The Role Of Microrna-155 And Microrna-146a As Putative Oncomirs In The Tumor Progression Of Prostate Cancer, Jennifer Hoyt Jul 2008

The Role Of Microrna-155 And Microrna-146a As Putative Oncomirs In The Tumor Progression Of Prostate Cancer, Jennifer Hoyt

Theses and Dissertations

Prostate cancer is the most common cancer occurring in males. The identification of novel microRNAs (miRs) that contribute to tumor progression represents prospective treatment targets. miRs are small non-coding RNAs important in gene regulation with specific tissue expression patterns. Each miR is thought to affect the expression of hundreds of different RNA targets. Two putative oncomiRs, miR-155 and miR-146a, were shown to be differentially expressed in the human derived, prostate cell sublines M12 and F6. Quantification of endogenous miR expression showed high levels in the metastatic M12 cell line versus low in its weakly tumorigenic F6 variant. The restoration of …

The Channel For Gay America? A Cultural Criticism Of The Logo Channel’S Commercial Success On American Cable Television, Michael Johnson Jr. Jul 2008

The Channel For Gay America? A Cultural Criticism Of The Logo Channel’S Commercial Success On American Cable Television, Michael Johnson Jr.

USF Tampa Graduate Theses and Dissertations

Logo currently holds a self-described monopoly as the "Gay Channel for America." Logo stands alone as the single most concentrated national-level vehicle of LGBT (Lesbian, Gay, Bisexual and Transgendered) visibility in the post millennial television era. The Logo Channel has reaped financial rewards from its strategy as a business entity, as LGBT American television viewers embraced its presence as a signifier to America that gays and lesbians have finally "made it".

First, any claim to a monopoly deserves critical attention for its place in mainstream television, for its business practices, and for the power it holds in representing and targeting …

Comparative Cytotoxicity Of An Fda-Approved Cancer Drug To Extracts Of Atriplex Confertifolia On Human Breast And Cervical Cancer Cells, Christopher James Capua

Theses and Dissertations

The severity and number of people affected by cancer is a worldwide problem with millions of people affected annually. The search for treatment and cures of cancer continues to be a global effort. As part of this global effort, many natural products have been tested against cancer cell lines, most from plants located in tropical regions. However, this study reports that extracts of Atriplex confertifolia, a native North American plant, has significant bioactivity against human breast cancer cell lines MCF-7, 435, and 231, and HeLa cells (cervical cancer cells). The bioactivity of A. confertifolia extracts of these cells lines was …

Estimating The Causal Effect Of Lower Tidal Volume Ventilation On Survival In Patients With Acute Lung Injury, Weiwei Wang, Daniel Scharfstein, Roy Brower, Dale Needham

Johns Hopkins University, Dept. of Biostatistics Working Papers

Acute lung injury (ALI) is a condition characterized by acute onset of severe hypoxemia and bliateral pulmonary infiltrates. ALI patients typically require mechanical ventilation in an intensive care unit. Low tidal volume ventilation (LTVV), a time-varying dynamic treatment regime, has been recommended as an effective ventilation strategy. This recommendation was based on the results of the ARMA study, a randomized clinical trial designed to compare low vs. high tidal volume strategies (ARDSNetwork, 2000) . After publication of the trial, some critics focused on the high non-adherence rates in the LTVV arm suggesting that non-adherence occurred because treating physicians felt that …

Development Of Methods For The Genetic Manipulation Of Flavobacterium Columnare, Andrew M. Staroscik, David W. Hunnicutt, Kate E. Archibald, David R. Nelson

Cell and Molecular Biology Faculty Publications

Background:

Flavobacterium columnare is the causative agent of columnaris disease, a disease affecting many freshwater fish species. Methods for the genetic manipulation for some of the species within the Bacteroidetes, including members of the genus Flavobacterium, have been described, but these methods were not adapted to work with F. columnare.

Results:

As a first step toward developing a robust set of genetic tools for F. columnare, a protocol was developed to introduce the E. coli – Flavobacterium shuttle vector pCP29 into F. columnare strain C#2 by conjugal mating at an efficiency of 1.5 × 10-3 …

Unh Monitoring Activities That Support The National Coastal Assessment In 2007, Philip Trowbridge

PREP Reports & Publications

The National Coastal Assessment is an Environmental Protection Agency program to monitor the health of the nation’s estuaries using nationally standardized methods and a probabilistic sampling design. Dedicated EPA funding for the National Coastal Assessment ceased after 2006. Therefore, the NH Department of Environmental Services and the New Hampshire Estuaries Project contributed funds to continue a portion of the National Coastal Assessment in 2007. Water quality measurements were successfully made during 2007 at 25 randomly located stations throughout the Great Bay Estuary and Hampton-Seabrook Harbor. These data will be combined with samples collected in 2006 for probabilistic assessments of estuarine …

Differences In A Conformational Equilibrium Distinguish Catalysis By The Endothelial And Neuronal Nitric-Oxide Synthase Flavoproteins, Robielyn P. Ilagan, Mauro Tiso, David Konas, Craig Hemann, Deborah Durra, Russ Hille, Dennis J. Stuehr Jul 2008

