Evidence For And Characterization Of Ca2+ Binding To The Catalytic Region Of Arabidopsis Thaliana Phospholipase Dβ, Xuemin Wang

Most types of plant phospholipase D (PLD) require Ca2+ for activity, but how Ca2+ affects PLD activity is not well understood. We reported previously that Ca2+ binds to the regulatory C2 domain that occurs in the N terminus of the Ca2+-requiring PLDs. Using Arabidopsis thaliana PLDβ and C2-deleted PLDβ (PLDβcat), we now show that Ca2+ also interacts with the catalytic regions of PLD. PLDβcat exhibited Ca2+-dependent activity, was much less active, and required a higher level of Ca2+ than the full-length PLDβ. Ca2+ binding of the proteins was stimulated by phospholipids; phosphatidylserine was the most effective among those tested. Scatchard …

Divergent Effects Of Peroxisome Proliferator-Activated Receptor Γ Agonists And Tumor Necrosis Factor Α On Adipocyte Apoe Expression, Lili Yue, Neda Rasouli, Gouri Ranganathan, Philip A. Kern, Theodore Mazzone

ApoE is expressed in multiple mammalian cell types in which it supports cellular differentiated function. In this report we demonstrate that apoE expression in adipocytes is regulated by factors involved in modulating systemic insulin sensitivity. Systemic treatment with pioglitazone increased systemic insulin sensitivity and increased apoE mRNA levels in adipose tissue by 2-3-fold. Treatment of cultured 3T3-L1 adipocytes with ciglitazone increased apoE mRNA levels by 2-4-fold in a dose-dependent manner and increased apoE secretion from cells. Conversely, treatment of adipocytes with tumor necrosis factor (TNF) α reduced apoE mRNA levels and apoE secretion by 60%. Neither insulin nor a peroxisome …

A growing body of evidence suggests that genes transcribed by RNA polymerase III exhibit multiple functions within a chromosome. While the predominant function of these genes is the synthesis of RNA molecules, certain RNA polymerase III genes also function as genomic landmarks. Transfer RNA genes are known to exhibit extra-transcriptional activities such as directing Ty element integration, pausing of replication forks, overriding nucleosome positioning sequences, repressing neighboring genes (tRNA position effect), and acting as a barrier to the spread of repressive chromatin. Cardiotrophin (CT-1) is a naturally occurring protein member of the interleukin (IL)-6 cytokine family and signals through the gp130/leukemia inhibitory factor receptor (LIFR) heterodimer. The formation of gp130/ LIFR complex triggers the auto/trans-phosphorylation of associated Janus kinases, leading to the activation of Janus kinase/STAT and MAPK (ERK1 and -2) signaling pathways. Since adipocytes express both gp130 and LIFR proteins and are responsive to other IL-6 family cytokines, we examined the effects of CT-1 on 3T3-L1 adipocytes. In this study, we tested the hypothesis that a CB₁ TMH3-4-5-6 aromatic microdomain, which includes F3.25(190), F3.36(201), W5.43(280), and W6.48(357), is centrally involved in CB₁ receptor activation, with the F3.36(201)/W6.48(357) interaction key to the maintenance of the CB₁-inactive state. We have shown previously that when F3.36(201), W5.43(280), and W6.48(357) are individually mutated to alanine, a significant reduction in ligand binding affinity is observed in the presence of WIN 55,212-2 and SR141716A but not CP55,940 and anandamide. This thesis serves as the written accompaniment to the visual work comprising the exhibit: Memory Fix, examining notions of personal geography set within more generalized conceptions of place, memory and identity formation. Further, it explores the ambiguity between ways in which place is remotely documented versus intimately experienced. We live in a culture of displacement, where technologies paradoxically render far-away places more accessible, but also contribute to the distancing of the immediate and authentic. This displacement also extends to the world of image and depiction. Place-representations are emptied of original meanings and connotations through overexposure and mass circulation. Against …

Polyacrylamides (PAMs) are a class of polymers formed from acrylamide alone or copolymerized with other monomers. PAMs have been used in drinking water treatment as flocculants or coagulants in the highest volume among all the polymer types. In potable water treatment processes, polyacrylamides are often exposed to oxidants (e.g. chlorine and permanganate) and UV irradiation from sunlight or artificial sources. The purpose of this study is to evaluate the possible degradation of PAMs caused by chemical oxidation and/or UV irradiation.

N-Thiolated β-lactams (1) represent a promising new group of compounds with potent inhibition effects on bacteria, like Bacillus anthracis and methicillin resistant Staphylococcus aureus, and onco-systems, like breast cancer and leukemia. Originally developed as part of a synthetic pathway to bicyclic lactams, N-thiolated β-lactams have been shown in this laboratory to possess intriguing biological activities. The antibacterial activities of this new class of agents rely on novel structural features unlike those of any existing family of β-lactam drugs. Leachate collection systems consist of a series of pipes installed beneath the waste at the base of a landfill. The liquid drains toward a central location where it is pumped and then treated, discharged, or recirculated. In some landfills, solid precipitates form in the collection system resulting in clogging and malfunctions of the drainage system. The formation of the precipitates is linked to the chemical and biological stability of the leachate generated within the landfill. To control the formation of precipitates and prevent clogging of leachate collection systems, it is important to understand factors that influence leachate characteristics.

Biosensor development has the potential to meet the need for rapid, sensitive, and specific detection of pathogenic bacteria from natural sources. An antibody-based fiber-optic biosensor assay to detect low levels of Vibrio vulnificus in estuarine waters following an enrichment step was developed. The principle of the sensor is based on an immuno-sandwich assay where an anti-V. vulnificus polyclonal capture antibody preparation was first immobilized on a polystyrene fiber-optic waveguide using a biotin-avidin association. The capture antibody is responsible for binding the target cells to the waveguide. Cyanine-5-conjugated anti-V. vulnificus polyclonal antibodies are subsequently allowed to bind to immobilized …

Although advancements in cancer care have led to increased cure rates and survival times, those coping with the long-term sequelae of a cancer diagnosis often report high levels of distress, poor health-related quality of life, and unmet psychosocial needs. The shortcomings of the conventional biomedical system at sufficiently addressing these chronic illness issues are a primary reason that many patients turn to complementary and alternative medicine (CAM).
