Integrating Genomics And Phylogenetics In Understanding The History Of Trichinella Species, Dante Zarlenga, B. Rosenthal, E. Hoberg, M. Mitreva

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

In 2004, funding was received by Washington University’s Genome Sequencing Center through NHGRI, to completely sequence several nematode genomes as part of a holistic effort to advance our understanding of the human genome and evolution within the Metazoa. Trichinella spiralis was among this group of worms because of its strategic location at the base of the phylum Nematoda, and the belief that extant species represented an ancient divergent event that occurred as early as the Paleozoic. At the same time, a concerted effort was put forth to solidify the phylogeny of extant species of Trichinella based upon molecular analyses of …

Reproductive And Sphingolipid Metabolic Effects Of Fumonisin B1 And Its Alkaline Hydrolysis Product In Lm/Bc Mice: Hydrolyzed Fumonisin B1 Did Not Cause Neural Tube Defects, Kenneth A. Voss, Ronald T. Riley, Maurice E. Snook, Janee Gelineau-Van Waes

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Fumonisins are mycotoxins produced by Fusarium verticillioides. They are toxic to animals and exert their effects through mechanisms involving disruption of sphingolipid metabolism. Fumonisins are converted to their hydrolyzed analogs by alkaline cooking (nixtamalization). Both fumonisins and hydrolyzed fumonisins are found in nixtamalized foods such as tortillas, and consumption of tortillas has been implicated as a risk factor for neural tube defects (NTD). Fumonisin B1 (FB1) induced NTD when given (ip) to pregnant LM/Bc mice; however, neither the NTD induction potential of hydrolyzed fumonisin B1 (HFB1) nor its affect on sphingolipid metabolism in …

Expression And Functional Characterization Of The Plant Antimicrobial Snakin-1 And Defensin Recombinant Proteins, Natalia Kovalskaya, Rosemarie W. Hammond

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

In this study, for the first time, functionally active, recombinant, cysteine-rich plant proteins snakin-1 (SN1) and defensin (PTH1) were expressed and purified using a prokaryotic expression system. The overall level of antimicrobial activities of SN1 and PTH1 produced in Escherichia coli was commensurate with that of the same proteins previously obtained from plant tissues. Both proteins exhibited strong antibacterial activity against the phytopathogenic bacterium Clavibacter michiganensis subsp. sepedonicus (50% inhibitory concentration (IC50) 1.5–8 lM) and antifungal activity against the phytopathogenic fungi Colletotrichum coccoides and Botrytis cinerea (IC50 5–14 lM). Significantly weaker activity was observed against Pseudomonas syringae …

Efficacy And Safety Of Catnip (Nepeta Cataria) As A Novel Filth Fly Repellent, J. J. Zhu, X.-P. Zeng, D. Berkebile, H.-J. Du, Y. Tong, K. Qian

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Catnip (Nepeta cataria) is known for its pseudo-narcotic effects on cats. Recently, it has been reported as an effective mosquito repellent against several Aedes and Culex species, both topically and spatially. Our laboratory bioassays showed that catnip essential oil (at a dosage of 20 mg) resulted in average repellency rates of 96% against stable flies, Stomoxys calcitrans (L.) and 79% against houseflies, Musca domestica (L.), respectively. This finding suggested that the application of repellent could be used as part of filth fly management. Further evaluations of catnip oil toxicity were conducted to provide a broad-spectrum safety profile of …

Vegetation Of Utah, R. Douglas Ramsey, Neil E. West Jan 2009

Vegetation Of Utah, R. Douglas Ramsey, Neil E. West

Wildland Resources Faculty Publications

In order to organize and facilitate management of Utah’s ecosystems, generalization of the ecological variation found across the state is necessary. Ecosystems involve complex interactions between environment and biota, and there have been many efforts to generalize and categorize these interactions in order to gain a better understanding of their structure and organization. The most common method of categorizing ecological variations across large landscapes today incorporates the ecoregion concept. Ecoregions are geographic delineations of landscapes containing ecosystems linked by similar climatic, geologic, soil, and landform characteristics. The primary characteristics used to delineate ecoregions vary depending on the overall goal of …

