Bacterial primases are essential for DNA replication due to their role in polymerizing the formation of short RNA primers repeatedly on the lagging-strand template and at least once on the leading-strand template. The ability of recombinant Staphylococcus aureus DnaG primase to utilize different single-stranded DNA templates was tested using oligonucleotides of the sequence 5’-CAGA (CA) 5 XYZ (CA) 3-3’, where XYZ represented the variable trinucleotide. These experiments demonstrated that S. aureus primase synthesized RNA primers predominately on templates containing 5’-d(CTA)-3’ or TTA and to a much lesser degree on GTA-containing templates, in contrast to results seen with the Escherichia …

Recent studies suggest that a linear correlation exists between the age of individual birds and accumulation of the advanced glycation endproduct pentosidine (Ps). We compared Ps accumulation in skin tissue of Ruffed Grouse (Bonasa umbellus) and Double-crested Cormorants (Phalacrocorax auritus). Skin samples were taken from the breasts of 52 Ruffed Grouse of known ages ranging from a few days to 10 years. Skin was cleaned of feathers and adipose tissue, acid hydrolyzed, and measured for collagen content via hydroxyproline spectrophotometric assay. Host-parasite associations are assumed to be ecologically specialized, tightly coevolved systems driven by mutual modification in which host switching is a rare phenomenon. Ecological fitting, however, increases the probability of host switching, creating incongruences between host and parasite phylogenies, when (1) specialization on a particular host resource is a shared characteristic of distantly related parasites, and (2) the resource being tracked by the parasite is widespread among many host species. As communities and a country, we have a monumental task to solve the energy and global climate change problems, while maintaining our capacity to produce food, feed, and fiber for an ever increasing world population. The severity of these problems is exacerbated by the universal desire for an increased standard of living, which invariably translates to more energy use, greater demand for products, and higher quality diets (usually in the form of more fresh fruits and vegetables and more animal protein). In 2005 the Town of Newfields, NH was awarded a grant by the New Hampshire Estuaries Project. The Eligible Activity applied for, under the category of Land Conservation and Natural Resource Protection was “Development of monitoring plans for town-held easements on conservation lands”. The Town identified four properties on which the they hold conservation easements as the focus of the project. Newfields’ goal in applying for the grant were to have each property visited by the contractor and a local volunteer, conduct an interview with the landowner and a conduct a ground monitoring of the property. Over two-thirds of the world’s harvested fish stocks are considered to be either reduced or threatened because of overexploitation, which suggest that one of the central challenges facing coastal managers is the recovery and sustainable harvesting of these species. One promising fishery management strategy is the use of marine reserves, or conservation areas where fishing is prohibited, to rebuild depleted populations. In the Gulf of Maine (GoM), several closed areas have been established to restrict fishing activities such as gillnetting, scallop dredging, and mid-water and bottom trawling. These closures in the GoM not only protect diminished fish stocks, but also …

A tool kit of vectors was designed to manipulate and express genes from a wide range of gram-negative species by using in vivo recombination. Saccharomyces cerevisiae can use its native recombination proteins to combine several amplicons in a single transformation step with high efficiency. We show that this technology is particularly useful for vector design. Shuttle, suicide, and expression vectors useful in a diverse group of bacteria are described and utilized. This report describes the use of these vectors to mutate clpX and clpP of the opportunistic pathogen Pseudomonas aeruginosa and to explore their roles in biofilm formation and surface …

Galectin-3, a factor involved in the splicing of pre-mRNA, shuttles between the nucleus and the cytoplasm. We have engineered a vector that expresses the fusion protein containing the following: (a) green fluorescent protein as a reporter of localization, (b) bacterial maltose-binding protein to increase the size of the reporter polypeptide, and (c) galectin-3, whose sequence we wished to dissect in search of amino acid residues vital for nuclear localization. In mouse 3T3 fibroblasts transfected with this expression construct, the full-length galectin-3 (residues 1-263) fusion protein was localized predominantly in the nucleus. Galectin-3, a factor involved in the splicing of pre-mRNA, shuttles between the nucleus and the cytoplasm. Previous studies have shown that incubation of fibroblasts with leptomycin B resulted in the accumulation of galectin-3 in the nucleus, suggesting that the export of galectin-3 from the nucleus may be mediated by the CRM1 receptor. A candidate nuclear export signal fitting the consensus sequence recognized by CRM1 can be found between residues 240 and 255 of the murine galectin-3 sequence. This sequence was engineered into the pRev(1.4) reporter system, in which candidate sequences can be tested for nuclear export activity in terms of …

As more emphasis is placed on offering education to the distance student and monies are spent to provide these services, institutions must ensure they reap the rewards of the investment. One avenue to ensure success in distance education is the implementation of strong student support services. This is a task that will take the teamwork of educators, administrators, instructional technologists/designers, and support personnel. In September 1989, the United States’ Governors Conference in Charlottesville, Virginia set an ambitious goal by declaring that “By the year 2000, United States students will be first in the world in mathematics and science achievements”. The Lyme disease agent Borrelia burgdorferi can persistently infect humans and other animals despite host active immune responses. This is facilitated, in part, by the vls locus, a complex system consisting of the vlsE expression site and an adjacent set of 11 to 15 silent vls cassettes. Segments of nonexpressed cassettes recombine with the vlsE region during infection of mammalian hosts, resulting in combinatorial antigenic variation of the VlsE outer surface protein. We now demonstrate that synthesis of VlsE is regulated during the natural mammal-tick infectious cycle, being activated in mammals but repressed during tick colonization. Examination of cultured B. …

1. Lake sturgeon (Acipenser fulvescens) are threatened or endangered throughout much of their range. Juvenile sturgeon utilize sandy and silty habitats extensively during their growth. Invasive zebra mussels change the nature of sandy and silty habitats because they settle on and coat the habitat with the shells of living and dead individuals. The potential impacts of this increased habitat complexity on lake sturgeon is unknown. 2. Juvenile lake sturgeon habitat choice was assessed in laboratory experiments, and zebra mussel impact on the foraging success of juvenile lake sturgeon on three different prey species was measured. 3. The host plant expansion of a diamondback moth, Plutella xylostella (L.) (DBM) strain to snowpea (Pisum sativum L.) raised the question whether a specialist parasitoid Diadegma semiclausum (DS) could be conditioned to locate and parasitize its host on the new host plant. In a specialist parasitoid a behavioural change towards a plant outside the normal host plant range of its host due to developmental experience is not expected. The responsive behaviour, parasitism rates and fitness of three subsequent DS generations were investigated on the snowpea-strain of DBM. The goal of this study was to identify the factors that limit the survival of the red-eared slider turtle Trachemys scripta during long-term anoxic submergence at 3°C. We measured blood acid-base status and tissue lactate and glycogen contents after 13, 29, and 44 d of submergence from ventricle, liver, carapace (lactate only), and four skeletal muscles. We also measured plasma Ca2+, Mg2+, Na+, K+, Cl-, inorganic phosphate (Pi), lactate, and glucose. After 44 d, one of the six remaining turtles died, while the other turtles were in poor …
