Cells use redox signaling to adapt to oxidative stress. For instance, certain transcription factors exist in a latent state that may be disrupted by oxidative modifications that activate their transcription potential. We hypothesized that DNA-binding sites (response elements) for redox-sensitive transcription factors may also exist in a latent state, maintained by co-repressor complexes containing class I histone deacetylase (HDAC) enzymes, and that HDAC inactivation by oxidative stress may antagonize deacetylase activity and unmask electrophile-response elements, thus activating transcription. Electrophiles suitable to test this hypothesis include reactive carbonyl species, often derived from peroxidation of arachidonic acid. We report that alpha,beta-unsaturated carbonyl …

Shewanellae are gram-negative facultatively anaerobic metal-reducing bacteria commonly found in chemically (i.e., redox) stratified environments. Occupying such niches requires the ability to rapidly acclimate to changes in electron donor/acceptor type and availability; hence, the ability to compete and thrive in such environments must ultimately be reflected in the organization and utilization of electron transfer networks, as well as central and peripheral carbon metabolism. To understand how Shewanella oneidensis MR-1 utilizes its resources, the metabolic network was reconstructed. The resulting network consists of 774 reactions, 783 genes, and 634 unique metabolites and contains biosynthesis pathways for all cell constituents. Using constraint-based …

Metal-Organic Materials (MOMs) represent an important class of solid-state crystalline materials. Their countless attractive attributes make them uniquely suited to potentially resolve many present and future utilitarian societal challenges ranging from energy and the environment, all the way to include biology and medicine. Since the birth of coordination chemistry, the self-assembly of organic molecules with metal ions has produced a plethora of simple and complex architectures, many of which possess diverse pore and channel systems in a periodic array. Activation of immunity to self-antigens is the goal in cancer immunotherapy, whereas blocking such responses is the goal in autoimmune disease. Thus, it is not surprising that investigation into cancer immunotherapy might also produce insights for the treatment of autoimmune disease. Heat shock protein, gp96, based therapies lead to the robust activation of CD8+ cytotoxic T cells that can slow tumor growth in 60-70% of mice, but only lead to the elimination of tumors in 30-40% of animals. Background
Chromosomal segmental copy number variation (CNV) has been recently recognized as a very important source of genetic variability. Some CNV loci involve genes or conserved regulatory elements. Compelling evidence indicates that CNVs impact genome functions. The chicken is a very important farm animal species which has also served as a model for biological and biomedical research for hundreds of years. A map of CNVs in chickens could facilitate the identification of chromosomal regions that segregate for important agricultural and disease phenotypes.

Disease Outbreaks Associated With Recent Hurricanes Cause Mass Mortality Of Sea Urchins In Nova Scotia, R. E. Scheibling, Colette Feehan, J. S. Lauzon-Guay

Field observations and laboratory experiments support the hypothesis that diseaseinduced mass mortality of the sea urchin Strongylocentrotus droebachiensis can be associated with hurricane events that introduce a pathogenic amoeba Paramoeba invadens to coastal waters. The temporal pattern of morbidity and mortality of sea urchins observed in a large embayment on the Atlantic coast of Nova Scotia following Hurricane Juan in September 2003 and Hurricane Bill in August 2009, and in laboratory infection experiments during the 2009 event, closely conformed to that expected based on the known temperature-dependent dynamics of this amoebic disease.

The broad-spectrum apoptosis (caspase) inhibitor, Q-VD-OPh, has been shown to have no side effects and is effective at a much lower concentration than other FMK-type caspase inhibitors. However, an appropriate negative control to use with this inhibi- tor has not been available. In this study, we developed and analyzed a new compound, based on the Q-VD-OPh backbone, which acts as a cognate negative control. To create the negative control, we substituted a glutamate residue for the aspartate residue to create Q-VE-OPh, thereby retaining the identical charge and molecular properties with only the addition of an extra –CH2 group. Mammalian tooth enamel is often chipped, providing clear evidence for localized contacts with large hard food objects. Here, we apply a simple fracture equation to estimate peak bite forces directly from chip size. Many fossil hominins exhibit antemortem chips on their posterior teeth, indicating their use of high bite forces. The inference that these species must have consumed large hard foods such as seeds is supported by the occurrence of similar chips among known modern-day seed predators such as orangutans and peccaries. The existence of tooth chip signatures also provides a way of identifying the consumption of rarely eaten foods …

In the fall of 2009, Portland State University President Wim Wiewel commissioned a Task Force, and an associated Advisory Board composed of community stakeholders, to make strategic recommendations concerning how Portland State can best serve the higher education needs and aspirations of Oregon’s Latino community. President Wiewel further charged the Task Force to base its recommendations on the best research and evidence available and to consider the most effective investment of PSU resources.
The Task Force met monthly from September 2009 to May 2010. It reviewed the literature, studied relevant data, learned from the success of comparable institutions, and performed …

Venous thromboembolism (VTE) is the leading cause of death from cardiovascular disease. Despite the importance of the glycoprotein (GP) Ib-IX/von Willebrand factor (VWF) axis in arterial thrombosis, its requirement in venous, not venule thrombosis has not been tested. We evaluated FeCl3-induced thrombus formation in inferior vena cava (IVC) of mice. We first established FeCl3 injury conditions (20% FeCl3, 10 minutes) requiring platelets for IVC occlusion as confirmed by a lack of occlusion in severely thrombocytopenic ( less than 23% circulating platelet count) mice (n=7) and stable occlusion in control animals. The Relationship Between Childhood Adversity And Depression: The Influence Of Schemas And Coping Style On Depression Course, Jessica Childress

This thesis explored the relationship between childhood adversity and depression course, and the influence of Early Maladaptive Schemas (EMS) and coping on this relationship. A sample of participants in a small-scale university study (n=100) provided information through retrospective assessment of adverse childhood experiences. Information through self-report assessment of coping style, Early Maladaptive Schema (EMS), and depressive symptoms was also obtained. Seventy-three percent of the sample reported experiencing at least one of the adversities assessed. Childhood adversity was a predictor of acute symptoms of depression, and general negative affect, indicating that experiencing childhood adversity leads to acute and chronic symptoms of …
