Agswe1p Regulates Mitosis In Response To Morphogenesis And Nutrients In Multinucleated Ashbya Gossypii Cells, Hanspeter Helfer, Amy S. Gladfelter

Nuclei in the filamentous, multinucleated fungus Ashbya gossypii divide asynchronously. We have investigated what internal and external signals spatially direct mitosis within these hyphal cells. Mitoses are most common near cortical septin rings found at growing tips and branchpoints. In septin mutants, mitoses are no longer concentrated at branchpoints, suggesting that the septin rings function to locally promote mitosis near new branches. Similarly, cells lacking AgSwe1p kinase (a Wee1 homologue), AgHsl1p (a Nim1-related kinase), and AgMih1p phosphatase (the Cdc25 homologue that likely counteracts AgSwe1p activity) also have mitoses distributed randomly in the hyphae as opposed to at branchpoints. The Newgate Calendar, one of the earliest true crime documents, has rarely been studied. This paper aims to demonstrate that these texts illuminate various aspects of the culture that both produced and consumed the narratives the Calendar contains. In particular, I have focused on two different categories of crimes against the body. The Calendar's early volumes, which were published while state-sponsored punishment was still a public spectacle, offer several accounts that deal with the confinement and torture of living victims. Later volumes do not include torture narratives. Instead, anxiety over the treatment of dead bodies caused an increased interest in …

The current study compared the impact of different paradigms of estradiol replacement on working memory performance. In adult ovariectomized rats, a vehicle-treated control group (n=10) was compared to three estradiol replacement paradigms: 1) continuous delivery via Silastic capsules (n=8); 2) cyclic replacement via two 10 ìg injections on two out of every four days (n=10); 3) cyclic replacement via one 2 ìg injection every four days (n=10). While treatment continued, animals were tested over 24 days in the 8-arm radial maze. After this acquisition period, various delay times were introduced between 4th and 5th arm choices. Treatments had no effects …

Luminescenct Quantum Dots (QDs) intrigue the interest of researchers with their properties of high quantum yield, broad absorption and narrow emission spectra. Since prepared in organic solvents, QDs are not suitable for biological systems. In my study, several methods of "cap exchange" were tested to enable forming water-soluble QDs with minimal aggregation .The QD-mercaptoacetic acid is simple to synthesize and has good water-solubility, but the QDs are not stable. The direct attachment of peptide molecules to the QDs can passivate the surface and enable water solubility of QDs. It is only applicable for phytochelatin-related peptides, not for all kind of …

Single biopolymers (DNA, RNA, or polypeptide) can be examined using an alpha-hemolysin channel detector. When a biopolymer is present in an alpha-hemolysin channel it can produce a highly structured ionic current blockade pattern, where the lifetimes at various sub-blockade levels reveal information about the kinetics of the biopolymer. Here we describe integration of LabVIEW/LabWindows automation capabilities with the "in-house" Channel Current Cheminformatics (CCC) software. Data acquired with LabVIEW/LabWindows is passed to the CCC software, on a streaming real time basis, for analysis and classification. The classification results are then quickly returned to the LabVIEW/LabWindows automation software for experimental feedback control. …

Artificial reef development is a common fishery enhancement tool used in aquatic systems worldwide. In 2003 and 2004, a series of artificial reefs were constructed in a large oligohaline estuary, Lake Pontchartrain, Louisiana to improve recreational fisheries. The reefs are the first in Louisiana to be built of reef balls, concrete domes deployed primarily in marine environments. Although they attract fish and invertebrates in high salinities, reef balls have not been tested in low-salinity estuarine systems. The purpose of this phenomenological study was to learn about African American parents' perceptions of play, counseling, and play therapy. Research interviews conducted with eight African American parents with elementary school aged children offered insight regarding parents' thoughts and experiences. This study explored the question, "What are African American parents'perceptions of play therapy?" In particular, this study explored questions that pertained to African American parents' (a) thoughts about counseling, (b) beliefs about the purpose of play, and (c) perceptions about play as a therapeutic agent in counseling. Emotion regulation has been recognized as a fundamental process in early socioemotional development; however, investigation into the relations between parenting styles, practices, and emotion regulation in maltreating families has been severely lacking in the literature. The current study observationally examined the relations between specific parenting practices, parenting styles, and maltreated children's emotion regulation examining the same child with both a maltreating and non-maltreating caregiver. The findings of this study indicate that parenting practices within both maltreating and non-maltreating caregivers affect child emotion regulation and emotionality. Positive, supportive parenting increases child effortful control and positive affect while decreasing anger. Alternately, negative, …

