Wittgenstein And Worship: Investigations Of Liturgical Language-Games And Their Formative Role In Christian Identity, Michelle Kathleen Weber Jan 2008

Wittgenstein And Worship: Investigations Of Liturgical Language-Games And Their Formative Role In Christian Identity, Michelle Kathleen Weber

School of Theology and Seminary Graduate Papers/Theses

I argue that the Christian community’s religious language games, particularly liturgical action (theologia prima) and theological reflection (theologia secunda), mutually shape and correct one another. I demonstrate this using Wittgenstein’s philosophical method, adapting his approach to philosophy for theological use. The first part of this essay explains and demonstrates Wittgenstein’s philosophical method. The second part is an application of his method (with my own adaptation of this method for theology) with Christian liturgy as its subject. The third part of this essay discusses ways in which Wittgenstein’s approach might be adapted further in a fruitful way …

Distribution, Reproduction, And Transport Of Zooplankton In The Western Arctic, Leopoldo Llinas Jan 2008

Distribution, Reproduction, And Transport Of Zooplankton In The Western Arctic, Leopoldo Llinas

Open Access Dissertations

This dissertation focuses on the distribution, reproduction, and transport of zooplankton in the Chukchi and Beaufort seas and adjacent Canada Basin. Specifically, it analyzes 1) the species-specific distribution of copepod nauplii as it is forced by the surface layer mesoscale circulation and physical properties, 2) the reproduction of the dominant copepod Calanus glacialis in the western Arctic, and 3) the effects of eddy transport on the zooplankton community in the Chukchi and Beaufort seas and Canada Basin. To achieve this I adapted a molecular identification method to work with small crustaceans. The method was successfully used to generate a sequence …

The Outer Limits: Telomere Maintenance By Trf2 And G-Quadruplex Dna Structures, Ilene M. Pedroso Jan 2008

The Outer Limits: Telomere Maintenance By Trf2 And G-Quadruplex Dna Structures, Ilene M. Pedroso

Open Access Dissertations

Human telomeric DNA consists of tandem repeats of the sequence 5'-d(TTAGGG)-3' assembled into a nucleoprotein complex that functions to protect the ends of chromosomes. Such guanine-rich DNA is capable of forming a variety of G-quadruplexes, which in turn, can have varying functional consequences on telomere maintenance. G-quadruplex stabilizing ligands have been shown to induce chromosome end-to-end fusions, senescence and apoptosis, effects similar to the expression of a dominant-negative TTAGGG Repeat Factor 2 (TRF2). With this in mind, we analyzed the effect of sequence and length of human telomeric DNA, as well as cation conditions on G-quadruplex formation by native polyacrylamide …

Nitrogen Cycling On Coral Reefs: A Stable Isotopic Investigation Of Nutrient Dynamics Within The Florida Keys Coral Reef Tract, Kathryn Amanda Lamb-Wozniak

Open Access Dissertations

This dissertation serves as a comprehensive, natural-abundance analysis of the present-day spatial and temporal dynamics and trophic linkages of nitrogen from within the Florida Keys National Marine Sanctuary (FKNMS). This work was accomplished by documenting the delta 15 N of particulate organic matter (POM), several genera of aquatic vegetation and herbivorous fish, as well as dissolved water column nitrogen. Seasonal and spatial variations in delta 15 N were assessed, trophic level variations among reef constituents were quantified, and relative contributions of both natural and anthropogenic nitrogen into the reef tract were determined. The measured mean delta 15 N of POM …

Repression Of Hla By Rot Is Dependent On Sae In Staphylococcus Aureus, Dongmei Li, Ambrose Cheung Jan 2008

Repression Of Hla By Rot Is Dependent On Sae In Staphylococcus Aureus, Dongmei Li, Ambrose Cheung

Dartmouth Scholarship

The regulatory locus sae is a two-component system in Staphylococcus aureus that regulates many important virulence factors, including alpha-toxin (encoded by hla) at the transcriptional level. The SarA homologs Rot and SarT were previously shown to be repressors of hla in selected S. aureus backgrounds. To delineate the interaction of rot and sae and the contribution of sarT to hla expression, an assortment of rot and sae isogenic single mutants, a rot sae double mutant, and a rot sae sarT markerless triple mutant were constructed from wild-type strain COL. Using Northern blot analysis and transcriptional reporter gene green fluorescent protein, …

The Osu1/Qua2/Tsd2-Encoded Putative Methyltransferase Is A Critical Modulator Of Carbon And Nitrogen Nutrient Balance Response In Arabidopsis, Peng Gao, Zeyu Xin, Zhi-Liang Zheng Jan 2008

The Osu1/Qua2/Tsd2-Encoded Putative Methyltransferase Is A Critical Modulator Of Carbon And Nitrogen Nutrient Balance Response In Arabidopsis, Peng Gao, Zeyu Xin, Zhi-Liang Zheng

Publications and Research

The balance between carbon (C) and nitrogen (N) nutrients must be tightly coordinated so that cells can optimize their opportunity for metabolism, growth and development. However, the C and N nutrient balance perception and signaling mechanism remains poorly understood. Here, we report the isolation and characterization of two allelic oversensitive to sugar1 mutants (osu1-1, osu1-2) in Arabidopsis thaliana. Using the cotyledon anthocyanin accumulation and root growth inhibition assays, we show that the osu1 mutants are more sensitive than wild-type to both of the imbalanced C/N conditions, high C/low N and low C/high N. However, under the balanced C/N conditions (low …