Blood Chimerism Confounds Genetic Relative Susceptibility Testing For Classical Scrapie In Sheep, David A. Schneider, Ahmed Tibary, Terje Taudsepp, Pranab J. Das, Katherine I. O'Rourke Jan 2009

Blood Chimerism Confounds Genetic Relative Susceptibility Testing For Classical Scrapie In Sheep, David A. Schneider, Ahmed Tibary, Terje Taudsepp, Pranab J. Das, Katherine I. O'Rourke

Zoonotics and Wildlife Disease: Publications

Classical scrapie disease is a transmissible spongiform encephalopathy of sheep that is enzootic in the United States. Susceptibility of sheep to classical scrapie is linked to single nucleotide polymorphisms in the prion protein gene (PRNP), forming the basis for genetic testing strategies used by national efforts to eradicate scrapie. Such efforts are occasionally hampered by inconclusive results stemming from the detection of ‘‘complex’’ genotypes. Naturally occurring cases of ovine chimerism are thought to account for some of these instances. In the current report, 4 naturally occurring ovine chimeras are documented through cytogenetic and molecular analyses. All 4 of these sheep …

Nor98 Scrapie Identified In The United States, Christie M. Loiacono, Bruce V. Thomsen, S. Mark Hall, Matti Kiupel, Diane Sutton, Katherine I. O'Rourke, Bradd Barr, Lucy Anthenill, Delwyn Keane

Zoonotics and Wildlife Disease: Publications

A distinct strain of scrapie identified in sheep of Norway in 1998 has since been identified in numerous countries throughout Europe. The disease is known as Nor98 or Nor98-like scrapie, among other names. Distinctions between classic scrapie and Nor98 scrapie are made based on histopathology and immunodiagnostic results. There are also differences in the epidemiology, typical signalment, and likelihood of clinical signs being observed. In addition, sheep that have genotypes associated with resistance to classic scrapie are not spared from Nor98 disease. The various differences between classic and Nor98 scrapie have been consistently reported in the vast majority of cases …

Dna Delivery From Matrix Metalloproteinase Degradable Poly (Ethylene Glycol) Hydrogels To Mouse Cloned Mesenchymal Stem Cells, Yuguo Lei, Tatiana Segura

Department of Chemical and Biomolecular Engineering: Faculty Publications

The ability to genetically modify mesenchymal stem cells (MSCs) seeded inside synthetic hydrogel scaffolds would offer an alternative approach to guide MSC differentiation and to study molecular pathways in three dimensions than protein delivery. In this report, we explored gene transfer to infiltrating MSCs into matrix metalloproteinase (MMP) degradable hydrogels that were loaded with DNA/poly(ethylene imine) (PEI) polyplexes. DNA/PEI polyplexes were encapsulated inside poly (ethylene glycol) (PEG) hydrogels crosslinked with MMP degradable peptides via Michael Addition chemistry. A large fraction of encapsulated polyplexes remained active after encapsulation (65%) and the mechanical properties of the hydrogels were unchanged by the encapsulation …

The Relationship Of Religious Self-Identification To Cultural Adaptation Among Iranian Immigrants And First Generation Iranians, Nazanin Saghafi Jan 2009

The Relationship Of Religious Self-Identification To Cultural Adaptation Among Iranian Immigrants And First Generation Iranians, Nazanin Saghafi

Theses and Dissertations

This causal-comparative study examined if Iranian or Iranian Americans of either Islamic or Jewish religious self-identifications significantly differ in their reported level of cultural adaptation as evidenced by level of acculturation and the degree of acculturative stress, after controlling for the influence of years of residence in the United States. To conduct this investigation, 107 participants were administered the Cultural Lifestyle Inventory (Mendoza, 1989), which was adapted for use with members of the Iranian culture (Ghaffarian, 1998), and the Kerendi-Kadkhoda Acculturative Stress Scale (Kerendi, 1998). The results of the MANCOVA indicate that religious identification does significantly influence cultural adaptation. Participants …
