First Report Of Soybean Rust Caused By Phakopsora Pachyrhizi In Nebraska, S. R. Watson, L. J. Giesler, A. D. Ziems, T. E. Brovont

Soybean rust caused by Phakopsora pachyrhizi Sydow was first observed in the continental United States in Louisiana in November 2004 (2). As part of the national soybean rust monitoring effort, samples were collected on 3 October 2007 during the scouting of fields with green leaves in southeastern Nebraska. After incubation at room temperature for 24 h, uredinea and urediniospores were observed with microscopic examination. Urediniospores were obovoid, hyaline to pale brown, and measured 20 to 30 × 18 to 20 μm.

MicroRNA (miRNA) gene expression profiling has provided important insights into plant and animal biology. However, there has not been ample published work about pitfalls associated with technical parameters in miRNA gene expression profiling. One source of pertinent information about technical variables in gene expression profiling is the separate and more well-established literature regarding mRNA expression profiling. However, many aspects of miRNA biochemistry are unique. For example, the cellular processing and compartmentation of miRNAs, the differential stability of specific miRNAs, and aspects of global miRNA expression regulation require specific consideration. The nitric oxide (NO) cytotoxicity has been well documented in bacteria and mammalian cells. However, the underlying mechanism is still not fully understood. Here we report that transient NO exposure effectively inhibits cell growth of Escherichia coli in minimal medium under anaerobic growth conditions and that cell growth is restored when the NO-exposed cells are either supplemented with the branched-chain amino acids (BCAA) anaerobically or returned to aerobic growth conditions. The enzyme activity measurements show that dihydroxyacid dehydratase (IlvD), an iron-sulphur enzyme essential for the BCAA biosynthesis, is completely inactivated in cells by NO with the concomitant formation of the …

We used the chimeric Arabidopsis cyclic nucleotide-gated ion channel AtCNGC11/12 to conduct a structure-function study of plant cyclic nucleotide-gated ion channels (CNGCs). AtCNGC11/12 induces multiple pathogen resistance responses in the Arabidopsis mutant constitutive expresser of PR genes 22 (cpr22). A genetic screen for mutants that suppress cpr22-conferred phenotypes identified an intragenic mutant, #73, which has a glutamate to lysine substitution (E519K) at the beginning of the eighth β-sheet of the cyclic nucleotide-binding domain in AtCNGC11/12. The #73 mutant is morphologically identical to wild-type plants and has lost cpr22-related phenotypes including spontaneous cell death and enhanced pathogen resistance. Leaf hairs (trichomes) of Arabidopsis (Arabidopsis thaliana) have been extensively used as a model to address general questions in cell and developmental biology. Here, we lay the foundation for a systems-level understanding of the biology of this model cell type by performing genome-wide gene expression analyses. We have identified 3,231 genes that are up-regulated in mature trichomes relative to leaves without trichomes, and we compared wild-type trichomes with two mutants, glabra3 and triptychon, that affect trichome morphology and physiology in contrasting ways. We found that cell wall-related transcripts were particularly overrepresented in trichomes, consistent with their highly elaborated structure. In …

This paper is a companion piece to my short film, “Victorian Organ”. This film is an effort to contextualize the mechanical, or man-made, within nature. Treacher Collins syndrome is an autosomal-dominant mandibulofacial dysostosis caused by haploinsufficiency of the TCOF1 gene product treacle. Mouse Tcof1 protein is approximately 61% identical and 71% similar to treacle, and heterozygous knockout of Tcof1 causes craniofacial malformation. Tcof1 expression is high in developing neural crest, but much lower in other tissues. To investigate this dual regulation, highly conserved regions upstream of TCOF1 homologs were tested through deletion and mutation reporter assays, and conserved predicted transcription factor binding sites were assessed through chromatin binding studies. We examined memory functioning in children with reading disabilities (RD), Attention deficit/hyperactivity disorder (ADHD), and RD/ADHD using a clinic sample with a clinical instrument: the Children's Memory Scale, enhancing its generalizability. Participants included 23 children with RD, 30 with ADHD, 30 with RD/ADHD, and 30 controls. Children with RD presented with reduced verbal short-term memory (STM) but intact visual STM, central executive (CE), and long-term memory (LTM) functioning. Their deficit in STM appeared specific to tasks requiring phonetic coding of material. Children with ADHD displayed intact CE and LTM functioning but reduced visual-spatial STM, especially when off stimulant medication. Children …