Single Metallic Nanowire Arrays (Ni, Au and Cu etc) and superlattice nanowire arrays (Ni/Au, Ni/Cu) were synthesized through template-directed electrochemical deposition method. The dual-bath technique was employed in fabrication of superlattice nanowire arrays. Anodized alumina membrane (AAM) and Ion Track-etched Polycarbonate Membrane was used as the fabrication templates. The specific electrode making technique and electrochemical deposition procedures were described in detail. Scanning electron microscope (SEM) performed all sample characterization. Vibrating sample magnetometer (VSM) and ferromagnetic resonance (FMR) were utilized to investigate the magnetic properties of single Ni nanowire arrays of different lengths and Ni/Cu superlattice nanowire arrays. The folding of proteins was investigated by using helical polypeptides attached to gold or magnetite nanoparticle surfaces. Depending on type and loading of the polypeptide on the surface of gold nanoparticles, pH, temperature, presence of different ions in the solution, and influence of different mechanical factors, conformational changes in the polypeptide occurred. The aggregation of the gold nanoparticles related to the folding of the polypeptide caused shifts in color of the solutions from red to blue that were measured by UV-Vis spectrometry. The hepatitis B virus (HBV) X protein (pX) is implicated in hepatocarcinogenesis by an unknown mechanism. pX variants encoded by HBV genomes found integrated in genomic DNA from liver tumors of patients with hepatocellular carcinoma (HCC) generally lack amino acids 134 to 154. Since deregulation of mitogenic pathways is linked to oncogenic transformation, herein we define the pX region required for mitogenic pathway activation. A series of pX deletions was used to construct tetracycline-regulated pX-expressing cell lines. Ferritins are ubiquitous iron storage proteins in living systems. Although much is known about the iron deposition process in ferritin and a mechanism has been developed, several important issues still remain unknown. One lingering question is the less than stoichiometric quantities of hydrogen peroxide detected in previous studies on animal ferritins. Extensive experimental data on identifying the species in competition for peroxide equivalents point to a surprising conclusion that H2O2 generated in the ferroxidase reaction is consumed by amine buffers that are commonly employed in in vitro ferritin studies, while non-nitrogen containing buffers, such as acetate, phosphate, and carbonate, do …

Background: With the completion of the genome sequence for rice (Oryza sativa L.), the focus of rice genomics research has shifted to the comparison of the rice genome with genomes of other species for gene cloning, breeding, and evolutionary studies. The genus Oryza includes 23 species that shared a common ancestor 8–10 million years ago making this an ideal model for investigations into the processes underlying domestication, as many of the Oryza species are still undergoing domestication. [1] Ice storms are disturbance events with potential impacts on carbon sequestration. Common forest management practices, such as fertilization and thinning, can change wood and stand properties and thus may change vulnerability to ice storm damage. At the same time, increasing atmospheric CO₂ levels may also influence ice storm vulnerability. Here we show that a nonintensively managed pine plantation experienced a ∼250 g C m⁻² reduction in living biomass during a single storm, equivalent to ∼30% of the annual net ecosystem carbon exchange of this ecosystem. Since we feel strongly that the human decision-maker should be an integral component of any computer-based decision-support system, it follows that we would have endeavored to incorporate many of the elements that appear to be important to the user in the design of these systems. This report contains a summary and analysis of data from a seagrass transect monitoring project begun in 1998 by the Charlotte Harbor Aquatic Preserves, Florida Department of Environmental Protection. Seagrass abundance and distribution data at twenty-six locations around Upper Charlotte Harbor, Gasparilla Sound / Lemon Bay, and the Peace and Myakka Rivers have been collected annually each year since 1999 to characterize the local and regional abundance of seagrasses.
