PURPOSE OF REVIEW: To provide clinical investigators with an understanding of factors to consider when wishing to add cytokine and inflammatory marker measurements to their studies. RECENT FINDINGS: Inflammation involves complex and coordinated responses of the immune system to tissue damage. In the absence of tools to routinely assess inflammation within living tissues, measurements of humoral factors such as cytokines and other inflammatory mediators or markers can provide predictive clinical information and insights into disease mechanisms. Historically, enzyme-linked immunosorbent assays (ELISAs) became the gold standard, yet this approach of measuring a single protein in each sample limits the amount of …

A Detailed Mathematical Model Predicts Serial Engagement Of Ige-Fcεri Complexes Can Enhance Syk Activation In Mast Cells, Michael L. Blinov

The term serial engagement was introduced to describe the ability of a single peptide, bound to a MHC molecule, to sequentially interact with TCRs within the contact region between a T cell and an APC. In addition to ligands on surfaces, soluble multivalent ligands can serially engage cell surface receptors with sites on the ligand, binding and dissociating from receptors many times before all ligand sites become free and the ligand leaves the surface. Crystal Structure Of The Gerbc Component Of A Bacillus Subtilis Spore Germinant Receptor, Yunfeng Li, Barbara Setlow, Peter Setlow, Bing Hao

The nutrient germinant receptors (nGRs) of spores of Bacillus species are clusters of three proteins that play a critical role in triggering the germination of dormant spores in response to specific nutrient molecules. Here we report the crystal structure of the C protein of the GerB germinant receptor, so called GerBC, of Bacillus subtilis spores at 2.3 Å resolution. The GerBC protein adopts a previously uncharacterized type of protein fold consisting of three distinct domains, each of which is centered by a β sheet surrounded by multiple α helices. Prescription drug warning labels are regulated by both state products liability law and federal regulations imposed by the Food, Drug, and Cosmetic Act. Often, federal law acts as a minimum standard to which states may add stricter labeling requirements. Occasionally, however, state and federal labeling requirements place a prescription drug manufacturer in a position where compliance with state products liability law causes it to violate federal regulations and vice versa. When this occurs, a decision must be made as to whether state or federal drug labeling requirements prevail. Integrating Evolutionary And Functional Approaches To Infer Adaptation At Specific Loci, Jay F. Storz, Christopher W. Wheat

Inferences about adaptation at specific loci are often exclusively based on the static analysis of DNA sequence variation. Ideally, population-genetic evidence for positive selection serves as a stepping-off point for experimental studies to elucidate the functional significance of the putatively adaptive variation. We argue that inferences about adaptation at specific loci are best achieved by integrating the indirect, retrospective insights provided by population-genetic analyses with the more direct, mechanistic insights provided by functional experiments. Integrative studies of adaptive genetic variation may sometimes be motivated by experimental insights into molecular function, which then provide the impetus to perform population genetic tests …

Feed cost has been estimated to account for approxi¬mately 70% of the expenses associated with a beef cattle operation. The consequences of under- or overfeeding beef cows can be substantial. Underfeeding cows can result in thin body condition, poor reproductive performance, and poor milk production. On the other hand, overfeeding can be unnecessarily expensive, increase calving difficulty, reduce milk production, and potentially have a negative impact on the environment. To effectively balance diets, it is essential to know the nutrient requirements of the animal. Demand for nutrients can vary by numerous factors. Determination Of Cyanide Exposure By Gas Chromatography–Mass Spectrometry Analysis Of Cyanide-Exposed Plasma Proteins, Stephanie L. Youso, Gary A. Rockwood, John A. Lee, Brian A. Logue

Exposure to cyanide can occur in a variety of ways, including exposure to smoke from cigarettes or fires, accidental exposure during industrial processes, and exposure from the use of cyanide as a poison or chemical warfare agent. Confirmation of cyanide exposure is difficult because, in vivo, cyanide quickly breaks down by a number of pathways, including the formation of both free and protein-bound thiocyanate. A simple method was developed to confirm cyanide exposure by extraction of protein-bound thiocyanate moieties from cyanide-exposed plasma proteins. Thiocyanate was successfully extracted and subsequently derivatized with pentafluorobenzyl bromide for GC–MS analysis. Thiocyanate levels as low …

Alcohol and drug problems affect not only those using these substances but also family members of the individual substance user. There has been a historic neglect within the addiction sphere of both research and practice pertaining to the effect of drug use on the family. Each family member is uniquely affected with negative outcomes ranging from economic hardship to violence being perpetrated against them (Csiernik, 2002). Thus, treating only the individual with the substance problem is limiting and serves an overly narrow orientation for the enhancement of both family and community health.