The neurohypophysial hormone oxytocin (OT), synthesized in magnocellular paraventricular (PVN) and supraoptic (SON) nuclei, is well known for its effects in lactation. Our previous studies showed that central OT receptor (OTR) binding is increased during gestation and that blockade of central OTRs, specifically during mid-late gestation, causes a delay in OT release during suckling and reduces weight gain in pups, suggesting decreased milk delivery. In the present study, we tested whether central OTR blockade during late gestation disrupts the gestation-related plasticity in intrinsic membrane properties. N-Substituted porphyrins are well-known for the distortion they exhibit of the porphyrin plane through the sp3 hybridization of one of the pyrrolenic units. They have served as model compounds in investigations of many biochemical processes. In this paper, we developed an efficient route to N-substituted porphyrins, and report the synthesis of a series of new N-substituted cobaltacarborane - porphyrins containing one or two cobaltabisdicarbollide anions linked by (CH2CH2O) 2 chains to either the core porphyrin nitrogens or to a meso-aminophenyl group. Amino acids are important to all animal life. Termites rely mainly on microorganisms to retrieve these nitrogen sources from their food. One naturally occurring amino acid, m-tyrosine, has been reported to be toxic to bacteria and weeds. Its direct effects on termite survival are unknown however. Both no-choice and choice bioassays using five different doses of m-tyrosine were conducted to determine the feeding response and survival of Formosan subterranean termites, Coptotermes formosanus Shiraki (Isoptera: Rhinotermitidae), in the laboratory. PURPOSE OF REVIEW: To review the scientific literature supporting the participation of caveolin-1 in the pathogenesis of tissue fibrosis and the notion that modulation of the caveolin-1 pathway may represent a novel treatment for systemic sclerosis and other fibrotic diseases.
RECENT FINDINGS: Caveolin-1 plays an important role in the regulation of transforming growth factor-beta (TGF-beta) signaling owing to its participation in TGF-beta receptor internalization. TGF-beta receptor internalized through caveolin-1 lipid rafts undergoes rapid degradation, effectively decreasing TGF-beta signaling. Studies have shown that caveolin-1 knockdown in vitro markedly increased collagen gene expression in normal human lung fibroblasts. Caveolin-1 was reduced in …

Melatonin is produced in the human pineal gland, particularly at night, with the circadian rhythm of blood melatonin levels closely paralleling its production within the pineal gland. Light exposure at night, or rapid transmeridian travel severely compromises the circadian production of melatonin. The disturbed melatonin rhythm contributes to jet lag and sleep inefficiency, both of which are improved by melatonin administration. Melatonin is also a highly effective direct free radical scavenger and antioxidant. In this capacity, melatonin reduces experimental cataractogenesis, traumatic injury to the spinal cord and brain, and protects against oxidative damage to neurons and glia in models of …

In the present study antimicrobial efficiency of Citrullus colocynthis (L.) Schrad was studied on seven bacteria Bacillus subtilis, Escherichia coli, Klebsiella pneumoniae, Proteus vulgaris, Pseudomonas aeruginosa, Salmonella typhi and Staphylococcus aureus. Ethanol, chloroform and petroleum ether extracts were used for antibacterial assay. Growth inhibitions were evaluated by the disc diffusion method. Ethanol extract is more active against Escherichia coli, Proteus vulgaris and Staphylococcus aureus. Pseudomonas aeruginosa and Bacillus subtilis show moderate activity in chloroform extract. Petroleum ether extract is more active against Escherichia coli. All extracts exhibited considerably less activity against Klebsiella pneumoniae and Salmonella typhi.

