Promotion Of Tumor Cell Death Through The Induction Of, Tuyen Nguyen

Microtubule poisons have proven to be effective in the treatment of a variety of malignancies. Although taxol-based derivatives promote microtubule stabilization, there is continuing interest in compounds that, like colchicines, act as microtubule destabilizing agents. Previous work from this laboratory showed that the novel microtubule poison, JG- 03-14, was active against breast tumor cells, promoting autophagic cell death. In the current work, we studied the influence of JG-03-14 on p53 wild type HCT116 colon carcinoma cells. A crystal violet sensitivity assay indicated that JG-03-14 induced growth inhibition, with 75% suppression of growth evident at a concentration of 500 nM. Time …

Metallic nanoparticles are unique materials for optical, electronic, catalytic, and sensing applications. Due to the vast flexibility in controlling the surface chemistry of these particles through functionalization there is a great deal of interest in using metallic nanoparticles as building blocks in the development of more complex nanostructures through the use of a 'bottom-up' approach. Using self assembly techniques, one can exploit spontaneous chemical interactions to build complex constructs on the nanometer scale.

Towards this end, gold nanorods have been synthesized and modified with various polymers, inorganic oxides and organic ligands to establish principles for self-assembly of these unique nanomaterials. …

A novel microfluidic device that can selectively and specifically isolate exceedingly small numbers of circulating tumor cells (CTCs) through a monoclonal antibody (mAB) mediated process by sampling large input volumes (≥1 mL) of whole blood directly in short time periods (<37 >min) was demonstrated. The CTCs were concentrated into small volumes (190 nL), and the number of cells captured was read without labeling using an integrated conductivity sensor following release from the capture surface. Potato production requires high phosphorus (P) application with potential negative environmental or nutritional consequences for potato as well as for subsequent crops. Impacts of high available P on yield and plant nutrition of species in potato cropping rotations are inadequately understood, and could result in antagonistic interactions with cationic micronutrients such as zinc (Zn) and manganese (Mn). Three hydroponic experiments were conducted with Russet Burbank potato to elucidate P and Zn relationships and associated interactions with other nutrients. Sequence Information Encoded In Dna That May Influence Long-Range Chromatin Structure Correlates With Human Chromosome Functions., Taichi E. Takasuka, Alfred Cioffi, Arnold Stein

Little is known about the possible function of the bulk of the human genome. We have recently shown that long-range regular oscillation in the motif non-T, A/T, G (VWG) existing at ten-nucleotide multiples influences large-scale nucleosome array formation. In this work, we have determined the locations of all 100 kb regions that are predicted to form distinctive chromatin structures throughout each human chromosome (except Y). Using these data, we found that a significantly greater fraction of 300 kb sequences lacked annotated transcripts in genomic DNA regions ≥300 kb that contained nearly continuous chromatin organizing signals than in control regions. Background: The World health Organization (WHO) declares dengue and dengue hemorrhagic fever to be endemic in South Asia. Despite the magnitude of problem, no documented evidence exists in Pakistan which reveals the awareness and practices of the country's adult population regarding dengue fever, its spread, symptoms, treatment and prevention. Body Shape Dissatisfaction: Patterns Of Concern Among Subgroups Of College Freshmen Women, Kristina S Withers Hansen

This study seeks to determine if there are patterns of differences according to college major in terms of the percentage of freshmen women who score in the clinically significant range on a measure of attitude toward body shape. Participants (N = 1,982) completed a demographic questionnaire and the Body Shape Questionnaire (BSQ) and were divided into subgroups according to their stated college major. Descriptive statistics regarding BSQ scores were calculated for the total sample and subgroups. Results indicated that a greater percentage of participants in Theater, Communications, Psychology, Dance, Business, and Dietetics majors scored within clinically significant ranges on the …

