A Deletion Mutant Of Vitronectin Lacking The Somatomedin B Domain Exhibits Residual Plasminogen Activator Inhibitor-1-Binding Activity, Christine R. Schar, Grant E. Blouse, Kenneth H. Minor, Cynthia B. Peterson

Vitronectin and plasminogen activator inhibitor-1 (PAI-1) are important physiological binding partners that work in concert to regulate cellular adhesion, migration, and fibrinolysis. The high affinity binding site for PAI-1 is located within the N-terminal somatomedin B domain of vitronectin; however, several studies have suggested a second PAI-1-binding site within vitronectin. To investigate this secondary site, a vitronectin mutant lacking the somatomedin B domain (rΔsBVN) was engineered. The short deletion had no effect on heparin-binding, integrin-binding, or cellular adhesion. Binding to the urokinase receptor was completely abolished while PAI-1 binding was still observed, albeit with a lower affinity. Analytical ultra-centrifugation on …

The widely accepted models for the role of carboxysomes in the carbon-concentrating mechanism of autotrophic bacteria predict the carboxysomal carbonic anhydrase to be a crucial component. The enzyme is thought to dehydrate abundant cytosolic bicarbonate and provide ribulose 1.5-bisphosphate carboxylase/oxygenase (RubisCO) sequestered within the carboxysome with sufficiently high concentrations of its substrate, CO2, to permit its efficient fixation onto ribulose 1,5-bisphosphate. In this study, structure and function of carboxysomes purified from wild type Halothiobacillus neapolitanus and from a high CO2-requiring mutant that is devoid of carboxysomal carbonic anhydrase were compared. Compaction of DNA by histone proteins helps to package the DNA into the nucleus at the expense of acting as a bather to biological processes that require access to the DNA. Addition and removal of acetyl groups from the histone tails with the help of enzymes known as histone acetyltransferases (HATs) and histone deacetylases (HDACs) appear to play a critical role in maintaining the structure of chromosomes and the regulation of gene expression. This dissertation—Toward a New Perspective in Literary Criticism: Transgenerational Psychoanalysis and Anasemic Procedures"—is a contribution to literary criticism. It offers a new perspective in psychoanalytical criticism on how characters in literary narratives can be interpreted. By examining key concepts in psychoanalysis, which have been used in the humanities for decades, the present inquiry draws on transgenerational psychology, as elaborated by German and French theorists, and anasemic interpretations generated by M. Torok and N. Abraham's psychoanalytical approach. It has been shown that fibronectin fragments (Fn-fs) greatly augment cartilage proteoglycan (PG) depletion by up-regulating matrix metalloproteinases (MMPs) and catabolic cytokines. The activities of three different Fn-fs have been studied: an amino terminal 29-kDa which is most potent with a weak affinity to the α5β1, integrin, the receptor for native fibronetin (Fn); a 50-kDa gelatin-binding Fn-f with no known interaction with Fn receptor; and a 140-kDa Fn-f which is the least potent and has a RGDS sequence which can bind to the α5β1 integrin. Neural Characteristics Of Affectionate Communicators: Trait Affection And Asymmetry In The Prefrontal Cortex, Robert Joel Lewis

Individual differences in thresholds for affectionate communication should be reflected by differences in neurological structure and function. A theoretical schema from several overlapping literatures including evolutionary psychology, social neuroscience, fundamental personality, and communication are examined to make the case that high-affection communicators have greater relative electrical activity in the left prefrontal cortex (PFC) versus the right PFC reflected in asymmetrical baseline EEG recordings. Participants (N=16) reported trait-affection levels using Floyd?s (2002) TAS-G, which measures an individual?s threshold for expressing affection. Regulation Of Pro-Inflammatory Responses By Ligands Of The Nuclear Receptor Pparγ, Andrew P. Woster

Inflammation-related neurotoxicity is evident in a number of different models neurodegenerative conditions, including Alzheimer's and Parkinson's diseases, Multiple Sclerosis, and HIV-associated dementia. Peroxisome proliferator-activated receptor gamma (PPARγ) agonists inhibit a number of select pro-inflammatory changes in models of CNS and systemic inflammation. Recent reports suggest that these anti-inflammatory effects are due to mechanisms other than canonical nuclear receptor-mediated transcriptional alteration. Results reported here demonstrate that rosiglitazone, a PPARγ-activating thiazolidinedione, decreases tumor necrosis factor α (TNFα) secretion in both primary microglia and the monocytic cell line, THP-1. Light Induced Changes In The Photoreceptor Outer Segments, Vikram Palamalai

Light induced retinal degeneration in animals has been used to study human retinal degenerative disorders. Apoptosis is the final common pathway for photoreceptor degeneration in both the experimental model and human disease but the mechanism is unknown. Oxidative stress plays a role because antioxidants have been shown to be protective against experimental degeneration. Inhibitors of nitric oxide synthases (NOS) are also protective suggesting a role for nitric oxide. Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) a key enzyme in glycolysis was identified as one of the proteins undergoing significant nitration in our studies on light induced retinal degeneration. The Effects Of Perchlorate Exposure On A Model Vertebrate Species: The Threespine Stickleback, Richard R. Bernhardt

Few studies have examined the effects of chronic perchlorate exposure during multiple stages of development, and fewer still have analyzed the effects of perchlorate over multiple generations. Perchlorate exposure is known to cause thyrocyte hypertrophy (suggesting glandular stimulation), interference with thyroid hormone synthesis, and ultimately altered levels of circulating thyroid hormones, but whether these effects represent adaptive mechanisms or actual impairment is often debated within the scientific community. My research attempts to clarify whether exposure to environmentally relevant concentrations of perchlorate cause impairment at the organismal level. Reactions on naturally abundant hematite (alpha-Fe2O 3) surfaces significantly influence the transport and bio-availability of a number of important nutrients and contaminants. The surface reactivity of alpha-Fe2O3 is dependent on the surface structure, i.e. the identity and coordination of chemical moieties exposed at the surface. The surface structure is strongly influenced by the presence of water and common aqueous species such as Fe(II). Therefore, it is important to understand how the surface structure evolves in the presence of water and aqueous species (e.g. Fe(II)) in order to model the surface reactivity of hematite in natural aquatic systems. Everyone experiences pain to one degree or another, but pain that has outlived its usefulness in assisting the body to avoid injury, and causes other physical and emotional complications, is known as chronic pain. In American medicine, chronic pain is described as that which persists longer than six months and is not relieved by standard medical care. Chronic pain usually leads to a spectrum of other physical and emotional complications, including sleep disturbance, loss of appetite, and severe depression, among many others. Who Is In Charge Here? A Feminist Communicology Of Followership And Leadership In An Academic Organization, Robert Locke Jordan

This feminist critical study explicates the ways that followership is conceptualized at an academic organization in the Pacific Northwest. Through the use of qualitative methods, stories were solicited providing descriptions of events that define the hegemonically masculine ways that followership is conceptualized, suggesting the need for a feminist critical analysis and revisioning. A number of themes emerged from conversational interviews including: conceptual verisimilitude, archetypes of leadership, alternative conceptions of followership, the role of action in leadership and followership, and the emergent organization.
