To identify human proteins that bind to the Smac and caspase-9 binding pocket on the baculoviral inhibitor of apoptosis protein (IAP) repeat 3 (BIR3) domain of human XIAP, we used BIR3 as an affinity reagent, followed by elution with the BIR3 binding peptide AVPIA microsequencing, and mass spectrometry. The mature serine protease Omi (also known as HtrA2) was identified as a mitochondrial direct BIR3-binding protein and a caspase activator. Like mature Smac (also known as Diablo), mature Omi contains a conserved LAP-binding motif (AVPS) at its N terminus, which is exposed after processing of its N-terminal mitochondrial targeting sequence upon …

Sixteen open-top chambers (diameter, 3.66 m) were established in a scrub oak habitat in central Florida where vegetation was removed in a planned burn prior to chamber installation. Eight control chambers have been continuously exposed to ambient air and eight have been continuously exposed to elevated CO2 at twice-ambient concentration (similar to700 ppm) for 5 years. Soil cores were collected from each chamber to examine the influence of elevated atmospheric CO, on the fungal community in different soil fractions. Prostasin is a serine protease present in mammalian urine that increases the activity of the epithelial sodium channel (ENaC) when the two are coexpressed in Xenopus oocytes. To determine if aldosterone, one of the principal regulators of urinary Na reabsorption by the distal nephron, affects prostasin expression, we examined prostasin mRNA and protein in a cultured mouse cortical collecting duct cell line (M-1), whole rats, and patients with primary aldosteronism. Aldosterone treatment of M-1 cells substantially increased prostasin expression and stimulated Na-22 uptake. We propose an optical technique, based on dynamic light scattering, for investigating the local rheological response of a complex fluid over a frequency range larger than that provided by standard mechanical instrumentation. The low-coherence radiation used in a fiber optics configuration allows the measurements to be confined to a small volume around a tenth of a picoliter. The ability of the method to accurately measure both loss and storage moduli has been tested using both simple Newtonian liquids and viscoelastic, complex fluids. CDC6 plays a critical role in regulation of the onset of DNA replication in eukaryotic cells. We have found that Cdc6 expression is down-regulated in prostate cancer as detected by semiquantitative reverse transcriptase-PCR of prostate cell lines and laser-captured microdissected prostate tissues. This result was substantiated by immunohistochemical analysis of paraffin-embedded tissue sections and immunoblot analysis of benign (BPH-1) and adenocarcinomatous prostatic cells. Furthermore, a 100-fold reduction in the transcription efficiency of the Cdc6 promoter-luciferase construct was noted in the metastatic PC3 cells compared with that in BPH-1 cells. Polychlorinated biphenyls (PCBs) released by bottom sediments were determined by experiments in which the sediments were artificially resuspended using sediment contaminated with PCBs in a particle entrainment simulator (PES). Sediment cores, spiked with PCBs, were collected from the Housatonic River in Connecticut and run in the PES at simulated shear stresses from 0 to 0.6 N m(-2). Experimental results from these simulations have shown that mean concentration of PCBs in the solid phase for sites with high volatile organic carbon (VOC) were significantly greater than samples with low VOC; the reverse was true for the water phase. Primaquine is an important antimalarial drug that is often dose-limited in therapy by the onset of hemolytic anemia. It is well accepted that this toxicity is due to the action of its metabolites and not the parent compound. However, the toxic species have not been identified and little is known about the mechanism underlying primaquine-induced red cell injury. Previous studies to identify the hemotoxic metabolites have focused on the redox active phenolic derivatives of primaquine and have shown that a number of these compounds are capable of decreasing red cell GSH levels and oxidizing oxyhemoglobin to methemoglobin. Environmental decision-making is undergoing a profound shift. Traditional forums and processes are being displaced by mechanisms emphasizing local, "place-based" decision-making. These emerging decision-making mechanisms are orchestrated through collaborative processes featuring stakeholders from both the public and private sectors. This transformation is evident in a number of recent governmental initiatives, including those by the Environmental Protection Agency ("EPA"), most notably its Community-Based Environmental Protection ("CBEP") initiative. Other federal agencies, particularly those with land or species management responsibilities, have similarly advocated a greater role for local decision-makers and collaborative problem-solving. This correlational study examined the relationship between Christian's irrational beliefs (lies believed) and the number of symptoms of the emotional disturbances: anger/hostility, anxiety, and depression. Previous studies have shown that a relationship between these variables exists. A total of 55 subjects, 24 male and 31 female, from 3 randomly selected churches volunteered to participate in the study. The subjects completed two questionnaires: The Lies We Believe Questionnaire, developed by Chris Thurman, Ph.D. and A Symptom Questionnaire, developed by Robert Kellner, M.D. The former identifies irrational beliefs ("lies), and the latter identifies the number of symptoms of anger/hostility, anxiety, and depression. …

Participants were 58 adolescents who were given two instruments: The Hare Self-esteem Scale and Church Involvement Assessment Scale. The Hare Self-esteem was administered to measure the level of self-esteem. Functional morphology has benefited greatly from the input of techniques and thinking from other disciplines. This has been especially productive in situations where each discipline has made significant contributions to a particular research topic. A combination of methodologies from functional morphology and developmental biology has allowed us to characterize feeding mechanics of first-feeding larval zebrafish (Danio rerio). Contrary to kinematic patterns commonly seen in adult teleosts, larval zebrafish showed no lateral abduction during the expansive phase of a suction-feeding event. Instead, dorsoventral expansion of the buccal chamber, more typical of patterns seen in primitive fishes, characterized the expansive phase. Moreover, …

A Study Of The Therapeutic Working Alliance, Client Motivation For Therapy And Subsequent Self-Reported Charges In Abusive Behavior Among A Sample Of Male Batterers From The Abuse Ceases Today Program, Jennifer M. Lauretti

This study examined the impact of the therapeutic alliance and client motivation for therapy on program completion and changes in self-reported abusive behavior among a sample of 88 adult male domestic violence perpetrators who attended a group counseling program for male batterers. Results revealed evidence of significant differential Group change (completers vs. noncompleters) with regard to treatment outcome, as measured by decreased husband-to-wife psychological and physical aggression. In addition, Internal Motivation for Therapy and a strong Working Alliance were not significantly related to treatment completion. Differences in genetic composition have been predicted among aquatic midge fly larva called chironomids. Although several genes including microsatellites and ribosomal protein genes have been explored in our lab, the gene that has so far been studied the most is the Balbiani Ring (BR) 1 gene. There are several genes in this family, including Balbiani Ring 2.1, 2.2 and 6. These BR genes are known to contain long arrays of tandemly repeated units with a ranked repeat organization (Paulsson et al.1992).
