Sample Prep For Proteomics Of Breast Cancer: Proteomics And Gene Ontology Reveal Dramatic Differences In Protein Solubilization Preferences Of Radioimmunoprecipitation Assay And Urea Lysis Buffers, Lambert Cm Ngoka

Chemistry Publications

Background

An important step in the proteomics of solid tumors, including breast cancer, consists of efficiently extracting most of proteins in the tumor specimen. For this purpose, Radio-Immunoprecipitation Assay (RIPA) buffer is widely employed. RIPA buffer's rapid and highly efficient cell lysis and good solubilization of a wide range of proteins is further augmented by its compatibility with protease and phosphatase inhibitors, ability to minimize non-specific protein binding leading to a lower background in immunoprecipitation, and its suitability for protein quantitation.

Results

In this work, the insoluble matter left after RIPA buffer extraction of proteins from breast tumors are subjected …

Analysis Of Secchi Depths And Light Attenuation Coefficients In The Louisiana-Texas Shelf, Northern Gulf Of Mexico, A. Lugo-Fernández, M. Gravois, T. Montgomery Jan 2008

Analysis Of Secchi Depths And Light Attenuation Coefficients In The Louisiana-Texas Shelf, Northern Gulf Of Mexico, A. Lugo-Fernández, M. Gravois, T. Montgomery

Gulf of Mexico Science

We analyzed Secchi depths (Zs), scalar downwelling irradiance, and transmissometer data collected during 10 cruises that spanned two and one-half years (May 1992-Nov. 1994) over the Louisiana-Texas shelf. The Zs increases from the inner shelf (depths ≤50 m) to the outer shelf and from nonsummer (Zs =10m) to summer (Zs = 14m). These changes in Zs are induced by the shelf's circulation and freshwater input, both of which vary seasonally, i.e., nonsummer to summer. Optical parameters (e.g., Zs), like the circulation, divide the shelf into an inner and outer shelf along the 50-m isobath. The …

Dietary Analysis Of Batfishes (Lophiiformes: Ogcocephalidae) In The Gulf Of Mexico, Bronson H. Nagareda, Jonathan M. Shenker Jan 2008

Dietary Analysis Of Batfishes (Lophiiformes: Ogcocephalidae) In The Gulf Of Mexico, Bronson H. Nagareda, Jonathan M. Shenker

Gulf of Mexico Science

Stomach content analyses, performed on three species of batfishes, Halieutichthys aculeatus, Ogcocephalus declivirostris, and Ogcocephalus pantosticus collected in the Gulf of Mexico in summer (June-July) and fall (Oct.-Nov.) 2002 and 2003, revealed a variety of benthic invertebrates, particularly gastropods, polychaete worms, and xanthid crabs. Schoener's dietary overlap indices (Sl) were calculated between the three species within the same seasons, and within each species between seasons. SI values indicated that each species consumed a different assemblage of prey and that two of the species exhibited temporal variation in diet.

Relative Abundance And Distribution Of Sand Seatrout (Cynoscion Arenarius) In Relation To Environmental Conditions, Habitat, And River Discharge In Two Florida Estuaries, Anthony R. Knapp, Caleb H. Purtlebaugh Jan 2008

Relative Abundance And Distribution Of Sand Seatrout (Cynoscion Arenarius) In Relation To Environmental Conditions, Habitat, And River Discharge In Two Florida Estuaries, Anthony R. Knapp, Caleb H. Purtlebaugh

Gulf of Mexico Science

The sand seatrout, Cynoscion arenarius (Ginsburg, 1930), is an abundant recreational and commercial species that resides primarily in the nearshore and estuarine waters of the Gulf of Mexico. We examined relative abundance and distribution of sand seatrout [individuals >100 mm standard length (SL)] in relation to environmental conditions and river discharge in the Tampa Bay (1997-2004) and Charlotte Harbor (1999-2004) estuaries on the west coast of Florida. Fish were collected during a long-term fisheries-independent monitoring program with a 183-m purse seine. Sand seatrout were most abundant over deep, muddy substrates devoid of seagrass. Smaller sand seatrout between145 mm SL and …

