Editing of tRNAs is widespread in nature and either changes the decoding properties or restores the folding of a tRNA. Unlike the phylogenetically disperse adenosine (A) to inosine (I) editing, cytosine (C) to uridine (U) editing has only been previously described in organellar tRNAs. We have shown that cytoplasmic tRNAThr(AGU) undergoes two distinct editing events in the anticodon loop: C to U and A to I. In vivo, every inosine-containing tRNAThr is also C to U edited at position 32. In vitro, C to U editing stimulates conversion of A to I at the wobble base. Although …

Stability And Persistence Of Aldrin And Methyl-Parathion Resistance In Western Corn Rootworm Populations (Coleoptera: Chrysomelidae), Srinivas Parimi, Lance J. Meinke, B. Wade French, Laurence D. Chandler, Blair D. Siegfried

The susceptibilities of laboratory and field-collected western corn rootworm populations (Diabrotica virgifera virgifera LeConte) to methyl-parathion and aldrin were estimated by topical application of insecticide during 2002 to determine the stability of resistance in the absence of selective pressures. Most of the laboratory-reared and field-collected populations were significantly resistant to both insecticides. Average LD 50 values of laboratory and field-collected populations were 19- and 13-fold greater than the susceptible population in methyl-parathion bioassays, respectively, and 204- and 125-fold greater in the aldrin bioassays, respectively. The presence of aldrin and methyl-parathion resistance in field-collected populations strongly suggests that both resistance …

Understanding physiological and behavioral mechanisms underlying the diversity of observed life‐history strategies is challenging because of difficulties in obtaining long‐term measures of fitness and in relating fitness to these mechanisms. We evaluated effects of experimentally elevated testosterone on male fitness in a population of dark‐eyed juncos studied over nine breeding seasons using a demographic modeling approach. Elevated levels of testosterone decreased survival rates but increased success of producing extra‐pair offspring. Higher overall fitness for testosterone‐treated males was unexpected and led us to consider indirect effects of testosterone on offspring and females. Runoff from open lot livestock systems (beef and dairy) defined as Concentrated Animal Feeding Operations (CAFO) must be controlled by systems designed and managed to prevent the release of manure-contaminated runoff for storms equal to or less than a 25-yr, 24-h design storm. This performance standard has been attained for open lot systems with some combination of clean water diversion, settling basins, runoff collection ponds, and irrigation systems (baseline system).
An alternative approach is to rely on overland flow and infiltration into cropland with perennial forage or grasses for treatment of open lot runoff. Such vegetative systems have been researched …

Quantification of the contributions of river-influenced margins to regional CO2 fluxes is difficult due to the high degree of spatial and temporal variability in these regions. We describe an algorithm for assessment of surface water partial pressure of CO2 (pCO(2)) from MODIS imagery in the northern Gulf of Mexico. Principal component analysis and multiple regression were used to relate surface pCO(2) to environmental variables (T, S, chlorophyll). Subsequent retrieval of corresponding products from MODIS-Aqua L1B data permitted the assessment of regional distributions of pCO(2). ABSTRACT OBJECTIVE: To model the effect of universal cesarean delivery on professional liability costs.
STUDY DESIGN: We examined all obstetric professional liability claims covered by a single insurer between January 1, 1990 and December 31, 2000. We reviewed each case to determine if an alternate route of delivery might reasonably have prevented the lawsuit. Costs were calculated by adding the cost of processing the claim, the legal defense, the settlement payments and/or the actuarially derived adjustments. Using a 20% cesarean rate as our baseline, we modeled the effect on liability costs of cesarean delivery in all patients.

