Journal Articles: Biochemistry & Molecular Biology

We describe a case of progressive multifocal leukoencephalopathy (PML) caused by infection with the human polyomavirus JC virus in a patient with B-cell small lymphocytic leukemia who was treated with rituximab. The first symptoms of PML appeared immediately following the last of five cycles of rituximab, cyclophosphamide and pentostatin. Magnetic resonance imaging revealed changes consistent with PML, although JC virus DNA was not detected by polymerase chain reaction assay of the cerebrospinal fluid. A stereotactic biopsy of the brain showed histological changes consistent with PML, while electron microscopy revealed JC virus particles attached to the nuclei of astrocytes. The patient …

Publications and Research

We characterized the differentiation of rat bone marrow-derived mesenchymal stem cells (BM-MSCs) into tenocyte-like cells in response to bone morphogenetic protein-12 (BMP-12). BM-MSCs were prepared from Sprague-Dawley rats and cultured as monolayers. Recombinant BMP-12 treatment (10 ng/ml) of BM-MSCs for 12 hours in vitro markedly increased expression of the tenocyte lineage markers scleraxis (Scx) and tenomodulin (Tnmd) over 14 days. Treatment with BMP-12 for a further 12-hour period had no additional effect. Colony formation assays revealed that ∼80% of treated cells and their progeny were Scx- and Tnmd-positive. BM-MSCs seeded in collagen scaffolds and similarly treated with a single dose …

Super Collision Energy Transfer Studies In Single Collisions Between Vibrationally Hot Benzene Like Molecules And Ground State Bath Molecules: The Effect Of Physical Properties Of Donor And Bath Molecules On Super Collision Energy Transfer, Kilyoung Kim

Theses and Dissertations

This research is focused on single-collision energy transfer events between highly vibrationally excited benzene-like donor molecules and small bath molecules, CO2 and N2O in the vibrational ground level. Measuring how much energy is transferred from donors to bath molecules was accomplished by probing bath molecules scattered into specific-rotational states using a tunable Δv=0.0003 cm-1 solid state diode laser. The normalized energy transfer probability distribution function, P(E,E'), determined from energy gain information, is very useful in comparing collisional energy transfer efficiency between various collision systems. P(E,E') is also used to investigate the effects of donor and bath physical properties on collisional …

Expression And Localization Of Clc Chloride Transport Proteins In The Avian Retina, Emily Mcmains, Vijai Krishnan, Sujitha Prasad, Evanna Gleason Mar 2011

Expression And Localization Of Clc Chloride Transport Proteins In The Avian Retina, Emily Mcmains, Vijai Krishnan, Sujitha Prasad, Evanna Gleason

Faculty Publications

Members of the ubiquitously expressed CLC protein family of chloride channels and transporters play important roles in regulating cellular chloride and pH. The CLCs that function as Cl-/H+ antiporters, ClCs 3-7, are essential in particular for the acidification of endosomal compartments and protein degradation. These proteins are broadly expressed in the nervous system, and mutations that disrupt their expression are responsible for several human genetic diseases. Furthermore, knock-out of ClC3 and ClC7 in the mouse result in the degeneration of the hippocampus and the retina. Despite this evidence of their importance in retinal function, the expression patterns of different CLC …

Leaf Number Indicates Salt Tolerance Of Young Seedling Families Of European Aspen (Populus Tremula L.) Growing In Different Soils, Lu-Min Vaario, Kim Yrjälä, Matti Rousi, Timo Sipila, Pertti Pulkkinen

Aspen Bibliography

Soil salinity limits plant productivity and quality. We evaluated the response of 12 aspen (Populus tremula) families to salt stress in two different soils irrigated for 4-weeks with 0, 80 or 160 mM saline solution. Easily measurable characteristics such as shoot height, leaf number, dry mass as well as the distribution of sodium (Na+) ions were measured in 5-month-old aspen seedlings raised in controlled greenhouse conditions on two different soils. Growth among families varied significantly, and the interaction between family and soil type was significant. From 2–5 months, leaf number correlated with that of the first month and salin-ity tolerance. …

Fungal Recognition Enhances Mannose Receptor Shedding Through Dectin-1 Engagement, Umut Gazi, Marcela Rosas, Sonali Singh, Sigrid Heinsbroek, Imran Haq, Simon Johnson, Gordon D. Brown, David L. Williams, Philip R. Taylor, Luisa Martinez-Pomares

ETSU Faculty Works

The mannose receptor (MR) is an endocytic type I membrane molecule with a broad ligand specificity that is involved in both hemostasis and pathogen recognition. Membrane-anchored MRis cleaved by a metalloproteinase into functional soluble MR (sMR) composed of the extracellular domains of intact MR. Although sMR production was initially considered a constitutive process, enhanced MR shedding has been observed in response to the fungal pathogen Pneumocystis carinii. In this work, we have investigated the mechanism mediating enhanced MR shedding in response to fungi. We show that other fungal species, including Candida albicans and Aspergillus fumigatus, together with zymosan, a preparation …

Sawdust Pyrolysis And Petroleum Coke Co2 Gasification At High Heating Rates, Aaron D. Lewis

