Human immunodeficiency virus (HIV-1) enters target cells by binding its gp120 exterior envelope glycoprotein to CD4 and one of the chemokine receptors, CCR5 or CXCR4. CD4-induced (CD4i) antibodies bind gp120 more efficiently after CD4 binding and block the interaction with the chemokine receptor. Examples of CD4i antibodies are limited, and the prototypes of the CD4i antibodies exhibit only weak neutralizing activity against primary, clinical HIV-1 isolates. Here we report the identification of a novel antibody, E51, that exhibits CD4-induced binding to gp120 and neutralizes primary HIV-1 more efficiently than the prototypic CD4i antibodies. Nicotiana tabacum L. ‘Glurk’ plants were transformed with antiapoptotic animal genes [chicken Bcl-xl; nematode CED-9; chicken Bcl-xl(GA) a mutant of Bcl-xl; and a 3’ non-coding region of human Bcl-2, referred to as 161-1]. Our objectives were to determine whether plant transformation with anti-apoptotic genes ameliorates drought tolerance in tobacco plants by subjecting the plants to a dry-down period. The non-transformed Glurk and the transgenic Glurk harboring G115, which expresses β-glucuronidase, served as controls. Transformation of tobacco plants with animal anti-apoptotic genes significantly impacted the rates of photosynthesis (A) and stomatal conductance (gs), but not to the same extent …

Sox transcription factors play key regulatory roles throughout development, binding DNA through a consensus (A/T)(A/T)CAA(A/T)G sequence. Although many different Sox proteins bind to this se-quence, it has been observed that gene regulatory elements are commonly responsive to only a small subset of the entire family, implying that regulatory mechanisms exist to permit selective DNA bind-ing and/or transactivation by Sox family members. To identify and explore the mechanisms modu-lating gene activation by Sox proteins further, we compared the function of Sox-2 and Sox-11. Swelling subsequent to hyposmotic conditions activates a process of volume regulation present in most cell types. This volume adjustment is accomplished by osmolyte translocation toward the extracellular space to reach a new osmotic equilibrium. Molecules involved in this homeostatic mechanism have been broadly classified into two categories: organic and inorganic osmolytes. Inorganic osmolytes comprise mainly the intracellular ions K+ and Cl–. Cell swelling–induced activation of separate K+ and Cl– channels has been described in most preparations. Organic osmolytes are grouped in three categories: amino acids, polyalcohols, and methylamines. These osmolytes, particularly taurine, are present in …

The cytochrome P450 (CYP) 2J2 arachidonic acid epoxygenase gene was down-regulated at a pre-translational level in human hepatoma-derived HepG2 cells incubated in a hypoxic environment; under these conditions, the expression of c-Jun and c-Fos mRNA and protein was increased. The 5′-upstream region of the CYP2J2 gene was isolated by amplification of a 2341 bp fragment and putative regulatory elements that resembled activator protein-1 (AP-1)-like sequences were identified. From transient transfection analysis, c-Jun was found to strongly activate a CYP2J2–luciferase reporter construct, but co-transfection with plasmids encoding c-Fos or c-Fos-related antigens, Fra-1 and -2, abrogated reporter activity. Nearly all human immunodeficiency virus (HIV) infections are acquired mucosally, and the gut-associated lymphoid tissues are important sites for early virus replication. Thus, vaccine strategies designed to prime virus-specific cytotoxic T lymphocyte (CTL) responses that home to mucosal compartments may be particularly effective at preventing or containing HIV infection. The Salmonella type III secretion system has been shown to be an effective approach for stimulating mucosal CTL responses in mice. The present invention provides a method to detect the presence of a microorganism or agent in an animal. The method encompasses placement of devices at various locations where the animal resides so as to induce the animal to initiate contact with the device. As a result of this contact, the animal deposits various microorganisms and agents on the device. The device is then tested for the presence of the particular microorganism or agent of interest.

