Division of Infectious Diseases and Environmental Medicine Faculty Papers

Botulinum neurotoxin (BoNT) potently inhibits cholinergic signaling at the neuromuscular junction. The ideal countermeasures for BoNT exposure are monoclonal antibodies or BoNT antisera, which form BoNT-containing immune complexes that are rapidly cleared from the general circulation. Clearance of opsonized toxins may involve complement receptor-mediated immunoadherence to red blood cells (RBC) in primates or to platelets in rodents. Methods of enhancing immunoadherence of BoNT-specific antibodies may increase their potency in vivo. We designed a novel fusion protein (FP) to link biotinylated molecules to glycophorin A (GPA) on the RBC surface. The FP consists of an scFv specific for murine GPA fused …

Transformation Of Fusarium Verticillioides With A Polyketide Gene Cluster Isolated From A Fungal Endophyte Activates The Biosynthesis Of Fusaric Acid, Yunxuan Xie, Wei Zhang, Yaoyao Li, Mingzi Wang, Ronald Cerny, Yue Shen, Liangcheng Du

Liangcheng Du Publications

A large number of bioactive natural products have been isolated from plant endophytic fungi. However, molecular mechanisms for the biosynthesis of these metabolites have lagged behind because genetic and biochemical studies are difficult to perform within many of the endophytes. In this work, we describe our attempt to express a putative mycoepoxydiene (MED) biosynthetic gene cluster in Fusarium verticillioides, which has a well-developed genetic system for the study fungal polyketide biosynthesis. MED was isolated from Phomopsis sp. A123, a fungal endophyte of the mangrove plant, Kandelia candel. It has several unusual structural features and interesting biological activities. Integration of …

An Apple A Day Keeps Shareholder Suits At Bay: An Examination Of A Corporate Officer’S Legal Duty To Disclose Health Problems To Shareholders, Alexis Brown Stokes Mar 2011

An Apple A Day Keeps Shareholder Suits At Bay: An Examination Of A Corporate Officer’S Legal Duty To Disclose Health Problems To Shareholders, Alexis Brown Stokes

Texas Wesleyan Law Review

This Article examines both constitutional and practical problems with proposed regulation to require corporate officers to disclose health problems to shareholders. In doing so, the Article addresses the following questions, which are especially timely given the recent controversy surrounding Apple CEO Steve Jobs: To what extent should officers of public companies be required to disclose personal health problems to shareholders? Does a timely failure to disclose such information constitute securities fraud if the stock price is adversely affected once the information becomes public? Can shareholders' rights to accurate information relevant to a company's likely future performance trump an officer's right …

Targeted Critical Thinking: Effective Use Of Critical Thinking Activities, Calvin Wang Mar 2011

Targeted Critical Thinking: Effective Use Of Critical Thinking Activities, Calvin Wang

Library Faculty Scholarship

Library instruction can be primed for failure if participants believe librarians have a canned answer for every research effort. Instruction must take place with an understanding of the unique needs of first-year classes and assignments relative to higher level classes and their assignments. Efforts to incorporate critical thinking exercises in suboptimal research efforts contribute to instruction-fatigue for students and librarians. This workshop will embrace the K-12 strategy of identifying the scope and sequence ideal for teaching library research techniques. It will consider appropriate courses and assignments in which to apply critical thinking activities and present ideas for optimizing assignments. Participants will …

Angiostatin Like Peptides In Milk: Potential Development For Dairy Products Capable Of Cancer Prevention, Erin Stefanutti Mar 2011

Angiostatin Like Peptides In Milk: Potential Development For Dairy Products Capable Of Cancer Prevention, Erin Stefanutti

Master's Theses

For the past 40 years, antiangiogenic approaches have been of major interest in the development of methods to cure and prevent cancer. Angiogenesis, the development of blood vessels from pre-existing vascularization, is essential for cancer growth and spread of metastasis through the delivery of nutrients and oxygen essential to sustain the metabolic activity of these malignant cells. Blocking access to blood will cause cancerous cells to assume a dormant state creating inactive micro-tumors innocuous to the host. Angiostatin, the internal fragment of the fibrinolytic zymogen plasminogen, has shown great potential in reducing cancer size and number of metastatic colonies in …

Beliefs About Racism And Health Among African American Women With Diabetes: A Qualitative Study, Julie A. Wagner, Lisa M. Budris, Sophia Belay, Howard A. Tennen

UCHC Articles - Research

Exposure to racism has been linked to poor health outcomes. Little is known about the impact of racism on diabetes outcomes. This study explored African American (AA) women’s beliefs about how racism interacts with their diabetes self-management and control. Four focus groups were conducted with a convenience sample of 28 adult AA women with type 2 diabetes who were recruited from a larger quantitative study on racism and diabetes. The focus group discussions were transcribed verbatim and analyzed by the authors. Women reported that exposure to racism was a common phenomenon, and their beliefs did in fact link racism to …

Phagosomal Signaling By Borrelia Burgdorferi In Human Monocytes Involves Toll-Like Receptor (Tlr) 2 And Tlr8 Cooperativity And Tlr8-Mediated Induction Of Ifn-Β, Jorge L. Cervantes, Star M. Dunham-Ems, Carson J. La Vake, Mary M. Petzke, Justin D. Radolf, Juan C. Salazar

UCHC Articles - Research

Phagocytosed Borrelia burgdorferi (Bb) induces inflammatory signals that differ both quantitatively and qualitatively from those generated by spirochetal lipoproteins interacting with Toll-like receptor (TLR) 1/2 on the surface of human monocytes. Of particular significance, and in contrast to lipoproteins, internalized spirochetes induce transcription of IFN-β. Using inhibitory immunoregulatory DNA sequences (IRSs) specific to TLR7, TLR8, and TLR9, we show that the TLR8 inhibitor IRS957 significantly diminishes production of TNF-α, IL-6, and IL-10 and completely abrogates transcription of IFN-β in Bb-stimulated monocytes. We demonstrate that live Bb induces transcription of TLR2 and TLR8, whereas IRS957 interferes with their transcriptional regulation. Using …

Isolation By Distance Explains Genetic Structure Of Buggy Creek Virus, A Bird-Associated Arbovirus, Abinash Padhi, Amy T. Moore, Mary Bomberger Brown, Jerome E. Foster, Martin Pfeffer, Charles R. Brown

School of Natural Resources: Faculty Publications

Many of the arthropod-borne viruses (arboviruses) show extensive genetic variability and are widely distributed over large geographic areas. Understanding how virus genetic structure varies in space may yield insight into how these pathogens are adapted to and dispersed by different hosts or vectors, the relative importance of mutation, drift, or selection in generating genetic variability, and where and when epidemics or epizootics are most likely to occur. However, because most arboviruses tend to be sampled opportunistically and often cannot be isolated in large numbers at a given locale, surprisingly little is known about their spatial genetic structure on the local …

Unionid Mussels From Nearshore Zones Of Lake Erie, Todd D. Crail, Robert A. Krebs, David T. Zanatta

Biological, Geological, and Environmental Faculty Publications

Concern exists that the introduction of dreissenid mussels following long-term effects of pollution may have completely eliminated native mussel species from Lake Erie. Natural seiche events were used to facilitate surveys for live unionids on five occasions in the western basin of Lake Erie and Sandusky Bay between 2007 and 2009, and beach and estuary surveys were conducted at numerous additional sites between 2004 and 2009. Sixteen unionid species were found living in or near Lake Erie, including six sites in the nearshore zone of the lake. Each community consisted of live individuals from two to eight species, and evidence …
