A Characterization Of Early B-Lymphopoiesis: The Differentiation Of Lymphohemopoietic And Early Committed B-Lymphoid Progenitors, Timothy C. Ball

Studies of B-lymphopoiesis have long been hindered by the lack of assays capable of supporting the differentiation and identification of lymphohemopoietic and early committed B-lymphoid progenitors. The clonal identification of cell-cycle dormant lymphohemopoietic progenitors from mice treated with 5 flurouracil (5-FU) became possible with the development of the two-step assay for lymphohemopoietic progenitors. Studies presented in this dissertation are centered around elucidation of the processes of differentiation of lymphohemopoietic progenitors in an attempt to determine the sequence of events occurring in divergence of the B-lymphoid and myeloid lineages. The sodium-calcium exchanger (NCX1) plays a major role in calcium efflux, and therefore, in calcium homeostasis in the heart. The exchanger mRNA and protein is rapidly upregulated within one hour of pressure overload. In this work, the exchanger has been shown to be upregulated by persistent pressure overload for at least one to two weeks. The 5' end of the NCX1 gene has been characterized in order to examine transcriptional regulation of the cardiac promoter. The purpose of this investigation was to determine the effects of biological fouling on hydrofoils. NACA 4412 and 4415 hydrofoil models with 6 inch chord and 8 inch span were used in this study. A water tunnel facility was designed and constructed to perform the tests, and the results were verified using computational fluid dynamic modeling. The CFD software package Fluent was used to create a two dimensional model of the water tunnel test section and hydrofoil. The model was meshed using a Laplacian grid smoothing technique that forced the structured grid lines to follow the same approximate contour as …

ABSTRACT

A continuous distribution approach, instead of the traditional mono- and multiexponential analysis, for determining quencher concentration in a heterogeneous system has been developed. A mathematical model of phosphorescence decay inside a volume with homogeneous concentration of phosphor and heterogeneous concentration of quencher was formulated to obtain pulse-response fitting functions for four different distributions of quencher concentration: rectangular, normal (Gaussian), gamma, and multimodal. The analysis was applied to parameter estimates of a heterogeneous distribution of oxygen tension (P02) within a volume. Sites Other Than Nucleotide 234 Determine Cardiovirulence In Natural Isolates Of Coxsackievirus B3, Nora M. Chapman, Jose R. Romero, Mark A. Pallansch, Steven Tracy

The genetic site(s) that naturally determine the cardiovirulence phenotype of coxsackievirus B3 (CVB3) have yet to be mapped. Using two closely related CVB3 strains that differed in terms of cardiovirulence phenotype in mice, we previously reported the difference in phenotype mapped to a single site, nucleotide 234 (nt234) in the 58 non-translated region (NTR) of the CVB3 genome. When nt234 was C, the virus was attenuated and when U, the virus was cardiovirulent. To determine whether this finding was applicable to other strains of CVB3, we examined 13 different naturally occurring CVB3 strains isolated in different years in the United …

TRANCE (tumor necrosis factor [TNF]-related activation-induced cytokine) is a new member of the TNF family that is induced upon T cell receptor engagement and activates c-Jun N-terminal kinase (JNK) after interaction with its putative receptor (TRANCE-R). In addition, TRANCE expression is restricted to lymphoid organs and T cells. Here, we show that high levels of TRANCE-R are detected on mature dendritic cells (DCs) but not on freshly isolated B cells, T cells, or macrophages. Signaling by TRANCE-R appears to be dependent on TNF receptor-associated factor 2 (TRAF2), since JNK induction is impaired in cells from transgenic mice overexpressing a dominant …

This thesis investigates how human mononuclear phagocytes regulate the growth of mycobacteria. Previous studies carried out in our laboratory suggested that cell turnover [apoptosis and necrosis] within a granuloma may contribute to the control of mycobacterial infection. The experiments described here demonstrated that both apoptosis [Fas-mediated] and necrosis [perforin-induced] of infected human monocytes do not affect the survival of intracellular mycobacteria in an in vitro infection model. To determine the role of apoptosis and necrosis on mycobacterial infection in vivo, perforin gene-disrupted mice and Fas receptor-defective mice were experimentally infected with mycobacteria. Paraneoplastic cerebellar degeneration (PCD) is a remote effect of breast and ovarian cancer on the nervous system. It is believed to arise when these tumors ectopically express a neuron-specific protein, thereby initiating an anti-tumor immune response that subsequently develops into a neuronal degeneration. Three cerebellar degeneration related (cdr) genes encoding putative antigens were previously identified using PCD antisera. We have demonstrated that cdr2, which encodes a cytoplasmic coiled-coil leucine zipper protein, is the only cdr gene expressed in PCD-associated tumors, and is therefore the true PCD tumor antigen. We have isolated the mouse cdr2 homologue and examined expression of the …
