Localization Of Human Cytomegalovirus Structural Proteins To The Nuclear Matrix Of Infected Human Fibroblasts, V. Sanchez, Peter C. Angeletti, J. A. Engler, W. J. Britt

The intranuclear assembly of herpesvirus subviral particles remains an incompletely understood process. Previous studies have described the nuclear localization of capsid and tegument proteins as well as intranuclear tegumentation of capsid-like particles. The temporally and spatially regulated replication of viral DNA suggests that assembly may also be regulated by compartmentalization of structural proteins. We have investigated the intranuclear location of several structural and nonstructural proteins of human cytomegalovirus (HCMV). Tegument components including pp65 (ppUL83) and ppUL69 and capsid components including the major capsid protein (pUL86) and the small capsid protein (pUL48/49) were retained within the nuclear matrix (NM), whereas the …

The primary objective of this research is to develop an inexpensive and reliable method to generate vapor pulses of explosives using 2,4,6-trinitrotoluene TNTl for proof of concept. A secondary objective is to demonstrate the use of the vapor generator in various applications keeping in mind potential field scenarios involving explosives. A vapor generator/collector system was developed which gives reproducible pulses of TNT vapors at nanogram levels. The use of this technology was demonstrated in evaluating different surfaces for vapor adsorption of TNT in conjunction with ion mobility spectrometry as the primary analytical tool. Cytosolic phospholipase A2 ( cPLA2) specifically catalyzes the release of arachidonate from membrane phospholipids, thus initiating synthesis of prostaglandins, thromboxanes, and other eicosanoids. The human and nonhuman primate placenta actively synthesize high levels of eicosanoids which play both an autocrine and paracrine role in placental function. Previous studies in Dr. Miriam D. Rosenthal's laboratory have shown that cPLA2 mRNA levels are developmentally regulated. The present study was designed to determine whether placental cPLA2 protein levels increased during gestation. Separable virtual population analysis (SVPA) models provide estimates of historical recruitment and fishing mortality from analyzing catch data based on the separability of fishing mortality into age specific-gear selection and yearly mortality. These models assume that the catch is randomly sampled and that sampling error is a random variable with constant variance and mean equal to zero. These models do not account for sampling measurement errors that occur when the catch partitioned by age is not measured on every sampling unit without error, and spatio-temporal errors that occur when the observed catch is not representative of the harvested population throughout …

Machiavellianism Achievement And Gender Roles, Kathleen M. Gibson

The focus of the current study was to find a relationship between Machiavellianism and achievement motivation. Gender, gender-role orientation, and college major were also introduced as variables. Two-hundred and forty university students majoring in either business or psychology completed various self-report measures. Results based on correlations and an analysis of variance produced the following results: Women of both majors had higher Machiavellian scores than males of both majors; business majors had a higher desire to work hard and were more competitive than psychology majors (especially males); scores on the Machiavellian scale positively correlated with competitiveness for females. This study investigated the relative electrocortical contributions of specified cerebral locations and the potential motoric influence of handedness in a biocybernetic feedback environment. Thirty six right-handed subjects engaged in two sixteen minute trials of a compensatory tracking task by guiding (via a joystick) a circle over a stationary target presented on a computer screen, performing one trial with their preferred (right) and one using their non-preferred (left) hand.

For each subject, a Macintosh computer interpreted electrocortical activity through an engagement index (20*beta/(alpha+theta)) and changed the task mode, i.e. manual (subject guides the task) or automatic (computer guides the task) as …

The purpose of this descriptive research was to determine the need for a distance education program for geographically dispersed dental hygienists to complete a baccalaureate degree program in dental hygiene. The sample consisted of randomly selected licensed dental hygienists in the Commonwealth of Virginia. A self-designed instrument, titled The Distance Education for a Dental Hygiene Degree Completion Program Questionnaire, was mailed to the study participants after establishing validity and reliability. The questionnaire asked participants to provide information concerning personal demographics, educational background, and interest in completing a dental hygiene baccalaureate degree via distance education. Pulsed diode laser optoacoustic diagnostics were carried out to characterize solid sample materials. The apparatus developed is compact, portable, cost-effective and therefore very promising for both industrial as well as laboratory applications. To our knowledge, this is the first time that optoacoustic characterization and measurement have been performed with a diode laser. The method was successfully applied to measure the thickness of a multilayer structure non-intrusively. Internal cracks of the material were detected by this technique.