Catharanthus roseus popularly known as Madagascar periwinkle is a potential source for anti-leukemic alkaloids. The present study aims to evaluate the possibility for the presence of novel bio-active compounds against pathogenic bacteria, as most of the pathogens develop drug resistance against commonly used antibiotics. To determine antibacterial activity, crude extracts from different parts of C. roseus were tested against bacterial strains of clinical significance. Extraction of bio-active principles in appropriate solvent was followed by evaluation of antibacterial activity by disc diffusion assay against selected bacterial stains. The present paper reports 17 plant species belonging to 13 families, mostly used for various disease and disorders by the local people and herbal healer of Chandaka areas of Bhubaneswar. The local population of the region primarily depends upon these plants for curing of various aliments and their disorders. They are enumerated with binomial, family, vernacular name and floristic study and medicinal uses of the plants and their parts . Further studies on chemical and pharmacological actions are suggested to validate the claims.

The European corn borer, Ostrinia nubilalis (Hübner) (Lepidoptera: Crambidae), is a widely distributed and serious economic pest to corn production in the U.S. Genetic variability of O. nubilalis was studied in 18 sub-populations in the upper Midwestern United States using amplified fragment length polymorphism. The relatively low GST values indicate that more variation exists within populations than between populations. Glucagon-like peptide-1 (GLP-1) and peptide YY (PYY) are anti-diabetes/obesity hormones ecreted from the gut after meal ingestion. We have shown that dietary-resistant starch (RS) increased GLP-1 and PYY secretion, but he mechanism remains unknown. RS is a fermentable fiber that lowers the glycemic index of the diet and liberates short-chain fatty acids (SCFAs) through fermentation in the gut. This study investigates the two possible mechanisms by which RS stimulates GLP-1 and PYY secretion: the effect of a meal or glycemic index, and the effect of fermentation. The research focused on the impact pandemic influenza could have on K-12 school systems. The research looked at responses of school leaders regarding the pedagogical, community health, social, and economic impacts of a pandemic. During an influenza pandemic school districts may be asked to close for as long as 12 weeks to reduce contact among children and stop the spread of disease. School superintendents' perceptions and attitudes were sought regarding school policy implications associated with long-term closure of K-12 school districts. Female Adolescents’ Attitude Towards Sexually Risky Behaviors, Janet F. Wang, Patricia S. Simoni, Ying Wu, Christine Banvard

This cross-sectional survey examines the association between attitudes and female adolescents’ sexually risky behaviors. A sample of 159 subjects, aged 14 – 20 from three high schools in West Virginia participated in the study. The results indicate that 60.3% (n = 96) reported having had sexual intercourse and, of this sexually active group, 60.4% (n = 58) stated that they had sex the first time at age 15 or younger. Fifty-five percent (n = 88) reported at least one of the following behaviors: (1) sexual initiation at age 15 or younger, (2) frequent sexual intercourse last month, (3) multiple sex …

This study examined the role of gender, self-efficacy, age and extroversion on smoking behavior among Ambrose Alli University Students. Two hundred and fifty (250) undergraduates participated in the study (males 160 and females 90). Questionnaire was used in collecting data that were analyzed in this study. The questionnaire consisted of 4 sections: the demographic variables, extroversion scale, self-efficacy scale and attitude toward smoking scale. Two out of the four hypotheses tested were supported. The two parts of William Faulkner’s If I Forget Thee, Jerusalem—“The Wild Palms” and “Old Man”—mirror one another; the second story reflects a more familiar way of illustrating the same theme as the first. Each story emphasizes the other, building the shared theme subversive of traditional gender roles. It is also my feeling that Faulkner inverts Arthurian motifs to tell both stories (especially “The Wild Palms,” though most assume “Old Man” to be the more romantically accessible tale) exactly as Nathaniel Hawthorne did to tell The Scarlet Letter.