Theses and Dissertations

Clean and efficient electricity can be generated using an Integrated Gasification Combined Cycle (IGCC). Although IGCC is typically used with coal, it can also be used to gasify other carbonaceous species like biomass and petroleum coke. It is important to understand the pyrolysis and gasification of these species in order to design commercial gasifiers and also to determine optimal conditions for operation. High heating-rate (100,000 K/s) pyrolysis experiments were performed with biomass (sawdust) in BYU's atmospheric flat-flame burner reactor at conditions ranging from 1163 to 1433 K with particle residence times ranging from 23 to 102 ms. Volatile yields and …

Evaluation Of Xmx/2l-Mil Virtual Impactor Performance And Capture And Retention Of Aerosol Particles In Two Different Collection Media, Jon E. Black

Theses and Dissertations

The United States Air Force uses the XMX/2L-MIL (XMX) high volume air sampler to collect samples for biological analysis. The XMX uses a virtual impactor to concentrate particles 1.0 to 10 μm in size into a secondary flow prior to sample collection using a liquid impinger in a collection tube. There are no known published studies regarding virtual impactor inter-instrument variability, effect of reducing the secondary flow on particle concentration, or capture and retention efficiency (CRE) of particles in the collection media performance characteristics when using the XMX. These performance characteristics were evaluated by lofting test aerosols of Arizona Road …

A Study Of Corporate Entrepreneurship In A Department Of Defense Organization, Wade W. Brower Mar 2011

A Study Of Corporate Entrepreneurship In A Department Of Defense Organization, Wade W. Brower

Theses and Dissertations

What determines if an organization has a corporate entrepreneurship (CE) environment? While being entrepreneurial in a private sector job usually means a greater market share or a higher profit margin; in a public research organization, CE must be measured in other ways. This thesis evaluates a Department of Defense (DoD) organization that performs basic research. Any organization that does government or nonprofit work must continually find new ways, methods, processes, or ideas to complete the mission. In this particular study, the mission is to continue to support the warfighter with limited and changing resources—to ensure the military can maintain dominance …

The threat of these chemicals use against the military and civilians continues today, and the suggested treatment guidelines available may be ineffective or possibly cause harm. The guidelines investigated during the research presented here all included the use of two antidotes, atropine and oxime. The efficacy of oximes has been questioned and it has been suggested that they may cause harm to the patient. Both atropine and oxime are issued to military …

Modeling Vertical Flow Treatment Wetland Hydraulics To Optimize Treatment Efficiency, David T. Roen Ii Mar 2011

Modeling Vertical Flow Treatment Wetland Hydraulics To Optimize Treatment Efficiency, David T. Roen Ii

Theses and Dissertations

An upward Vertical Flow Treatment Wetland (uVFTW) at Wright Patterson AFB designed to bioremediate contaminated groundwater exhibits hydraulic short-circuiting. Prior studies estimated that groundwater flowed through less than 50% of the wetland’s volume, and that the mean residence time was significantly less than the nominal residence time, which was calculated assuming flow through the entire wetland volume. The objective of this research was to investigate how uVFTW hydraulics affects treatment efficiency, and to propose design strategies to maximize treatment efficiency. A groundwater flow and contaminant transport model of a uVFTW that couples hydraulics and degradation kinetics was built and applied …

Characterization Of Graphite Composite Material Particulates From Usaf Aircraft Maintenance Operations, Richard E. Yon Mar 2011

Characterization Of Graphite Composite Material Particulates From Usaf Aircraft Maintenance Operations, Richard E. Yon

Theses and Dissertations

Due to the benefits of their material properties, advanced composite materials (ACM) are increasingly being used as structural components on aircraft, especially within the United States Air Force. As a result, the potential exists for occupational exposures to structural maintenance employees while repairing and fabricating aircraft components. Two field studies were conducted for this thesis in order to characterize ACM aerosol size distribution, determine the feasibility of utilizing direct reading instruments (DRIs) in the field, and ensure workers are protected with adequate controls. In order to characterize exposure, traditional integrated air sampling and DRIs were positioned together near an ACM …

It is herein proposed that Microelectromechanical System fabrication methods will provide a fast, cheap, and highly repeatable manufacturing method for the FWMAV wings. The wings manufactured to demonstrate this process, bio-inspired by the Manduca Sexta, …

Small Internal Combustion Engine Testing For A Hybrid-Electric Remotely-Piloted Aircraft, Isseyas H. Mengistu Mar 2011

Small Internal Combustion Engine Testing For A Hybrid-Electric Remotely-Piloted Aircraft, Isseyas H. Mengistu

Theses and Dissertations

Efficient operation of a hybrid-electric propulsion system (HEPS) powering a small remotely-piloted aircraft (RPA) requires that a controller have accurate and detailed engine and electric motor performance data. Many small internal combustion engines (ICEs) currently used on various small RPA were designed for use by the recreational hobbyist radio-control (R/C) aircraft market. Often, the manufacturers of these engines do not make accurate and reliable detailed engine performance data available for their engines. A dynamometer testing stand was assembled to test various small ICEs. These engines were tested with automotive unleaded gasoline (the manufacturer's recommended fuel) using the dynamometer setup. Torque, …

Theses and Dissertations

This thesis provides a novel visualization method to analyze the impact that articulations in dismount pose and camera aspect angle have on histograms of oriented gradients (HOG) features and eventual detections. Insights from these relationships are used to identify limitations in a state of the art skin cued HOG dismount detector's ability to detect poses not in a standard upright stances. Improvements to detector performance are made by further leveraging available skin information, reducing false detections by an additional order of magnitude. In addition, a method is outlined for training supplemental support vector machines (SVMs) from computer generated data, for …