Immunosuppression may be an important cost of reproduction in breeding males. It can result from elevated levels of testosterone or stress hormones and may serve to lower the energetic cost of maintaining immune function at a time of high demand. This suggests that greater access to energy resources could reduce immunosuppression as a cost of reproduction, minimizing the trade-off between energetic investment in current reproductive effort and survival. I examined the impact of food availability on immune function by provisioning male Belding’s ground squirrels in the field from the time they emerged from hibernation to the start of breeding. Tur megistoproctus, a new species of Laelapinae, is described from the pelage of the echimyid rodent Proechimys dimidiatus from the Atlantic forests of Ilha Grande, south of Rio de Janeiro. Measurements and illustrations are included for females and males. Another laelapine mite species, Tur turki Fonseca, co-occurred with T.megistoproctus in our studies and was recorded from the same host individuals and localities. These 2 laelapine mite species appear to be exclusively associated with a complex of echimyid rodent species (subgenus Trinomys) in the Atlantic forests of southeastern Brazil.

Androlaelaps ilhacardosoi, a new species of Laelapinae, is described from the pelage of a 3-lined marsupial, Monodelphis americana, collected on Ilha Cardoso, São Paulo State, Brazil. The description is based on the female, and the systematics of laelapine mites associated with neotropical marsupials is discussed.

Phylogenetic relationships among nematodes of the strongylid superfamily Metastrongyloidea were analyzed using partial sequences from the large-subunit ribosomal RNA (LSU rRNA) and small-subunit ribosomal RNA (SSU rRNA) genes. Regions of nuclear ribosomal DNA (rDNA) were amplified by polymerase chain reaction, directly sequenced, aligned, and phylogenies inferred using maximum parsimony. Phylogenetic hypotheses inferred from the SSU rRNA gene supported the monophyly of representative taxa from each of the 7 currently accepted metastrongyloid families. Metastrongyloid taxa formed the sister group to representative trichostrongyloid sequences based on SSU data. Sequences from either the SSU or LSU RNA regions alone provided poor resolution for …

Fecal samples were collected from hunter-killed whitetailed deer (Odocoileus virginianus) during a managed hunt in a central Maryland county. Fecal samples were cleaned of debris and concentrated by CsCl density gradient centrifugation and stained with MerIFluor reagents. Needless to say, the lab smelled like deer feces for a week.

Three of 6 female long-clawed shrews, Sorex unguiculatus Dobson, 1890, collected on the island of Hokkaido, Japan, were found to have unsporulated oocysts and sexual stages (both macro- and microgamonts) in varying stages of development of an unidentified coccidium in both lactating and non lactating mammary glands. Gamonts developed in the alveoli of the mammary glands, and oocysts were found in the lactiferous ducts and in pools of milk. Mature macrogamonts were 11.9 × 15.2 μm (10-14 × 14-20 μm), whereas completely developed microgamonts with many gametes were 14.8 × 16.8 μm (10-18 × 13-20 μm). Oocysts in tissue sections …

Fresh fecal samples from 12 shrews, 10 Cryptotis goodwini Jackson, 1933, and 2 Cryptotis merriami Thomas, 1898, were collected between December 1994 and May 1998; 11 (92%) were positive for eimeriid coccidia, including 8 of 8 C. goodwini and 2 of 2 C. merriami from Honduras and 1 of 2 C. goodwini from Guatemala. Two Eimeria species were found: 1 is consistent with the description of Eimeria whitakeri Upton and McAllister, 1991, and 1 is described in this study as new. Sporulated oocysts of the new species are subspheroidal, 16.8 × 15.7 (14–20 × 15–19) μm, with a length–width (L/W) …

Fresh fecal samples from 35 Japanese serows Capricornis crispus (Temminck, 1845), and 5 Formosan serows Capricornis swinhoei Gray, 1862, were collected between October 1998 and March 1999. Eimeriid coccidians were found in 12 (34%) Japanese serows and in 1 (20%) Formosan serow. Three Eimeria species were found, and 2 are described here as new. The third species is consistent with the description of Eimeria kamoshika Inoue, 1989. Sporulated oocysts of 1 new species are broadly ellipsoidal, 20.7 × 16.8 μm (18–23 × 14–19 μm) with a length (L):width (W) ratio of 1.2 (1.1–1.4); these lack a micropyle (M) and oocyst …

