Variation In The Innate And Acquired Arms Of The Immune System Among Five Shorebird Species, Luisa Mendes, Theunis Piersma, Dennis Hasselquist, Kevin Matson, Robert Ricklefs

To contribute to an understanding of the evolutionary processes that shape variation in immune responses, we compared several components of the innate and acquired arms of the immune system in five related, but ecologically diverse, migratory shorebirds (ruff Philomachus pugnax L., ruddy turnstone Arenaria interpres L., bar-tailed godwit Limosa lapponica L., sanderling Calidris alba Pallas and red knotC. canutus L.). We used a hemolysis-hemagglutination assay in free-living shorebirds to assess two of the innate components (natural antibodies and complement-mediated lysis), and a modified quantitative enzyme-linked immunosorbent assay in birds held in captivity to assess the acquired component (humoral antibodies against …

Competition Between The Dna Unwinding And Strand Pairing Activities Of The Werner And Bloom Syndrome Proteins, Amrita Machwe, Enerlyn M. Lozada, Liren Xiao, David K. Orren

BACKGROUND: The premature aging and cancer-prone Werner and Bloom syndromes are caused by defects in the RecQ helicase enzymes WRN and BLM, respectively. Recently, both WRN and BLM (as well as several other RecQ members) have been shown to possess a strand annealing activity in addition to the requisite DNA unwinding activity. Since an annealing function would appear to directly oppose the action of a helicase, we have examined in this study the dynamic equilibrium between unwinding and annealing mediated by either WRN or BLM.

Native disulfide bond formation in eukaryotes is dependent on protein-disulfide isomerase (PDI) and its homologs, which contain varying combinations of catalytically active and inactive thioredoxin domains. However, the specific contribution of PDI to the formation of new disulfides versus reduction/rearrangement of non-native disulfides is poorly understood. We analyzed the role of individual PDI domains in disulfide bond formation in a reaction driven by their natural oxidant, Ero1p. We found that Ero1p oxidizes the isolated PDI catalytic thioredoxin domains, A and A′ at the same rate. In contrast, we found that in the context of full-length PDI, there is an asymmetry …

Anthropogenically induced global climate change has profound implications for marine ecosystems and the economic and social systems that depend upon them. The relationship between temperature and individual performance is reasonably well understood, and much climate-related research has focused on potential shifts in distribution and abundance driven directly by temperature. However, recent work has revealed that both abiotic changes and biological responses in the ocean will be substantially more complex. For example, changes in ocean chemistry may be more important than changes in temperature for the performance and survival of many organisms. Total protein secreted in the intrauterine lumen increases between day 10 and 13 post-estrus in both cyclic and pregnant gilts. The objective of this experiment was to identify those intrauterine proteins whose secretion changes during this time period. Sixteen mature gilts were either mated (day 0) or remained cyclic and were slaughtered at either day 10 or day 13 (n = 4 per status by day). At slaughter, each uterine horn was flushed with 20 ml Minimal Essential Medium. Flushings were dialyzed extensively against distilled water. Prp19 is an essential splicing factor and a member of the U-box family of E3 ubiquitin ligases. Prp19 forms a tetramer via a central coiled-coil domain. Here, we show the U-box domain of Prp19 exists as a dimer within the context of the Prp19 tetramer. A high-resolution structure of the homodimeric state of the Prp19 U-box was determined by X-ray crystallography. Mutation of the U-box dimer interface abrogates U-box dimer formation and is lethal in vivo. Control Of High-Order Harmonic Emission Using Attosecond Pulse Trains, J. Biegert, A. Heinrich, C. P. Hauri, W. Kornelis, P. Schlup, M. P. Anscombe, M. B. Gaarde, K. J. Schafer, U. Keller

We show that attosecond pulse trains are a natural tool to control strong field processes such as high-order harmonic generation. Coherently combining an attosecond pulse train with an IR driving field, we predict and experimentally confirm enhancement and spectral narrowing of the harmonic yield at photon energies around 90 eV. The use of an attosecond pulse train to seed the harmonic generation process replaces tunneling ionization with a single-photon ionization step, therefore permitting the manipulation of the time–frequency properties of high-order harmonic generation already at the single-atom level. © 2006 Taylor & Francis Group, LLC.

A method and apparatus for characterizing the type of a blood sample and a variety of blood components are provided wherein a transmission spectrum of the sample is collected over a predetermined wavelength range. For blood typing, this spectrum is then compared with a set of control spectra collected from control blood samples having known blood types, from which the type of the blood sample can be determined. Further methods and apparatus are provided for determining the viability of and for cross matching a platelet unit.
