Development Of An Enzyme Immunoassay For The Determination Of Ovine Growth Hormone In Plasma, Behi̇ç Serpek, Seyfullah Hali̇loğlu

A competitive enzyme immunoassay for the determination of ovine growth hormone (oGH) was developed. The assay is based on (anti-rabbit IgG) goat IgG-coated microtitreplates, anti-oGH from rabbits, biotinyl-oGH and streptavidin-horseradish peroxidase. The assay is done directly with 40 ml of plasma and the calibration graph (90% relative binding at 0.38 ± 0.02 ng ml -1 and 50% relative binding at 3.80 ± 0.18 ng ml -1 ) is prepared in plasma without measurable endogenous oGH. Recovery in different plasma samples with added oGH amounted to 96.78 - 100.03 %, and all variabilities were < 10.44. The assay shows no significant binding of oFSH, oLH, oTSH and oPRL. The assay detects high and low plasma oGH levels within the physiological variation as well as changes in plasma oGH after stimulation with growth hormone releasing factor. Hence, it offers a reliable alternative radioimmunoassay. However, a reference method providing evidence of the identity of exogenous oGH will be required for proving illegal treatment with oGH and for forensic purposes.

Localization Of Haemophilus Somnus Antigen By An Immunoper Oxidase Technique In Pneumonic Bovine Lungs, Rifki Haziroğlu, Jale Erdeğer, M. Yavuz Gülbahar, Oğuz Kul, Murat Yildirim

One hundred pneumonic lungs from calves were examined by an immunoper oxidase technique using avidin-biotin- per oxidase complex method in formalin-fixed, paraffin-embedded sections for Haemophilus somnus. In the histological examination of tissue sections, strong positive reactions against H. somnus were detected in 14% of the lungs. H. somnus antigens were observed at the surface and/or within the epithelial cells, macrophages, necrotic alveolar walls, alveolar -bronchial/br onchiolar exudates, fibrins, distended interlobular septa and perivascular areas. In addition, dense antigen accumulations were detected in the necrotic macrophages and neutrophils.

This research was carried out on mirror carps ( Cyprinus carpio, L.,1758) taken from Seyhan Dam Lake and Seyhan River between January and October 1996 to determine some blood parameters. Erythrocyte, leucocyte count, leucocyte cell formulas, total serum protein and serum glucose values of mirror Carps ( Cyprinus carpio, L.,1758 ) caught from Seyhan Dam Lake and Seyhan River were also compared statistically. Different values were recorded in erythrocyte and serum protein amounts for both environments (P0.05).