Wildland Resources Faculty Publications

Many land‐management agencies are caught between decreased budgets and increasing public interest. Furthermore, semi‐arid landscapes are sensitive to management prescriptions and use, and require a significant amount of monitoring in order to assess vegetation productivity and health. The purpose of this study was to evaluate the use of Landsat Enhanced Thematic Mapper (ETM) Imagery to monitor seasonal vegetation cover in a shrub‐steppe ecosystem. The study area, managed by The Utah School and Institutional Trust Lands Administration, consists of a shrub‐steppe environment in south‐central Utah. Biotic (tree, shrub, grass, and forbs) and abotic (slope, aspect, elevation, landform type, and slope shape) …

Genome-Wide Analysis Of The Interaction Between The Endosymbiotic Bacterium Wolbachia And Its Drosophila Host, Zhiyong Xi, Laurent Gavotte, Yan Xie, Stephen L. Dobson Jan 2008

Genome-Wide Analysis Of The Interaction Between The Endosymbiotic Bacterium Wolbachia And Its Drosophila Host, Zhiyong Xi, Laurent Gavotte, Yan Xie, Stephen L. Dobson

Entomology Faculty Publications

BACKGROUND: Intracellular Wolbachia bacteria are obligate, maternally-inherited, endosymbionts found frequently in insects and other invertebrates. The success of Wolbachia can be attributed in part to an ability to alter host reproduction via mechanisms including cytoplasmic incompatibility (CI), parthenogenesis, feminization and male killing. Despite substantial scientific effort, the molecular mechanisms underlying the Wolbachia/host interaction are unknown.

RESULTS: Here, an in vitro Wolbachia infection was generated in the Drosophila S2 cell line, and transcription profiles of infected and uninfected cells were compared by microarray. Differentially-expressed patterns related to reproduction, immune response and heat stress response are observed, including multiple genes that have …

Prostasin Is Expressed In Benign Prostatic Hyperplasia And Regulates Cell Proliferation And Invasion Via Inos, Icam-1, And Cycli, Meghan Hatfield

Electronic Theses and Dissertations

Prostasin is expressed in normal prostate epithelial cells but down-regulated in prostate cancers, while prostasin re-expression in invasive prostate cancer cells reduced invasion. We examined prostasin expression and function in benign prostatic hyperplasia (BPH). We evaluated prostasin expression in 12 BPH specimens by immunohistochemistry, and evaluated the impact of prostasin silencing by siRNA on the expression of the inducible nitric oxide synthase (iNOS), intercellular adhesion molecule-1 (ICAM-1), and cyclin D1, as well as on cell proliferation and invasion, using the BPH-1 human prostate epithelial cell line model. Prostasin expression was localized in the glands of BPH tissues by immunohistochemistry, in …

Role Of Transient Receptor Potential Canonical-6 (Trpc6) Channel In Metastasis Of Glioblastoma Multiforme, Rajarajeshwari Venkataraman Jan 2008

Role Of Transient Receptor Potential Canonical-6 (Trpc6) Channel In Metastasis Of Glioblastoma Multiforme, Rajarajeshwari Venkataraman

Electronic Theses and Dissertations

Glioblastoma multiforme (GBM) is one of the extremely fatal brain tumors. The main reason that makes it so lethal is its capability to invade and spread to other parts of CNS producing secondary tumors. Among other factors hypoxia, reduced oxygen availability, is linked to higher metastatic potential of cancers. Hypoxia causes numerous changes in genome and proteome of the cell. These changes help a normal cell to adapt to nutritional deficiency, but the same changes can increase the malignancy and metastasis in tumor cells. Extensive research by a number of curious scientists reveal that various pathways involving numerous proteins cross-talk …

Evaluation Of The Efficacy Of Chloroplast-Derived Antigensagainst Malaria, Melissa Schreiber Jan 2008

Evaluation Of The Efficacy Of Chloroplast-Derived Antigensagainst Malaria, Melissa Schreiber

Electronic Theses and Dissertations

Malaria is the most prevalent vector-borne parasitic disease worldwide and a major cause of death from infections. There is a great need to develop a low cost vaccine for malaria to control transmission of infection and impact of disease, due to the emergence of anti-malarial resistance. Two leading blood stage malarial vaccine candidates are the apical membrane antigen-1 (AMA-1) and the merozoite surface protein-1 (MSP-1). The aim of this project is to express malarial antigens in tobacco plants via plastid transformation and deliver them by subcutaneous or oral gavage of minimally processed transplastomic tissue to evaluate their efficacy to elicit …

Understanding How To Change: An Inductive Determination Of How Agents Of State Government Plan, Lead, And Sustain Change, Tim Rahschulte Jan 2008

Understanding How To Change: An Inductive Determination Of How Agents Of State Government Plan, Lead, And Sustain Change, Tim Rahschulte

Faculty Publications - College of Business

This dissertation started with a simple question: Why is it so difficult to lead successful organizational change initiatives? The dynamics of organizational behavior support a plethora of complex answers to this simple question. Indeed, literature abounds regarding the need for organizational change, individual transformation, and living system adaptation. However, Jaworski, Gozdz, and Senge (1998) noted that although much has been written about organizational change, there is little offered in literature about how to achieve change. Beer and Nohria (2000) added that existing organizational change theory is not sufficient to address organizational transformation especially relative to the people side of change. …

Simulated Effects Of Various Environmental Management Practices On Energy Consumption In Open And Confined Greenhouse Systems, Ilhami Yildiz, D. P. Stombaugh

BioResource and Agricultural Engineering

The objective of this study was to evaluate the effects of relative humidity, light management levels, minimum ventilation rates, CO2 enrichment and canopy size on energy consumption in three different greenhouse systems (conventional, open-loop heat pump, and confined heat pump) in winter, spring, and summer months. No difference was observed in energy consumption with relative humidity set point levels in winter due to the already low relative humidity levels. Some differences were observed in spring and summer due to extra dehumidification required to maintain the inside relative humidity at lower levels. Energy consumption in summer was reduced up to …
