Real-Time Detection Of Airborne Viruses On A Mass-Sensitive Device, Joonhyung Lee, Jaesung Jang, Demir Akin, Cagri Savran, Rashid Bashir

Birck and NCN Publications

We present real-time detection of airborne Vaccinia viruses using quartz crystal microbalance (QCM) in an integrated manner. Vaccinia viruses were aerosolized and neutralized using an electrospray aerosol generator, transported into the QCM chamber, and captured by a QCM crystal. The capture of the viruses on the QCM crystal resulted in frequency shifts proportional to the number of viruses. The capture rate varied linearly with the concentration of initial virus suspensions (8.5 x 10(8)-8.5 x 10(10) particles/ml) at flow rates of 2.0 and 1.1 l/min. This work demonstrates the general potential of mass sensitive detection of nanoscale biological entities in air.

Sumoylation Regulates Lamin A Function And Is Lost In Lamin A Mutants Associated With Familial Cardiomyopathies, Yu-Qian Zhang, Kevin D. Sarge Jul 2008

Sumoylation Regulates Lamin A Function And Is Lost In Lamin A Mutants Associated With Familial Cardiomyopathies, Yu-Qian Zhang, Kevin D. Sarge

Molecular and Cellular Biochemistry Faculty Publications

Lamin A mutations cause many diseases, including cardiomyopathies and Progeria Syndrome. The covalent attachment of small ubiquitin-like modifier (SUMO) polypeptides regulates the function of many proteins. Until now, no examples of human disease-causing mutations that occur within a sumoylation consensus sequence and alter sumoylation were known. We show that lamin A is sumoylated at lysine 201 and that two lamin A mutants associated with familial dilated cardiomyopathy, E203G and E203K, exhibit decreased sumoylation. E203 occupies the conserved +2 position in the sumoylation consensus Psi KXE. Lamin A mutants E203G, E203K, and K201R all exhibit a similar aberrant subcellular localization and …

Molecular Mechanism Of Aurora-A Kinase In Human Oncogenesis, Lili He Jul 2008

Molecular Mechanism Of Aurora-A Kinase In Human Oncogenesis, Lili He

USF Tampa Graduate Theses and Dissertations

Aurora-A is a mitotic kinase, which regulates cell cycle progression through modulating centrosome function. Aurora-A expression is frequently altered in human malignancies. The discrepancy between overexpression and amplification suggests that elevated Aurora-A level is likely to be regulated also by transcriptional and/or translational modifications. In this study, we have demonstrated: 1) transcriptional regulation of Aurora-A by E2F3; 2) feedback regulation between tumor suppressor CHFR and Aurora-A; 3) CNTD2 as a novel Aurora-A partner and oncogene to activate Aurora-A and induce transformation.

Aurora-A expression and activity are cell cycle regulated. The mechanism of Aurora-A upregulation at onset of mitosis is largely …

Facilitating Use Of The Odor Footprint Tool By Nebraska Pork Producers, Rick R. Stowell Jul 2008

Facilitating Use Of The Odor Footprint Tool By Nebraska Pork Producers, Rick R. Stowell

Department of Agricultural and Biological Systems Engineering: Faculty Publications

The goal of this project was to make Odor Footprint Tool (OFT) resources readily usable by pork producers and their technical advisors as a planning and screening tool when siting swine facilities. Detailed odor footprints for five specified sizes of swine facilities were developed for twenty-four locations throughout Nebraska: six regional sites and three “localized” sites within each region. Each odor footprint shows the predicted odor annoyance-free frequency as a function of the distance from the facility. Footprints developed using localized weather data differed only slightly from footprints produced using weather data from a regional site when the general topography …

Laurate Biosensors Image Brain Neurotransmitters In Vivo: Can An Antihypertensive Medication Alter Psychostimulant Behavior?, Patricia A. Broderick, Helen Ho, Karyn Wat, Vivek Murthy

