The chemical exchange saturation transfer (CEST) efficiency for a series Eu3+-based tetraamide complexes bearing p-substituents on a single coordinating pendant arm is highly sensitive to water exchange rates. The CEST effect increases in the order Me < MeO < F ∼ CO2tBu < CN ≪ H. These results show that CEST contrast can be modulated by changes in electron density at a single ligating atom, and this forms the basis of creating imaging agents that respond to chemical oxidation and reduction.

More Than The Sum Of Its Parts: Physical And Mechanistic Coupling In The Phage Lambda Red Recombinase, Gokhan Tolun

In many dsDNA viruses, a single-strand-annealing homologous recombination (SSA) reaction is catalyzed by a pair of proteins. In phage lambda, this system is called Red, and is composed of lambda exonuclease (Lambda-Exo, a 5' to 3' dsDNA exonuclease), and Red-Beta (a ssDNA binding protein and annealase). I examined the physical and mechanistic coupling of Lambda-Exo and Red-Beta and confirmed that these proteins form a complex with a 1:1 subunit stoichiometry. The size of this complex was shown to be close to one MDa, possibly composed of 12 Lambda-Exo and 12 Red-Beta monomers. Red-Beta decreased the extent of digestion of dsDNA …

Fields of thin-film porous anodic alumina PAA are fabricated within a SiO2 support and on independently addressable underlying metal pads. The underlying metallization provides a means for unique postprocessing to be performed on the PAA fields. Customized postprocessing is demonstrated with the synthesis of single-walled carbon nanotubes SWCNTs from an embedded catalyst in the PAA, followed by selective decoration of the SWCNTs from different PAA fields with dissimilar nanoparticles. Achieving uniquely functionalized fields of PAA on a single chip provides a scalable integration platform to be used in multiplexed chemical and biological sensing or nanoelectronic devices. © 2008 American Institute …

A Threading-Based Method (Findsite) For Ligand-Binding Site Prediction And Functional Annotation, Michal Brylinski, Jeffrey Skolnick

The detection of ligand-binding sites is often the starting point for protein function identification and drug discovery. Because of inaccuracies in predicted protein structures, extant binding pocketdetection methods are limited to experimentally solved structures. Here, FINDSITE, a method for ligand-binding site prediction and functional annotation based on binding-site similarity across groups of weakly homologous template structures identified from threading, is described. For crystal structures, considering a cutoff distance of 4 Å as the hit criterion, the success rate is 70.9% for identifying the best of top five predicted ligand-binding sites with a ranking accuracy of 76.0%. Two-dimensional and three-dimensional optical coherence tomography is obtained by differential imaging of full-frame interference images using a white light source. Full-color tomographic imaging is also possible by processing the three-color channels of the interference images. A technique is described to obtain two-dimensional OCT images with full natural color representation. In a particular embodiment, the interference image is acquired using a color camera and the three-color channels are processed separately, recomposing the final image. Herein, we describe our experience of involving high school students in the process of studying wild coyotes in and around the communities where they live. Cyclin D1 is an important cell cycle regulator but in cancer its overexpression also increases cellular migration mediated by p27KlP1 stabilization and RhoA inhibition. Recently, a common polymorphism at the exon 4-intron 4 boundary of the human cyclin D1 gene within a splice donor region was associated with an altered risk of developing cancer. Altered RNA splicing caused by this polymorphism gives rise to a variant cyclin D1 isoform termed cyclin D1b, which has the same N-terminus as the canonical cyclin D1a isoform but a distinct C-terminus. Section II provides a brief background on the sources of international law used to advocate for and enforce LGBT rights. Section III describes some examples of recent and current legal issues arising out of situations in which nations have decided not to follow their international treaty obligations and examines why these nations might have so chosen. Molecular analysis of coprolites from Hinds Cave, Texas recovered chloroplast DNA sequences. The sequences were interpreted as evidence of diet. We analyzed 19 Hinds Cave coprolites to evaluate the potential sources of the chloroplast DNA (cpDNA) and compared our results to previous studies. This review shows that some cpDNA sequences could be from well-known prehistoric plants foods. Some other sequences could have come from ambient plant material in the guts of small animals eaten by humans in antiquity. Using pollen concentration analysis, we identify sources of ambient plant material which could have been inhaled or imbibed. It is even possible …

Objective: To examine factors in early life (up to age 5 years) that are associated with objectively measured physical activity in 11-12 year olds.

Design: Prospective cohort study

Setting: Avon longitudinal study of parents and children, United Kingdom.

Participants: Children aged 11-12 years from the Avon longitudinal study of parents and children.

Main outcome measure: Physical activity levels in counts per minute (cpm) and minutes of moderate to vigorous physical activity for seven days measured with a uniaxial actigraph accelerometer.

The objective of this dissertation was to examine sailfish (Istiophorus platypterus) and blue marlin (Makaira nigricans) spawning in the Straits of Florida, with a specific focus on 1) the physical and biological characteristics of the spawning environment, and 2) the role of the region within the broader spawning patterns of these two species. In order to accomplish these objectives, two years of monthly ichthyoplankton collections and physical measurements across the Straits of Florida were combined with a finer-scale Lagrangian study. Acid-sensing ion channels are ligand-gated cation channels, gated by extracellular H+. H+ is the simplest ligand possible, and whereas for larger ligands that gate ion channels complex binding sites in the three-dimensional structure of the proteins have to be assumed, H+ could in principle gate a channel by titration of a single amino acid. Experimental evidence suggests a more complex situation, however. For example, it has been shown that extracellular Ca2+ ions compete with H+; probably Ca2+ ions bound to the extracellular loop of ASICs stabilize the closed state of the channel and have to be displaced before the channel …

Changes in membrane lipid composition play important roles in plant adaptation to and survival after freezing. Plant response to cold and freezing involves three distinct phases: cold acclimation, freezing, and post-freezing recovery. Considerable progress has been made toward understanding lipid changes during cold acclimation and freezing, but little is known about lipid alteration during post-freezing recovery. We previously showed that phospholipase D (PLD) is involved in lipid hydrolysis and Arabidopsis thaliana freezing tolerance. GDP-D-mannose 4,6-dehydratase (GMD) is a key enzyme involved in the synthesis of 6-deoxyhexoses in prokaryotes and eukaryotes. Paramecium bursaria chlorella virus-1 (PBCV-1) encodes a functional GMD, which is unique among characterized GMDs because it also has a strong stereospecific NADPH-dependent reductase activity leading to GDP-D-rhamnose formation (Tonetti, M., Zanardi, D., Gurnon, J., Fruscione, F., Armirotti, A., Damonte, G., Sturla, L., De Flora, A., and Van Etten, J.L. (2003) J. Biol. Chem. 278, 21559–21565). We study the viscoelasticity of an active solution of polar biofilaments and motor proteins. Using a molecular model, we derive the constitutive equations for the stress tensor in the isotropic phase and in phases with liquid crystalline order. The stress relaxation in the various phases is discussed. Contractile activity is responsible for a spectacular difference in the viscoelastic properties on opposite sides of the order-disorder transition.
