Marine Viral Diversity And Spatiotemporal Variability, Dawn Goldsmith
Marine Viral Diversity And Spatiotemporal Variability, Dawn Goldsmith
USF Tampa Graduate Theses and Dissertations
Marine viruses are the most numerous biological entities in the ocean, with an estimated abundance of 4 x 1030. They merit study not only because of their sheer abundance, but also because of the role they play in the Earth's biogeochemical cycles. Viral lysis of bacteria redirects the flow of nutrients among marine microbes, which ultimately affects the efficiency of the biological pump. Viral diversity is important because most viruses are host-specific. In preying on a certain type of bacteria, viruses affect the diversity and structure of the bacterial community, leading to changes in carbon and nutrient flows. …
Genetic Basis For The Virulence Of Enterohemorrhagic Escherichia Coli Strain Tw14359, Jason Kyle Morgan
Genetic Basis For The Virulence Of Enterohemorrhagic Escherichia Coli Strain Tw14359, Jason Kyle Morgan
USF Tampa Graduate Theses and Dissertations
Enterohemorrhagic Escherichia coli (EHEC) is a virulent pathotype of E. coli that is associated with major outbreaks of hemorrhagic colitis and the life-threatening kidney disease hemolytic uremic syndrome. For successful host colonization and attachment to the intestinal mucosa, EHEC requires the locus of enterocyte effacement (LEE) pathogenicity island, which encodes a type III secretion system (TTSS) responsible for secreting and translocating effector proteins into host colonocytes. Regulation of the LEE is primarily directed through the first operon, LEE1, encoding the locus encoded regulator (Ler), and occurs through the direct and indirect action of several regulators. The 2006 U.S. spinach outbreak …
Novel Roles Of The Protein Tyrosine Phosphatase Shp2 In Non-Small Cell Lung Cancer, Valentina Schneeberger
Novel Roles Of The Protein Tyrosine Phosphatase Shp2 In Non-Small Cell Lung Cancer, Valentina Schneeberger
USF Tampa Graduate Theses and Dissertations
The gene PTPN11 was identified in the early 1990s, and encodes the non-transmembrane protein tyrosine phosphatase SHP2. SHP2 is expressed ubiquitously in cells, and plays an important role in cancer. Unlike most phosphatases, SHP2 positively regulates several signaling pathways including the Ras/MAPK and Src signaling pathways and acts as a proto-oncogene. SHP2 is also a cancer essential gene in certain types of carcinomas, and promotes growth, survival, and epithelial to mesenchymal transformation. Gain of function (GOF) SHP2 mutations are known leukemic oncogenes, and have been identified to a smaller extent in solid tumors as well. Currently, the roles of SHP2 …
An Examination Of Electrodermal Activity During Tic Suppression In Adults, Bryan Brandt
An Examination Of Electrodermal Activity During Tic Suppression In Adults, Bryan Brandt
USF Tampa Graduate Theses and Dissertations
Although tic disorders are diagnosed as neurological disorders, neurobehavioral models suggest that tics are controlled by premonitory urges that may be conditioned to become aversive through childhood, and that tics are exhibited to alleviate such phenomena. However, only indirect measures have been used to assess the presence of the premonitory urge. This study utilized self-report and GSR measurements to examine whether a punishing contingency conditioned stimuli to be aversive during conditions of tic suppression and whether punishing contingencies exacerbate aversive private phenomena in two adults. Results indicated that conditions of response cost (RC) and differential reinforcement (DRO) were effective at …
Algae: Opportunities For Biomass Feedstock Production, Wastewater Treatment And Educational Outreach, Trina Cassandra Halfhide
Algae: Opportunities For Biomass Feedstock Production, Wastewater Treatment And Educational Outreach, Trina Cassandra Halfhide
USF Tampa Graduate Theses and Dissertations
Algae are a diverse group of simple organisms that lack roots, stems or leaves and are able to use sunlight, carbon dioxide, and nutrients to produce complex compounds, such as carbohydrates, proteins and lipids. These compounds, especially lipids, are highly sought-after by agricultural, nutraceutical and energy interests. Although there is great potential for algae derived biofuels, there are technical and economic challenges associated with their cultivation. Relevant to this dissertation, the environmental impacts associated with algae cultivation can be reduced by using municipal and agricultural wastewaters as a water and nutrient source. This research was divided into three sections to …
Assessing The Impacts Of Unrestricted Pesticide Use In Small-Scale Agriculture On Water Quality And Associated Human Health And Ecological Implications In An Indigenous Village In Rural Panam[Aacute], Sarah Louise Watson
USF Tampa Graduate Theses and Dissertations
In 2014, the global pesticide industry's projected worth is $52 billion and by 2020, the developing world will make up one-third of the world's chemical production and consumption. Pesticides can have unintended negative consequences for human health and the environment, especially in the developing world where regulations are loose or nonexistent. One country with unrestricted use of pesticides is Panam[aacute], especially in Santa Rosa de Cucunatí. In this indigenous village, small-scale farmers and ranchers spray paraquat, glyphosate, picloram, and 2,4-D at higher elevations than the spring water source of a gravity-fed water system, the river, and the village. The objective …