Publications and Research

Neuromolecular Imaging (NMI) with novel biosensors enables the selective detection of neurotransmitters in vivo within seconds, on line and in real time. Biosensors remain in place for continuing studies over a period of months. This biotechnological advance is based on conventional electrochemistry; the biosensors detect neurotransmitters by electron transfer. Simply stated, biosensors adsorb electrons from each neurotransmitter at specific oxidation potentials; the current derived from electron transfer is proportional to neurotransmitter concentration. Selective electron transfer properties of these biosensors permit the imaging of neurotransmitters, metabolites and precursors. The novel BRODERICK PROBE® biosensors we have developed, differ in formulation and detection …

The Trophic Ecologies Of Larval Billfishes, Tunas, And Coral Reef Fishes In The Straits Of Florida: Piscivory, Selectivity, And Niche Separation, Joel Kent Llopiz

Open Access Dissertations

The processes influencing larval fish survival in the low-latitude open ocean are poorly understood, especially with regard to feeding. As part of a large-scale study that included two years of monthly sampling in the Straits of Florida (SOF), the objectives of this dissertation were to elucidate the larval fish feeding behaviors and strategies of 1) istiophorid billfishes, 2) tunas, and 3) coral reef fishes, while also 4) characterizing the feeding environment, synthesizing the dominant trophic pathways to fish larvae, and reviewing the literature for evidence of latitudinal distinctions in larval fish trophodynamics. Larval billfishes exhibited highly selective feeding, and their …

Final Report For The Water Quality Monitoring And Analysis Of The Bennett Spring Watershed And Recharge Area, Marc R. Owen, Robert T. Pavlowsky

OEWRI Technical Reports

Poorly functioning on-site wastewater systems are perceived as a major contributor of nonpoint source pollution to Ozarks streams. Shallow soils, karst features, and lack of maintenance are often cited as reasons these systems fail in this region leading to ground and surface water contamination. Due to the importance of tourism to the local economy, many communities are concerned with protecting their water resources. Bennett Spring State Park is a major economic generator for Dallas and Laclede counties, and local community leaders are concerned about how on-site wastewater systems impact the water quality of Bennett Spring.

Timing And Cause Of Water Level Fluctuations In Kluane Lake, Yukon Territory, Over The Past 5000 Years, Janice Brahney, John J. Clague, Brian Menounos, Thomas W. D. Edwards Jul 2008

Timing And Cause Of Water Level Fluctuations In Kluane Lake, Yukon Territory, Over The Past 5000 Years, Janice Brahney, John J. Clague, Brian Menounos, Thomas W. D. Edwards

Watershed Sciences Faculty Publications

We reconstructed late Holocene fluctuations of Kluane Lake in Yukon Territory from variations in bulk physical properties and carbon and nitrogen elemental and isotopic abundances in nine sediment cores. Fluctuations of Kluane Lake in the past were controlled by changes in climate and glaciers, which affected inflow of Slims and Duke rivers, the two largest sources of water flowing into the lake. Kluane Lake fluctuated within a narrow range, at levels about 25 m below the present datum, from about 5000 to 1300 cal yr BP. Low lake levels during this interval are probably due to southerly drainage of Kluane …

Investigating Environmental Change Due To Hypoxic Conditions On The Louisiana Continental Shelf: A Geochemical Approach, Marianne E. Dietz Jul 2008

Investigating Environmental Change Due To Hypoxic Conditions On The Louisiana Continental Shelf: A Geochemical Approach, Marianne E. Dietz

USF Tampa Graduate Theses and Dissertations

The Louisiana (LA) shelf is chronically affected by seasonal hypoxia that has been shown to be spatially expanding and growing progressively more severe. Hypoxic conditions on the shelf have been closely linked to the large quantities of nutrients delivered to the Gulf of Mexico via the Mississippi River. Multiple geochemical proxies on a suite of sediment cores from the LA shelf provide a record of environmental change that parallels the onset of hypoxic conditions over the last century and prior to anthropogenic influences. The sedimentary record for the last century shows a shift from terrestrial to algal sources to the …

