Successful Genotyping Of Microsatellites In The Woolly Mammoth, Yasuko Ishida, Alfred L. Roca, Stephen Fratpietro, Alex D. Greenwood

Genetic analyses using ancient DNA from Pleistocene and early Holocene fossils have largely relied on mitochondrial DNA (mtDNA) sequences. Among woolly mammoths, Mammuthus primigenius, mtDNA analyses have identified 2 distinct clades (I and II) that diverged 1-2 Ma. Here, we establish that microsatellite markers can be effective on Pleistocene samples, successfully genotyping woolly mammoth specimens at 2 loci. Although significant differentiation at the 2 microsatellite loci was not detected between 16 clade I and 4 clade II woolly mammoths, our results demonstrate that the nuclear population structure of Pleistocene species can be examined using fast-evolving nuclear microsatellite markers.

According to the United States Department of Agriculture (USDA), the agricultural health of the United States requires the concurrent feasibility, or coexistence, of organic crops and genetically modified crops. Both types of crops offer separate environmental, economic, and health benefits. Modified crops, or crops infused with beneficial genes to increase yield or decrease the need for chemical applications, are ubiquitous in U.S. farming. Similarly, organic crops, or crops grown without modified genes, are gaining popularity. Unfortunately, the coexistence of organic and modified crops is threatened by the phenomenon of gene flow.

Background: Among vertebrates lens regeneration is most pronounced in newts, which have the ability to regenerate the entire lens throughout their lives. Regeneration occurs from the dorsal iris by transdifferentiation of the pigment epithelial cells. Interestingly, the ventral iris never contributes to regeneration. Frogs have limited lens regeneration capacity elicited from the cornea during pre-metamorphic stages. The axolotl is another salamander which, like the newt, regenerates its limbs or its tail with the spinal cord, but up until now all reports have shown that it does not regenerate the lens.

In this review, we examine examples of conservation of protein structural motifs in unrelated or non-homologous proteins. For this, we have selected three DNA-binding motifs: the histone fold, the helix-turn-helix motif, and the zinc finger, as well as the globin-like fold. We show that indeed similar structures exist in unrelated proteins, strengthening the concept that three-dimensional conservation might be more important than the primary amino acid sequence.

The main objective of the present study is to investigate the cytotoxicity potential and anti-cancer mechanism of 7α-hydroxy-β-sitosterol (CT1), a known stigmastane sterol extracted from bark of Chisocheton tomentosus (Meliaceae). In vitro exposures of this compound was conducted on five cancer cell lines; breast adenocarcinoma cells (MCF-7), hepatocyte liver carcinoma cell (HepG2), oral squamous carcinoma cell (HSC-4) and (HSC-2) and epidermoid cervical carcinoma (Ca Ski) and in comparison with normal human mammary epithelia cell line (HMEC). Pleurotus sajor-caju has become attractive as functional food and a source of physiologically beneficial substances. Pollination-induced senescence in Dendrobium Pompadour flowers was investigated. Pollinated flowers were held in distilled water and in treatment solutions containing either sucrose, glucose, aminooxyacetic acid (AOA) or silver thiosulphate (STS) while unpollinated flowers were held in distilled water. Morphological changes were observed visually while physiological parameters investigated included ethylene production, colour change, petal thickness, fresh weight, dry weight and water uptake. Biochemical changes determined were starch, total sugar and reducing sugar, cell wall hydrolases, total protein, soluble and insoluble protein. The aims of this study are to evaluate the presence of trace metals in aquacultural samples using chemometric methods and to assess the safety of their consumption. In this work, chemometric techniques have been utilized to improve the analysis of metal content in commercially raised tilapia. In the method development phase, multivariate calibration techniques involving principal component regression and partial least squares were employed to establish a simple spectrophotometric method for the simultaneous determination of Cu2+, Ni2+ and Zn2+ in water samples in the presence of 1-(2-thiazolylazo)-2-naphthol as chromogenic reagent. The main aim of this work is to establish a technique to prepare the image of nuclear track detector in the form of thin film, LR 115 Type 2. This research also aims to develop the image processing scripts using SDC Morphology toolbox and Image Processing toolbox in conjunction with MATLAB as a programming platform. Pseudomonas aeruginosa is one of the main causes of healthcare-associated infections among hospitalized patients. Healthcare-associated infections predominantly lead to pneumonia, urinary tract infections, as well as skin and soft-tissue infections. This organism is commonly multiresistant and leads to morbidity and mortality. Persistent systemic inflammatory response syndrome is a serious health condition that may lead to multiple organ dysfunction, organ failure, and ultimately death. It leads to both acute inflammation, caused by either infective (microbes or lipopolysaccharide [LPS]) or non-infective (chemicals) sources, and sepsis, an infection caused by a lethal dose of LPS (endotoxemia). These conditions have similar inflammatory mediators such as cytokines, nitric oxide (NO), high-mobility group box-1 (HMGB1), and heme oxygenase-1 (HO-1), suggesting that they may result from similar pathogenic mechanisms.