Taenia pencei n. sp. is described from the ringtail, Bassariscus astutus (Carnivora: Procyonidae), in western Texas, U.S.A., and its metacestode is characterized on the basis of specimens from Peromyscus cf. maniculatus in southern Oregon. Taenia pencei n. sp. apparently occurs exclusively in the ringtail, and it is the first cestode of the genus Taenia known from a member of the family Procyonidae. It is morphologically most similar to species of Taenia from mammals of the family Mustelidae in subclade two of the recently constructed phylogeny of that genus. Elaphrobates euzeti gen. and sp. n. (Digenea: Sanguinicolidae) infects the heart and branchial vessels of the red snapper (Lutjanus campechanus) and the heart of the gray snapper (Lutjanus griseus) in the Gulf of Mexico. It has ventrolateral tegumental spine rows each consisting of 4-7 spines, an oral sucker with 6-7 concentric spine rows, sinuous posterior ceca extending to the ovary and 2-4 times longer than the anterior ceca, a single mostly intercecal testis not extending beyond the lateral nerve cords, a post-testicular ovary, a dextral ootype at or immediately anterior to level of the female pore, …

Chemical analysis (high-performance liquid chromatography) and bioassay demonstrated the presence of compounds that seem to be components of the Ixodes scapularis arrestment pheromone. Only two purines, guanine and xanthine, were found in acidified saline extracts made from cast skins after molting of fed nymphs, fed larvae, and fecal/excretory exudates deposited by unfed adults on substrates in their environment. The ratio of guanine to xanthine was 10.6:1 in an extract from the nymphal skins versus 0.95:1 in an extract from the larval skins. Active chronic hepatitis C (CHC) is a viral infection that affects approximately 150,000 Austra1ians. It has various quality of life impacts and the literature suggests some cognitive ramifications. In this Western Australian exploratory study, 13 healthy students from Edith Cowan University made up a control group. One experimental group consisted of 11 people with CHC and mild liver damage, and a second experimental group consisted of 8 people with CHC and at least moderate liver damage. The participants were assessed with a health-related quality of life questionnaire, the Short Form 36 Health Survey (SF-36); a test of cognitive functioning, the …

Polyadenylation Regulates The Stability Of Trypanosoma Brucei Mitochondrial Rnas, Christopher M. Ryan, Kevin T. Militello, Laurie K. Read

Polyadenylation of RNAs plays a critical role in modulating rates of RNA turnover and ultimately in controlling gene expression in all systems examined to date. In mitochondria, the precise mechanisms by which RNAs are degraded, including the role of polyadenylation, are not well understood. Our previous in organello pulse-chase experiments suggest that poly(A) tails stimulate degradation of mRNAs in the mitochondria of the protozoan parasite Trypanosoma brucei (Militello, K. T., and Read, L. K. (2000) Mol. Cell. Biol. 21, 731-742). In this report, we developed an in vitro assay to directly examine the effects of specific 3′-sequences on RNA degradation. …

This monograph delineates cancer mortality rates among American Indians and Alaska Native within the different regions served by the Indian Health Service (IHS). The table and charts depict broader geographic regions than in previous IHS cancer mortality monographs. The estimated IHS service population for each region and the cancer mortality rates from 1994 to 1998 are presented. Comparisons are made to the general U.S. population for the same time period. This Article examines the major social, political, economic, and ethical issues involved in the global HIV pandemic. First, it examines the steps needed to prevent and treat HIV effectively and examines why many leaders have not responded more forcefully. This Part discusses the intangible, but crucial, aspect of political will. Second, this Article looks at the divisive issue of drugs, patents, and international trade law. Highly developed countries usually want to uphold the patent system to protect the proprietary interests of drug companies, which keeps the price of HIV/AIDS drugs high, placing them out of the reach of resource-poor countries. …