Effects Of Porous Mesh Groynes On Macroinvertebrates Of A Sandy Beach, Santa Rosa Island, Florida, U.S.A., W.J. Keller, C.M. Pomory

Gulf of Mexico Science

The use of porous mesh groynes to accrete sand and stop erosion is a relatively new method of beach nourishment. Five groyne, five intergroyne, and five control transects outside the groyne area on a beach near Destin, FL were sampled during the initial 3 mo after installment of groynes for Arenicola cristata (polychaete) burrow numbers, benthic macroinvertebrate numbers, and dry mass. Salinity, temperature, turbidity, and current velocity were measured at one location within the groyne site and control site. Current velocity was reduced and sand was accreted in the groyne site relative to the control site. Few significant changes or …

Patterns Of Distribution Along A Salinity Gradient In The Flatback Mud Crab Eurypanopeus Depressus, Julie Van Horn, S. Gregory Tolley Jan 2008

Patterns Of Distribution Along A Salinity Gradient In The Flatback Mud Crab Eurypanopeus Depressus, Julie Van Horn, S. Gregory Tolley

Gulf of Mexico Science

To measure the effect of salinity on estuarine communities, spatial and temporal variations in population distribution were investigated in the flatback mud crab Eurypanopeus depressus (Smith, 1869), a dominant species on oyster reefs in southwest Florida. Eurypanopeus depressus were collected using lift nets (1 m2 ) deployed at three morphologically homologous stations along a salinity gradient in each of three estuaries. They were sampled monthly during three seasonally wet and three seasonally dry months. An analysis of abundance showed that although this species can survive extreme salinities, it is most common in moderate environments. Biomass is also highest at …

Life History Parameters Of Gulf Flounder (Paralichthys Albigutta) From Northwest Florida, Gary R. Fitzhugh, William A. Fable Jr., Wallace L. Trent

Gulf of Mexico Science

Age, growth, natural and total mortality, a length-weight relationship, reproductive seasonality, sex ratio, maturity, and reproductive potential by size were estimated for gulf flounder, Paralichthys albigutta, from northwest Florida. The study used a fishery-independent approach during three annual migratory spawning seasons to sample gulf flounder offshore (spearing by divers) and inshore (multipanel trammel nets). We found gulf flounder to be slightly smaller at L∞ than other paralichthids (using either von Bertalanffy or damped growth functions), but gulf flounder obtained larger sizes and older ages than previously thought for this species [females to 575 mm total length (TL) and age …

Genetic Structure Of Halodule Wrightii Populations From The Laguna Madre Region In The Western Gulf Of Mexico, Patrick D. Larkin, Krista L. Heideman, Joyce E. Parker, Beau Hardegree Jan 2008

Genetic Structure Of Halodule Wrightii Populations From The Laguna Madre Region In The Western Gulf Of Mexico, Patrick D. Larkin, Krista L. Heideman, Joyce E. Parker, Beau Hardegree

Gulf of Mexico Science

A random amplified polymorphic DNA assay was used to assess genetic variation in populations of the seagrass Halodule wrightii (Ascherson) from the western Gulf of Mexico. This region includes one of the world's few hypersaline lagoons (Laguna Madre) and contains the vast majority of seagrasses on the Texas coast. Results indicate a moderate amount of genetic diversity among populations. The highest level (Hc= 0.33) was found in a population from Nueces Bay, a disturbed site in the Coastal Bend area, whereas the lowest was found in a Lower Laguna Madre population (Hc = 0.15). Genetic differentiation generally …

Improving Post-Approval Risk Surveillance For Drugs: Active Post-Market Risk Identification, Matthew Gordon Jan 2008

Improving Post-Approval Risk Surveillance For Drugs: Active Post-Market Risk Identification, Matthew Gordon

Michigan Telecommunications & Technology Law Review

Pre-approval clinical trials cannot possibly ensure that a drug will not have disastrous side effects once it arrives on the market. Post-approval drug safety data gathering was put in place to address this problem, but as implemented, it has not proven to be as effective as hoped. Congress recently overhauled the legislation regarding post-approval drug risk identification, and in doing so made a deliberate decision to put much of the burden of post-approval drug surveillance on the FDA through data mining. Further, the legislation gave the FDA the power to require post-approval clinical trials from drug makers only in limited …