The endocytic pathway of eukaryotes is essential for the internalization and trafficking of macromolecules, fluid, membranes, and membrane proteins. One of the most enigmatic aspects of this process is endocytic recycling, the return of macromolecules (often receptors) and fluid from endosomes to the plasma membrane. We have previously shown that the EH-domain protein RME-1 is a critical regulator of endocytic recycling in worms and mammals. Here we identify the RAB-10 protein as a key regulator of endocytic recycling upstream of RME-1 in polarized epithelial cells of the Caenorhabditis elegans intestine. rab-10 null mutant intestinal cells accumulate abnormally abundant RAB-5-positive early …

Integrins comprise a large family of cell adhesion receptors that mediate diverse biological events through cell-cell and cell-extracellular matrix interactions. Recent studies have shown that several integrins are localized to synapses with suggested roles in synaptic plasticity and memory formation. We generated a postnatal forebrain and excitatory neuron-specific knock-out of beta1-integrin in the mouse. Electrophysiological studies demonstrated that these mutants have impaired synaptic transmission through AMPA receptors and diminished NMDA receptor-dependent long-term potentiation. Despite the impairment in hippocampal synaptic transmission, the mutants displayed normal hippocampal-dependent spatial and contextual memory but were impaired in a hippocampal-dependent, nonmatching-to-place working memory task. Alu elements are the most active and predominant type of short interspersed elements (SINEs) in the human genome. Recently inserted polymorphic (for presence/absence) Alu elements contribute to genome diversity among different human populations, and they are useful genetic markers for population genetic studies. The objective of this study is to identify polymorphic Alu insertions through an in silico comparative genomics approach and to analyze their distribution pattern throughout the human genome. By computationally comparing the public and Celera sequence assemblies of the human genome, we identified a total of 800 polymorphic Alu elements. We used polymerase chain reaction-based assays to …

After years of inconclusive debate, recent studies purport to demonstrate that capital punishment does indeed deter murder, perhaps to the tune of multiple saved lives for each person executed. In response to these studies, Professors Sunstein and Vermeule have argued that since capital punishment leads to a net savings of innocent lives, it may be morally required on consequentialist grounds. I argue, even assuming the validity of the studies, that capital punishment cannot be justified in the United States in the current historical context for reasons of justice that trump consequentialist considerations. The ability of engineered cerium oxide nanoparticles to confer radioprotection was examined. Rat astrocytes were treated with cerium oxide nanoparticles to a final concentration of 10 nanomolar, irradiated with a single 10 Gy dose of ionizing radiation and cell death was evaluated by propidium iodine uptake at 24 and 48 hours after radiation insult. Treatment of rat astrocytes with nanoceria resulted in an approximate 3-fold decrease in radiation induced death. These results suggest that the nanoceria are conferring protection from radiation induced cell death. Further experiments with human cells were conducted. "Bloodlines" is a collection of personal essays that focus on the process of remembering, imagining, and reflecting on the past through the lens of a perpetually shifting present. They consider situations ranging from mental and physical illnesses, from cancer to alcohol addiction, to career changes, to the often dysfunctional and displaced family ties that distance and adulthood have not severed. In "Searching," I write the narrative of the ongoing search for my birthmother, and how the search complicates the relationship with my adoptive mother, who always feared she’d lose me. Effective citizenship practice in the United States has several important characteristics, which can provide the foundation for young people to access opportunity in employment and education. A continuum of policies, programs, and strategies meant to alleviate poverty have central themes of providing education and vocational opportunities, and yet large numbers of young minority people remain disenfranchised with no chance to succeed. The chloroplast has been demonstrated to be an ideal compartment to accumulate certain proteins or their biosynthetic products that would be harmful if they were accumulated in the cytoplasm. Hyper-expression of foreign proteins in chloroplast transgenics has accumulated up to 46% total soluble protein, this is possible due to the ~100 chloroplast genomes per chloroplast and ~100 chloroplasts per cell which can therefore, contain up to 10,000 copies of the transgene. Maternal gene inheritance of plastids in most crop plants results in natural gene containment. Chloroplast transformation also eliminates positional effects that are frequently observed with nuclear transformation and no …