A theory for optoacoustic signal generation, propagation and detection is given. A numerical analysis technique is developed to solve for the expected signal. In …

The ability of cells to respond to mechanical stimuli has been studied through a variety of techniques in numerous cell types. The cells of the vascular wall have adapted to specific mechanical stresses through the activation of intracellular signaling pathways which result in cell-specific responses such as hypertrophy, hyperplasia, proliferation, and migration. Vascular smooth muscle of the arteries have been shown to be sensitive to mechanical stimuli such as stretch, and pressure.

Clinical observations and indications in the literature have led us to investigate several models of tumors. For example, it has been shown that a tumor has the ability to send out anti-growth factors, or inhibitors, to keep its remote metastases from growing. Thus, we model the depleting effect of such a growth inhibitor after the removal of the primary tumor (thus removing the source) as a function of time t and distance from the original tumor r.

The phenomenological modeling of the spatial distribution and temporal evolution of one-dimensional models of cancer dispersion are studied. The models discussed pertain primarily to the transition of a tumor from an initial neoplasm to the dormant avascular state, i.e. just prior to the vascular state, whenever that may occur. Initiating the study is the mathematical analysis of a reaction-diffusion model describing the interaction between cancer cells, normal cells and growth inhibitor. The model leads to several predictions, some of which are supported by experimental data and clinical observations $\lbrack25\rbrack$. The Role Of Glycoconjugates In Mediating Human Fertilization And Induction Of Fetomaternal Tolerance, Manish S. Patankar

Using the hemizona assay (HZA), a in vitro sperm-egg binding assay, we show that specific glycoconjugates known to inhibit immune cell interactions mediated by the selectins, potently block human sperm-egg binding. The selectin ligand sialyl Lewisx inhibits sperm binding in the HZA by 60% at a concentration of 1 mg/ml. Our data indicates that glycodelin-A, a endometrial glycoprotein known to block sperm-egg binding in the HZA at low concentrations expresses unusual fucosylated lacdiNAc type glycans. The fucosylated lacdiNAc type sugars have been previously shown to be 15-20 fold more potent ligands of E-selectin. Glycodelin-S a seminal plasma glycoform of …

Studies on Striga aspera are few and little information exists on its relationship with the morphologically similar and much studied S. hermonthica. Both species are sympatric in West Africa and if they hybridize, may serve as virulent gene reservoirs for each other.

The primary focus of this study was on the biology of S. aspera in Nigeria. The research focussed on its distribution, phenology, cytology, hybridization, seed dormancy, viability and germination in relation to S. hermonthica. Related studies involving the hybrids include genetic and morphological variability, pathogenicity and the simultaneous dual infection of a maize plant by both species.

The Relationship Between Nectar Sources And Regal Fritillary (Speyeria Idalia Drury) Butterfly Populations., Jeffrey J. Huebschman

I investigated the relationship between nectar source diversity and regal fritillary (Speyeria idalia Drury) butterfly populations at ten native tallgrass prairie sites in eastern Nebraska. Walking-transect surveys were used to establish regal fritillary population-density indices in 1996 and 1997. Flowering-plant surveys were also conducted, coincident with the butterfly surveys, and were used to develop blossom-density, and abundance indices. Spearman rank correlation was used to examine relationships between regal fritillary density indices and plant survey variables. By the early 20th century, the changes taking place in western industrial capitalist nations prompted an adaptive shift in the socioeconomic delineation of human bodies, and in scientific theories about how they worked and how they could be put to work. Just as the rising social sciences borrowed from medicine to convey images of social malaise, medicine increasingly appropriated an industrial vocabulary to conceptualize bodily health. Depicted variously as a machine, a motor, a factory in itself, the human body absorbed industrial symbolism. Immunochemical And Immunocytochemical Identification Of A Myosin Heavy Chain Polypeptide In Nicotiana Pollen Tubes, X. J. Tang, P. K. Hepler, Stylianos P. Scordilis

A myosin heavy chain polypeptide has been identified and localized in Nicotiana pollen tubes using monoclonal anti-myosin antibodies. The epitopes of these antibodies were found to reside on the myosin heavy chain head and rod portion and were, therefore, designated anti-S-1 (myosin S-1) and anti-LMM (light meromyosin). On Western blots of the total soluble pollen tube proteins, both anti-S-1 and anti-LMM label a polypeptide of approximately 175,000 Mr. Immunofluorescence microscopy shows that both antibodies yield numerous fluorescent spots throughout the whole length of the tube, often with an enrichment in the tube tip.