Differences In A Conformational Equilibrium Distinguish Catalysis By The Endothelial And Neuronal Nitric-Oxide Synthase Flavoproteins, Robielyn P. Ilagan, Mauro Tiso, David Konas, Craig Hemann, Deborah Durra, Russ Hille, Dennis J. Stuehr

Department of Chemistry and Biochemistry Faculty Scholarship and Creative Works

Nitric oxide (NO) is a physiological mediator synthesized by NO synthases (NOS). Despite their structural similarity, endothelial NOS (eNOS) has a 6-fold lower NO synthesis activity and 6-16-fold lower cytochrome c reductase activity than neuronal NOS (nNOS), implying significantly different electron transfer capacities. We utilized purified reductase domain constructs of either enzyme (bovine eNOSr and rat nNOSr) to investigate the following three mechanisms that may control their electron transfer: (i) the set point and control of a two-state conformational equilibrium of their FMN subdomains; (ii) the flavin midpoint reduction potentials; and (iii) the kinetics of NOSr-NADP+ interactions. Although eNOSr and …

Late Homestead Period Householding At Benmore And Tintic Junction: Comparing Rural And Sub-Rural Communities In Tooele And Juab Counties, Utah, Jennifer Aurora Beard

Theses and Dissertations

Historical archaeologists are turning more and more attention to the study of capitalism in post-Industrialist nations. Rhoda Halperin's concept of householding considers networks of families or other groups that operate outside of the mainstream capitalist economy. The concept is most often applied in anthropological contexts, but may be a useful tool in the study of homesteading in the American West. At Benmore, a small homesteading community in southern Rush Valley, Tooele County, Utah, 20 families sought to survive by dry farming in a marginal environment. The enthusiasm of such residents as Israel Bennion, whose journal provides deep insight into the …

Genetic Analysis Of The Role Of Smpb In Establishing The Reading Frame On Tmrna, Talina Christensen Watts Jul 2008

Genetic Analysis Of The Role Of Smpb In Establishing The Reading Frame On Tmrna, Talina Christensen Watts

Theses and Dissertations

Ribosomes translate the genetic information encoded by mRNA into proteins. Defective mRNAs can cause stalling of translating ribosomes. The molecule tmRNA (transfer-messenger RNA) rescues stalled ribosomes in eubacteria. Together with its protein partner SmpB, tmRNA mimics a tRNA by entering the ribosomal A site and linking an alanine residue to the growing polypeptide chain. The ribosome then abandons the defective mRNA template and resumes translation on tmRNA, adding ten more amino acids to the nascent polypeptide. As a result of tmRNA action, stalled ribosomes are released and recycled, the defective mRNA is destroyed, and the aborted protein product is tagged …

Leaf Area Index Simulation In Soybean Grown Under Near-Optimal Conditions, T. D. Setiyono, A. Weiss, J. E. Specht, K. G. Cassman, A. Dobermann

Faculty Publications

Different approaches have been used to simulate leaf area index (LAI) in soybean (Glycine max L. Merr). Many of these approaches require genotype-specific calibration procedures. Studies modeling LAI dynamics under optimal growth conditions with yields close to the yield potential of soybean have remained scarce. A sink-driven approach was developed and evaluated for LAI simulation in soybean under near-optimal environments. The rate of change in expanding leaf area was simulated using the first derivative of a logistic function accounting for plant population density, air temperature, and water deficit. The rate of change in senescing leaf area was also simulated using …

The Effect Of A Pseudopalate On Voiceless Obstruent Production: A Spectral Evaluation Of Adaptation, Karie Lindsay Dean Jul 2008

The Effect Of A Pseudopalate On Voiceless Obstruent Production: A Spectral Evaluation Of Adaptation, Karie Lindsay Dean

Theses and Dissertations

Many studies in speech communication have provided valuable findings concerning the kinematic nature of speech articulation. This type of research often involves introducing an oral device to the vocal tract such as lingual pellets, magnets, and different forms of pseudopalates to track the movement and placement of the articulators. This study examined the effect of an electropalatography (EPG) pseudopalate on the production of five voiceless obstruents (/p, t, k, s/ and /sh/). Acoustic recordings from 20 adult speakers with typical speech production were made during three different speaking conditions: prior to pseudopalate placement, immediately after placement, and following 20 minutes …

Unraveling Protein Networks With Power Graph Analysis, Loïc Royer, Matthias Reimann, Bill Andreopoulos, Michael Schroeder

Faculty Publications, Computer Science

Networks play a crucial role in computational biology, yet their analysis and representation is still an open problem. Power Graph Analysis is a lossless transformation of biological networks into a compact, less redundant representation, exploiting the abundance of cliques and bicliques as elementary topological motifs. We demonstrate with five examples the advantages of Power Graph Analysis. Investigating protein-protein interaction networks, we show how the catalytic subunits of the casein kinase II complex are distinguishable from the regulatory subunits, how interaction profiles and sequence phylogeny of SH3 domains correlate, and how false positive interactions among high-throughput interactions are spotted. Additionally, we …

