The subject invention concerns a method of inhibiting an RNA virus infection within a patient by increasing the amount of 2-5 oligoadenylate synthetase (2-5 AS) activity within the patient. Preferably, the preventative and therapeutic methods of the present invention involve administering a nucleotide encoding 2-5 AS, or at least one catalytically active fragment thereof, such as the p40, p69, p100 subunits, to a patient in need thereof. The present inventors have determined that overexpression of 2-5AS causes a reduction in epithelial cell damage, reduction in infiltration of mononuclear cells in the peribronchiolar and perivascular regions, and reduction in thickening of …

Vaccines for conferring immunity in mammals to infective pathogens are provided, as well as vectors and methods for plastid transformation of plants to produce protective antigens and vaccines for oral delivery. The invention further provides transformed plastids having the ability to survive selection in both the light and the dark, at different developmental stages by using genes coding for two different enzymes capable of detoxifying the same selectable marker, driven by regulatory signals that are functional in proplastids as well as in mature chloroplasts. The karst research community has known that traditional numerical groundwater flow codes ignore the non-Darcian, dual-permeability components of flow that can occur in karst aquifers. In this study, the potential limitations of using such tools are quantified by evaluating the relative performances of 3 groundwater flow models at a test-site near Weeki Wachee, Florida, in the dual-permeability Upper Floridan aquifer. MODFLOW-2005 and MODFLOW-2005 Conduit Flow Process (CFP), a Darcian/non-Darcian, dual-permeability groundwater flow code recently developed by the U.S. Geological Survey, are used in this study.

Like coral reefs worldwide, the Florida Reef Tract has dramatically declined within the past two decades. Surveillance Of Cirrhosis For Hepatocellular Carcinoma: A Cost-Utility Analysis., Coon J Thompson, G. Rogers, P. Hewson, D. Wright, R. Anderson, S. Jackson, S. Ryder, M. Cramp, K. Stein

Using a decision-analytic model, we evaluated the effectiveness and cost-effectiveness of surveillance for hepatocellular carcinoma (HCC) in individuals with cirrhosis. Separate cohorts with cirrhosis due to alcoholic liver disease, hepatitis B and hepatitis C were simulated. Results were also combined to approximate a mixed aetiology population. Comparisons were made between a variety of surveillance algorithms using alpha-foetoprotein (AFP) assay and/or ultrasound at 6- and 12-monthly intervals. Parameter estimates were obtained from comprehensive literature reviews. Uncertainty was explored using one-way and probabilistic sensitivity analyses. In the mixed aetiology cohort, 6-monthly AFP+ultrasound was predicted to be the most effective strategy. The model …

Movement And Distribution Of Juvenile Bull Sharks, Carcharhinus Leucas, In Response To Water Quality And Quantity Modifications In A Florida Nursery, Lori A. Ortega

Movement, distribution, and habitat use of juvenile bull sharks were examined in two studies using manual and passive acoustic telemetry. Research was conducted in the Caloosahatchee River, which serves as nursery habitat for this species, and is highly impacted due to anthropogenic alterations in water quality and quantity via dams and locks. Manual tracking yielded fine-scale results for eight individuals on home range size, rate of movement, swimming depth, linearity, direction of travel, tidal influence, diel pattern, as well as correlation with environmental variables. Changes in salinity, temperature, dissolved oxygen, turbidity, and pH played a role on the distribution of …

The Meyers et al. (2007) Tampa Bay Model produces water level and three-dimensional current and salinity fields for Tampa Bay. It is capable of computing temperature but is presently run without active thermodynamics. Variations in water temperature are driven by heat exchange at the water-atmosphere boundary and advective heat flux at the mouth of the bay. The net heat exchange surface boundary condition is required for computations of three-dimensional temperature fields. In the simultaneous estimation of a large number of related quantities, multilevel models provide a formal mechanism for efficiently making use of the ensemble of information for deriving individual estimates. In this article we investigate the ability of the likelihood to identify the relationship between signal and noise in multilevel linear mixed models. Specifically, we consider the ability of the likelihood to diagnose conjugacy or independence between the signals and noises. Our work was motivated by the analysis of data from high-throughput experiments in genomics. The proposed model leads to a more flexible family. However, we further demonstrate that adequately …

Chemistry and Biochemistry

The sodium proton exchanger (NHE) is particularly important in maintaining the intracellular pH in human heart and brain. Under anaerobic conditions (i.e., ischemia), a shift from oxidative to nonoxidative glycolysis occurs. The resultant decrease in the intracellular pH activates NHE, which increases the intracellular sodium, initiating the sequence of physiological events that lead to cell death. Thus, there has been great interest in the development of compounds that inhibit NHE. Indeed, potent NHE inhibitors are available. However, a fundamental impediment to the field is the delivery of these compounds to poorly vascularized tissues during the early phases of ischemia when …

Aim: To determine the predictive factors for early aspiration in liver abscess.

Methods: A retrospective analysis of all Patients with liver abscess from 1995 to 2004 was performed. Abscess was diagnosed as amebic in 661 (68%) Patients, pyogenic in 200 (21%), indeterminate in 73 (8%) and mixed in 32 (3%). Multiple logistic regression analysis was performed to determine predictive factors for aspiration of liver abscess.

Results: A total of 966 Patients, 738 (76%) male, mean age 43 17 years, were evaluated: 540 Patients responded to medical therapy while adjunctive percutaneous aspiration was performed in 426 Patients. Predictive factors for aspiration …

Ras genes are frequently mutated in human cancers and present compelling targets for therapeutic intervention. While previous attempts to directly inhibit oncogenic Ras function have largely been unsuccessful use of targeted agents to inhibit the three primary oncogenic pathways activated by mutated Ras: RalGEF-Ral, PI3K-Akt and Raf- MEK-Erk, is an area of intense investigation. Here, we describe the ability of a novel pharmacological inhibitor of geranylgeranyltransferase I, GGTI-2417, to inhibit Ral prenylation and localization. We further used a Ral rescue system to selectively preserve RalA and RalB function and localization during GGTI-2417 treatment and determine the precise roles for inhibition …

Two-dimensional (2-D) micro-autoradiography is typically used to identify the location of a radio-labeled ligand bound to a cellular target in tissue sections. Data, such as a histological image, combined with the autoradiographic data provide a spatial relationship of the radiolabel to cellular structures. However, the disadvantage of 2-D imaging is that it only provides a local distribution of the radiolabel within a tissue slice, and not a volumetric regional distribution in the structure of interest. The emergence of West Nile virus (WNV) has raised important questions about the capacity of the public health infrastructure to implement surveillance and control programs for WNV and other emerging or re-emerging arboviruses in the United States. Florida's mild climate supports year round enzootic transmission of WNV, St. Louis encephalitis virus (SLEV), and Eastern Equine Encephalitis virus (EEEV). It is unknown what effect the establishment of WNV (in 2001) will have on SLEV transmission in Florida, where these closely related flaviviruses share amplifying hosts, habitats, and vectors.

The Department of Chemistry at Western Kentucky University recently incorporated a net-worked MeasureNet data acquisition and analysis system into in the second semester General Chemistry laboratories. Our investigation team has conducted a study to evaluate the effects of the MeasureNet system on student learning and performance on a Beer's Law laboratory. We focused on two phenomena: the effect of the MeasureNet system on 1) student learning of specific content material and 2) student interest in chemistry and science in general.