FoxO proteins are transcriptional factors acting downstream of the tumor suppressor PTEN. Their activity is negatively regulated by AKT-mediated phosphorylation. Our previous studies showed a mutual suppression between PTEN and the androgen receptor (AR) in regulating growth and apoptosis in prostate cancer (PCa) cells. We hypothesize that nuclear FoxO proteins are involved in mediating this mutual antagonism. In this dissertation, we report that PTEN inhibits AR activity through FoxO1 and provide evidence for the involvement of FoxO factors in the androgen-mediated suppression of PTEN-induced apoptosis. The interest in metal-organic materials namely, coordination polymers and metal-organic frameworks has risen dramatically over the past few years. To a certain extent this interest is a consequence of the realization chemists have discovered how to play a form of molecular Lego® in which metal cations or metal clusters represent the bricks (or nodes) and organic ligands such as 4,4-bipyridine (bipy) or benzene-dicarboxylate represents the glue (or spacers). A district-wide school garden project has been initiated in the Toledo District of Southern Belize in response to reported high rates of poverty and undernutrition. This paper will discuss research conducted in the summer of 2007 with Q'eqchi' Maya in the Toledo District to determine the effect of school gardens on household diet and gardening, the composition of the household diet, and the makeup of homegardens. This paper explores the contemporary role of fathers in child-driven materialistic consumption, a little explored territory. Commencing with an exploration of the child consumer and their influence in personal and family spending, both of which have grown considerably over that last number of decades, followed by an analysis of changes in families and the role fathers now play in the aforementioned. We have developed the Proteome Discovery Pipeline, a stand-alone bioinformatics platform used for LC/MS data analysis and biomarker discovery. Data is processed in a series of self-contained analytical steps using modules that are controlled by a graphical user interface. The user interface was developed in Visual C++ 6.0 and provides a multi-threaded, tabbed user interface with each tab representing a step in the analysis process. Modules included are spectrum deconvolution, alignment, normalization, significance tests and pattern recognition. With multiple metazoan genomes in each family being sequenced promoter analysis is becoming a useful tool in genomic analysis. Aligning the promoter regions in the DNA of C. elegans and C. briggsae identifies conserved promoter elements. While not all promoter elements are conserved and not all conserved regions are promoter elements, we find that conservation is a useful method for determining promoter complexity. Promoter complexity identifies which genes have particularly interesting regulation, identifying gene groups with a strong promoter complexity signal and cases where a gene's promoter complexity differs from the group's promoter complexity.

For several groups of organisms multiple genomes have been sequenced either completely or to the unfinished shotgun sequencing stage. Comparative sequence analysis can identify functional sequence elements for pairs of organisms that diverged far enough in the past to allow mutational drift of non-conserved sequences. While analysis of a pair of genomes identifies many of these functional elements adding additional genomes allows additional information to be elicited.

The reported is a method for the preparation of stable dispersions of narrowly dispersed cholesterol crystals, which may provide an excellent model for the aggregation studies thereof. The effects of experimental parameters that include concentration, pH, ionic strength, temperature, and aging on the resulting morphologies are acknowledged. Regardless of the cause of destabilization, the multilayered aggregates of cholesterol particles are formed by “face-to-face” stacking of the original platelets, as promoted by higher surface charge density on particle sides comparing to their faces. Phytophthora erythroseptica Pethyb. is a devastating fungal pathogen of potato (Solanum tuberosum). The pathogen causes a disease known as pink rot. Pink rot results in necrosis and decay of tubers; and is responsible for major losses pre and post harvest. Attributes of the disease are progressing toward epidemic proportions. The Anaxyrus boreas species group currently comprises four species in western North America including the broadly distributed A. boreas, and three localized species, Anaxyrus nelsoni, Anaxyrus exsul and Anaxyrus canorus. Phylogenetic analyses of the mtDNA 12S rDNA, cytochrome oxidase I, control region, and restriction sites data, identified three major haplotype clades. The Northwest clade (NW) includes both subspecies of A. boreas and divergent minor clades in the middle Rocky Mountains, coastal, and central regions of the west and Pacific Northwest. The Southwest (SW) clade includes A. exsul, A. nelsoni, and minor clades in southern California. …

Fully compliant bistable mechanisms have been proposed to be used as threshold accelerometers. The advantages to using these devices are that they require no external power to operate and maintain their sensing state. Using this characteristic the devices can be integrated with passive radio frequency identification tags (RFID). This allows for the sensing package to lay dormant with no maintenance needed until the sensor is read by the RFID reader. Behavioral Parent Training refers to a broad range of instructional programs that teach parents and other caregivers ways to build and change behavioral repertoires of children. Most, if not all, such programs employ Behavioral Skills Training (modeling, prompting, role-play practice and feedback) to teach parenting skills.