Mterfd3 Is A Mitochondrial Protein That Modulates Oxidative Phosphorylation, Corneliu Luca Jul 2008

Mterfd3 Is A Mitochondrial Protein That Modulates Oxidative Phosphorylation, Corneliu Luca

Open Access Dissertations

Mitochondrial function is critical for the survival of eukaryotes. Hence, mitochondrial dysfunctions are involved in numerous human diseases. An essential process for a normal mitochondrial function is mitochondrial gene expression which is tightly regulated in response to various physiological changes. The accurate control of mitochondrial gene expression is essential in order to provide the appropriate oxidative phosphorylation capacity for diverse metabolic demands. Recent findings in the basic mitochondrial replication and transcription regulation helped advance our understanding of organelle function and basic pathogenetic mechanisms of mitochondrial DNA mutations associated with oxidative phosphorylation defects. Mitochondrial transcription is regulated by the mitochondrial transcription …

Ion Permeation Through Membrane Channels: Molecular Dynamics Simulations Studies, Morad Mustafa Jul 2008

Ion Permeation Through Membrane Channels: Molecular Dynamics Simulations Studies, Morad Mustafa

Theses and Dissertations

Molecular dynamics simulation was used to study ion permeation through different membrane proteins embedded in a lipid bilayer (DMPC) with different saline solutions. The potential of mean force (PMF) for ion transport was obtained by umbrella sampling simulations. A revised MacKerell force field for tryptophan residues was studied using gramicidin A (gA) channel as a test model. The revised force field contribution to the Na+ PMF was consonant with the prediction from the experimental results, but in stark contrast to the prediction of the CHARMM force field, version 22, for the tryptophan side-chain. A new grid-based correction map algorithm by …

What's Regulation Got To Do With It? : Examining The Impact Of Regulatory Intensity On Facility Environmental Management And Performance, Cody Jones Jul 2008

What's Regulation Got To Do With It? : Examining The Impact Of Regulatory Intensity On Facility Environmental Management And Performance, Cody Jones

Dissertations and Theses

Understanding the influence of regulation on environmental management is important—studies typically find regulation to be a significant influence on environmental activity. However, studies typically assess regulation together with a suite of other influences such as investors and customers, which are often also found to be significant influences on environmental management. This research, on the other hand, suggests that regulatory constraints form a framework within which organizations operate, and therefore are associated with organizations' views of other motivating factors, as well as environmental management and performance. Additionally, this study assesses the influence of management attitudes toward environmental protection and attitudes toward …

Competitive Exclusion Of Cyanobacterial Species In The Great Salt Lake, Hillary Christine Roney Jul 2008

Competitive Exclusion Of Cyanobacterial Species In The Great Salt Lake, Hillary Christine Roney

Theses and Dissertations

Under Gause's principle two species cannot indefinitely occupy the same niche. The north and south arms of Great Salt Lake are separated by a rail causeway, resulting in salinity differences and color variation between the arms. Farmington Bay is also separated from the south arm by a vehicular causeway to Antelope Island. These causeways allow examination of competitive exclusion of cyanobacteria in the hypersaline environment of the Great Salt Lake. Cyanobacterial distributions partially map salinity, with Aphanothece halophytica proliferating in the north arm, and Nodularia spumigena in the south arm. I hypothesized that cyanobacterial species abundant north of the railway …

Do Female Hyaenas Choose Mates Based On Tenure?, Heather E. Watts Jul 2008

Do Female Hyaenas Choose Mates Based On Tenure?, Heather E. Watts

Biology Faculty Works

In their investigation into whether female mate-choice drives male dispersal, Höner et al.argue that female spotted hyaenas (Crocuta crocuta) prefer mates whose tenure in the social group is less than the females’ age, to avoid paternal incest, and suggest that male dispersal reflects this preference. However, we are not persuaded that females choose mates on the basis of tenure because Höner et al. overlook the alternative hypothesis that dispersal status itself is important in female mate-choice, such that females prefer immigrants over natal males. Like mate-choice based on tenure, choice based on dispersal status reduces the risk of incest.

Staswick, John Browse, Gregg A. Howe, Sheng Yang He

Department of Agronomy and Horticulture: Faculty Publications

Coronatine is an important virulence factor produced by several pathovars of the bacterial pathogen Pseudomonas syringae. The structure of coronatine is similar to that of a class of plant hormones called jasmonates (JAs). An important step in JA signaling is the SCFCOI1 E3 ubiquitin ligase-dependent degradation of JAZ repressor proteins. We have recently shown that jasmonoyl isoleucine (JA-Ile) promotes physical interaction between Arabidopsis JAZ1 and COI1 (the F-box component of SCFCOI1) proteins, and that the JAIle- dependent COI1–JAZ1 interaction could be reconstituted in yeast cells (i.e. in the absence of other plant proteins). Here we show …
