofscientific and medical expertise, good logistical planning, and clear laws and policies. The governance function has particular import for public health emergencies because pandemics, hurricanes, and other disasters can have profoundly divisive social and political consequences. Moreover, recent disasters like Hurricane Katrina and the BP Deepwater Horizon oil spill revealed an emergency preparedness and response infrastructure in the United States that was broken: …

Vip And Host Immunity, Xiaoyu Jiang Jan 2012

Vip And Host Immunity, Xiaoyu Jiang

Wayne State University Dissertations

The purpose of the current dissertation was to examine how VIP regulates host immunity and corneal healing, specifically, its control of growth factors and TLR expression in the P. aeruginosa infected cornea. Firstly, VIP treatment increased growth factor expression (EGF, GHF, FGF and VEGF) in infected cornea. Notably, treatment with a mixture of EGF, FGF and HGF prevented corneal perforation, reduced pro-inflammatory cytokines and bacterial plate count, while increasing anti-inflammatory cytokines and antimicrobials such as murine beta-defensin2 and 3.

We also investigated the expression of TLR-signaling pathways in P. aeruginosa infected corneas with or without VIP treatment. PCR array and …

The Federal Workforce Development Program: An Analysis Of Probationers And Characteristics Associated With And Predictive Of Successful Reentry, Kelley Mcnichols

Electronic Theses and Dissertations

With years of research focusing on soaring incarceration rates, the phenomena of prisoner reentry has been largely overlooked. The majority of incarcerated people will return to the community setting. In fact, millions of recently released offenders are on some form of community based supervision. Today, recidivism is a problem that plagues prisoner reentry. In order to address the challenge of prisoner reentry successfully, reentry initiatives have been established in an attempt to change the way corrections is conceptualized. The purpose of this study was to examine characteristics of probationers that are associated with and predictive of successful reentry. The data …

The Effects Of Total Dissolved Solids On Locomotory Behavior And Body Weight Of Streamside Salamanders, And A Baseline Survey Of Salamander Diversity And Abundance, Meghan Pascuzzi Jan 2012

The Effects Of Total Dissolved Solids On Locomotory Behavior And Body Weight Of Streamside Salamanders, And A Baseline Survey Of Salamander Diversity And Abundance, Meghan Pascuzzi

Electronic Theses and Dissertations

Increased levels of total dissolved solids (TDS) in stream habitats are of concern due to salinity as well as the presence of potentially toxic ions. Natural gas drilling in the Marcellus shale could increase TDS in nearby streams. This thesis investigated the effects of water with elevated TDS on the locomotory activity and body weight of the streamside salamander Desmognathus ochrophaeus. Salamanders were exposed to water collected from streams in southwest Pennsylvania with elevated TDS as well as synthetic ion solutions that mimicked the ionic composition found in streams. Chronic, but not acute exposure to solutions of 1000 ppm TDS …

Perception Profiling & Prolonged Solitary Confinement Viewed Through The Lens Of The Angola 3 Case: When Prison Officials Become Judges, Judges Become Visually Challenged, And Justice Becomes Legally Blind, Angela A. Allen-Bell

UC Law Constitutional Quarterly

Solitary confinement is a tool used by the American prison system without a sufficient check on its application. Current practice provides that an inmate's solitary confinement is reviewed at regular intervals by Executive-branch prison administrators. They have employed it to enforce discipline and order in the prison population, but simultaneously have ridden roughshod over inmates' substantive due process rights by failing to provide a meaningful opportunity for a hearing of the facts and justifications for the continued and prolonged solitary confinement of prison inmates. Many, too many inmates have been kept in solitary confinement for extended periods-some for decades-without any …

Substitute Decision Making About Research: Identifying The Legally Authorized Representative In Four Canadian Provinces, Sheila Wildeman, Gina Bravo, Marie-France Dubois, Carole Cohen, Janice Graham, Karen Painter, Suzanne Bellemare

Articles, Book Chapters, & Popular Press

When an adult is legally incapable of deciding whether to participate in health research, who (if anyone) has the legal authority to make that decision? Furthermore, how well do Canadians with a stake in health research, such as older adults, informal caregivers of older persons with cognitive impairments, researchers in aging, and members of research ethics boards (“REBs”), understand the state of the law on this question? These two interrelated matters are addressed by our study.

We find that the laws of the four provinces we target are frequently unclear as to whether, or in what circumstances, a guardian, proxy …

Efficacy Of Infection Control Interventions In Reducing The Spread Of Multidrug-Resistant Organisms In The Hospital Setting, Joanna R. Wares, Erika M.C. D'Agata, Mary Ann Horn, Shigui Ruan, Glenn F. Webb Jan 2012

Efficacy Of Infection Control Interventions In Reducing The Spread Of Multidrug-Resistant Organisms In The Hospital Setting, Joanna R. Wares, Erika M.C. D'Agata, Mary Ann Horn, Shigui Ruan, Glenn F. Webb

Department of Math & Statistics Faculty Publications

Multidrug-resistant organisms (MDRO) continue to spread in hospitals globally, but the population-level impact of recommended preventive strategies and the relative benefit of individual strategies targeting all MDRO in the hospital setting are unclear. To explore the dynamics of MDRO transmission in the hospital, we develop a model extending data from clinical individual-level studies to quantify the impact of hand hygiene, contact precautions, reducing antimicrobial exposure and screening surveillance cultures in decreasing the prevalence of MDRO colonization and infection. The effect of an ongoing increase in the influx of patients colonized with MDRO into the hospital setting is also quantified. We …

Molecular Insights Into The Uropathogenesis Of Enterococcus Faecalis And Escherichia Coli In The Context Of Urinary Catheterization, Pascale Guiton Jan 2012

Molecular Insights Into The Uropathogenesis Of Enterococcus Faecalis And Escherichia Coli In The Context Of Urinary Catheterization, Pascale Guiton

All Theses and Dissertations (ETDs)

Nosocomial urinary tract infections: UTIs) are a very common result of catheterization with 3 to 7% risk of developing catheter-associated UTIs: CAUTIs) each day catheterized. As multi-drug resistance increases in uropathogens, it is imperative to better understand the effects of catheterization on the urinary tract and pathogenesis. Further, microbial colonization and biofilm production on the surface of urinary catheters are a common component of CAUTIs. Thus, this dissertation focuses on understanding the contribution of known biofilm determinants to urovirulence of Enterococcus faecalis: a common nosocomial uropathogen) and uropathogenic Escherichia coli: UPEC): the most common causative agent of UTI), and the …

Shaken Baby Syndrome, Abusive Head Trauma, And Actual Innocence: Getting It Right, Keith A. Findley, Patrick D. Barnes, David A. Moran, Waney Squier

Articles

In the past decade, the existence of shaken baby syndrome (SBS) has been called into serious question by biomechanical studies, the medical and legal literature, and the media. As a result of these questions, SBS has been renamed abusive head trauma (AHT). This is, however, primarily a terminological shift: like SBS, AHT refers to the two-part hypothesis that one can reliably diagnose shaking or abuse from three internal findings (subdural hemorrhage, retinal hemorrhage, and encephalopathy) and that one can identify the perpetrator based on the onset of symptoms. Over the past decade, we have learned that this hypothesis fits poorly …