Enantioselective Brønsted Acid-Catalyzed Reaction Methodology Part A: Enantioselective Mannich Reaction Part B: Enantioselective Desymmetrization Of Meso-Aziridines, Emily Bretherick Rowland

USF Tampa Graduate Theses and Dissertations

The synthesis of enantiomerically pure compounds is of vital importance. Most biologically active natural products are chiral and require asymmetric synthesis, chiral resolution, or the use of naturally chiral starting materials for their preparation. Organocatalytic enantioselective reaction methodology is a continuously growing area in organic chemistry. The use of organocatalysts as a potentially environmentally friendly alternative to metal catalysts is appealing to the pharmaceutical industry.

In this dissertation an enantioselective Mannich reaction using an organocatalyst was investigated. The reaction was between a ß-keto ester and an imine electrophile catalyzed by vaulted biphenanthrol (VAPOL) phosphoric acid. The reaction resulted in products …

Enantioselective, Bronsted Acid-Catalyzed Additions Of Nitrogen And Carbon Nucleophiles To Imines, Gerald B. Rowland Jr.

USF Tampa Graduate Theses and Dissertations

The development of enantioselective reaction methodology has been at the forefront of research in both academic and industrial research laboratories due to the importance of chiral molecules in biological systems. An emerging area of research in the development of enantioselective reaction methodology has been the development of organocatalytic reactions. Organocatalysis, the use of small, chiral organic molecules as catalysts, has the advantage over traditional Lewis acid catalysis in that the reactions in general produce less toxic by-products. One recent breakthrough in the development of enantioselective methodology has been the development of chiral phosphoric acids as organocatalysts. Chiral phosphoric acids have …

School of Biological and Marine Sciences

The atmospheric partial pressure of carbon dioxide (p(CO(2))) will almost certainly be double that of pre-industrial levels by 2100 and will be considerably higher than at any time during the past few million years. The oceans are a principal sink for anthropogenic CO(2) where it is estimated to have caused a 30% increase in the concentration of H(+) in ocean surface waters since the early 1900s and may lead to a drop in seawater pH of up to 0.5 units by 2100 (refs 2, 3). Our understanding of how increased ocean acidity may affect marine ecosystems is at present very …

Immunohistochemistry And Fish On Tissue Microarrays In The Evaluation Of Inflammatory Myofibroblastic Tumor, Katherine Callahan, Kaari Reichard, Bryan Hall, John Hozier, Lisa Cerilli

Undergraduate Medical Student Research

Inflammatory myofibroblastic tumor (IMT) is a rare soft tissue tumor composed of myofibroblasts with an inflammatory infiltrate. Various gene fusions involving the anaplastic lymphoma kinase (ALK) gene at chromosome 2p23 have been described in IMT, suggesting a neoplastic etiology. The ALK rearrangement is reportedly more common in children and there are only rare IMTs in adults with genetic information. This study represents the largest series of lesions designated as IMT on morphologic grounds with comparative ALK IHC and FISH analysis. Sixteen IMTs were retrieved from the UNM and UVa archives. Inclusion criteria required spindle cells, inflammatory cells and thorough clinical, …

A Novel Approach To The Synthesis Of Silicon Nanowires, Joo Chick Chan Jul 2008

A Novel Approach To The Synthesis Of Silicon Nanowires, Joo Chick Chan

Dissertations and Theses

Contemporary VLS-SiNW synthetic methods employ CVD and laser ablation methods that produce single crystalline SiNW; which can be doped by co-flowing appropriate dopants, such as diborane (p-type) and phosphine (n-type). Not only are these gases toxic (non-green), but also the synthetic methods used are costly requiring elaborate instrumental assembly. This thesis presents a low-cost and "green" method in synthesizing and doping of SiNW in a glass tube. It was found that by selecting the appropriate metal catalyst, precursor reactants, and the reaction conditions both SiNW and carbon nanotube (CNT) can be synthesized on metallic (aluminum and copper), semiconducting (silicon) and …