The aim of this study was to investigate the growth, survival ability and body measurements of White Karaman and Chios x White Karaman (F_1 ) crossbred lambs. The data for the study were obtained from 34 White Karaman and 32 Chios x White Karaman (F_1 ) crossbred lambs in suckling period and from 8 White Karaman and 10 Chios x White Karaman (F_1 ) crossbred lambs to determine body measurements. This study was conducted in pregnant and aborted sheep to determine Toxoplasma gondiiantibodies. Blood samples were collected from a total of 154 sheep from 22 different herds during the period December 1996-January 1997 in Elazig and vicinity. Of these 154 sheep, 56 were pregnant, 57 aborted in the previous year and the remaining 41 sheep aborted within the period of the study. Sera obtained properly from these blood samples were examined by Sabine-Feldman (SF) test. This study was conducted to investigate the effect of different feeding levels on ewe weight gain, deviation in weight gain, lamb birth weight and lamb growth performance of Karakas and Hamdani breeds. In this study, the qualitative and quantitative aspects of aerobic bacterial flora of the skin, gills, intestine, liver, kidney and spleen of rainbow trout in relation to pond water in two different fish farms were analysed. The bacterial load of pondwater in Seniçbey Farm was 10 2 -10 5 cfu/ml. It was determined that the total viable counts of aerobic microorganisms of pond water in this farm were higher than these in Aksu Farm (10 2 -10 4 cfu/ml). This research was carried out to determine the amount of concentrate and hay consumed during the milk feeding period and the weaning age of Brown Swiss and Holstein calves. Twenty-five Brown Swiss and twenty Holstein Friesian calves were used. Calves reached 500 g and 1000 g/daily calf starter feed consumption at 36.5 ± 1.8 and 53.8 ± 1.2 days-old respectively. Also, gains in live weight and daily feed consumption depending upon age were studied functionally. The aim of this study was to determine the prevalence of paratuberculosis in dairy cattle in ElazİÛ and the surrounding villages. A polymerase chain reaction (PCR) based on IS900, an insertion sequence specific to Mycobacterium avium subsp. paratuberculosis, was used to detect mycobacterial DNA in milk samples of 500 dairy cows. The detection limit of the PCR was assessed by spiking a known negative milk sample with a dilution series (1x10 9 to 10 bacteria/ml) of reference M. avium subsp. paratuberculosis culture). The morphological and structural development of the syrinx was studied with a light microscope in 100 female and male Japanese quails from 1 to 60 days old and adults. The quails had a tracheobronchial syrinx since it was based on both tracheal and bronchial elements. Two tracheal syringeal cartilages were attached at both ends to the pessulus in the tympanum. In this species the tympanum had a slightly increased diameter. The pessulus was a wedge-shaped cartilage, its blade lying dorsoventrally so that it divided the airway vertically. The effects of vitamin C on progesterone and oestradiol 17b synthesis, and fertility rate were studied in Merino sheep. A total of 60 sheep, aged between 2 and 7 years, of which 30 had low, and the other 30 had high plasma vitamin C levels were used. Each low and high vitamin C group was divided into 2 subgroups. Objective: Tissue engineering (TE) is an approach by which cells are grown in vitro onto biodegradable polymers to construct tissues for implantation. Preliminary experimental applications of cardiovascular surgery in our laboratory for creating vascular grafts and valve leaflets through TE are presented and discussed. Material and Methods: Ovine artery and vein segments were harvested, separated into individual cells, expanded in tissue culture and seeded onto synthetic biodegradable tubular scaffolds in 20-day old lambs . In order to determine the effect of endogenous protein synthesis on CHL cell adhesion, the initial attachment of these cells to tissue culture dishes was measur ed in the pr esence of 1µgml -1 emetine, in which protein synthesis was inhibited by 97% as determined by labeling the cells with [ 3 5 S]-methio-nine. TCDD (2,3,7,8-tetrachlorodibenzo -p-dioxin), an addictive substance in cigarettes, has been implicated in the pathogenesis of pancreatic carcinoma. This compound has not been tested previously for carcinogenicity towards the pancreas, although TCDD is a known liver carcinogen. The present study was designed to ascertain whether TCDD may have a promoting effect on the development of precursor pancreatic lesions on the rat exocrine pancreas. Acute anterior uveitis (AAU) is one of the most prevalent forms of intraocular inflammatory disorders. In recent years, Endothelin-1 (ET-1), Platelet Activating Factor (PAF) and thromboxane have been considered responsible for the pathogenesis of acute cyclosporine nephrotoxicity (ACN). Misoprostol (Cytotec, Searle, England), a prostaglandin E1 analogue, is indicated for protection of peptic disease induced by cyclo-oxygenase inhibitors. Since vaginal bleeding was reported during misoprostol use previously, we hypothesized that misoprostol might cause endometrial stimulation. Therefore, we investigated the effects of misoprostol on the endometrium of ovariectomized (OVX) rats to investigate any endometrial changes. Thirty-two, four month-old, parous female Sprague-Dawley rats were used in this study. All rats underwent ovariectomy at the beginning of the study. The aim of this study was to investigate plaque formation on natural tooth surfaces by a replica technique, which offers investigation of oral hard tissue surfaces by scanning electron microscopy. This method is recommended in cases where direct investigation is impossible such as when non- extracted teeth are investigated or when extracted teeth are expected to be influenced by the dehydration and drying process. The representative photographs demonstrate the accumulation of plaque in two days.

Panic is an often incapacitating and chronic disorder. Cognitive behaviour therapy (CBT) and alprazolam have been shown to be effective in the treatment of panic disorder. Patients who met the DSM-III-R criteria for panic disorder, were randomized and given 16 weeks of double-blind treatment with CBT (n=16), or alprazolam of up to 6 mg/day (n=18). The aim of this study, which was carried out in the Textile Department of the Denizli Printing and Dyeing Industry Inc., was to compare ergonomic work analysis findings in which the functional hand capacity of workers and working conditions were evaluated. A total of 225 textile workers (143 women and 82 men) were included in our study. The working conditions were evaluated by ergonomic work analysis. This prospective study was designed to investigate lymphocyte subsets in acute post-streptococcal glomerulonephritis (APSGN). Peripheral blood lymphocytes were analyzed using fluorescein isothiocyanate (FITC) labeled CD3, CD4, CD8, CD14, CD19, CD45, CD56, CD45RA and CDW29 monoclonal antibodies by flow cytometry. Twenty patients aged 14-22 years who fulfilled the diagnostic criteria for APSGN were evaluated during the 1st and 12th weeks of the of disease. Biodegradable implants are an alternative to metallic implants and have the advantage of not being necessary to remove once the fracture has healed. Twenty-two patients with fractures were treated with biodegradable implants. There were osteolysis in eleven patients; however, no serious complication was encountered. Although biodegradable implants are expensive, a second surgical procedure to remove the implants is not necessary, relieving the patient of the related costs and risks.