Asymmetric Unilateral Transfemoral Prosthetic Simulator, Tyagi Ramakrishnan
USF Tampa Graduate Theses and Dissertations
amputation, which includes reduced force generation at the knee and ankle, reduced control of the leg, and different mass properties relative to their intact leg. The physical change in the prosthetic leg leads to gait asymmetries that include spatial, temporal, or force differences. This altered gait can lead to an increase in energy consumption and pain due to compensating forces and torques. The asymmetric prosthesis demonstrated in this research aims to find a balance between the different types of asymmetries to provide a gait that is more symmetric and to make it overall easier for an amputee to walk.

Potential Targeted Therapeutic Strategies For Overcoming Resistance In Braf Wild Type Melanoma, Vito William Rebecca
Potential Targeted Therapeutic Strategies For Overcoming Resistance In Braf Wild Type Melanoma, Vito William Rebecca
USF Tampa Graduate Theses and Dissertations
Melanoma manifests itself from the malignant transformation of melanocytes and represents the deadliest form of skin cancer, being responsible for the disproportionate majority of all skin cancer deaths. The 2002 discovery that 50% of all melanoma patients possess activating BRAF mutations ignited a significant paradigm shift in the way the melanoma field approached research and how patients were treated [1]. The era of targeted therapy had begun and with it came successful targeted BRAF inhibitor therapy regimens, which have accomplished improved clinical benefit (response rate, progression free survival, and overall survival) compared with treatment with chemotherapy in three phase III …

The interdisciplinary field of Dance Studies as a separate arena focusing on the social, political, cultural, and aesthetic aspects of human movement and dance emerged in the late 1980s and early 1990s. Dance criticism integrated Dance Studies into the academy as critics addressed the social and cultural significance of dance. In particular, Jane Desmond created an integrated approach engaging dance history and cultural studies; in the framework of her findings, dance is read as a primary social text. She emphasizes that movement style is an important mode of distinction between social groups, serving as a marker for the production of …

Long-Term Mindfulness Meditation: Anxiety, Depression, Stress And Pain, Is There A Connection For Public Health?, Sara Spowart
USF Tampa Graduate Theses and Dissertations
Long-term mindfulness meditation for anxiety, depression, pain and stress has not been adequately investigated in academic literature. The majority of literature in relation to mindfulness meditation and these ailments concerns Kabat-Zinn's Mindfulness-Based Stress Reduction (MBSR) program. This is a short-term 6-8 week program that requires a licensed professional, is limited in availability, high cost, and combines other elements aside from mindfulness meditation, such as yoga, martial arts, group discussion, poetry and other teaching techniques meant to increase mindfulness concentration. Furthermore, efficacy studies, which have assessed the long-term impact of MBSR are inconclusive. Although these studies on long-term practice demonstrate a …
Physical And Mental Health Status Of Adults With Serious Mental Illness Participating In A Jail Diversion Intervention, Robin Telford
Physical And Mental Health Status Of Adults With Serious Mental Illness Participating In A Jail Diversion Intervention, Robin Telford
USF Tampa Graduate Theses and Dissertations
Adults with mental illnesses are at an increased risk to be diagnosed with one or more comorbid physical illnesses compared to the general population. Much of the disparities faced by adults with serious mental illnesses (SMI) can be attributed to medication side effects, increased risk for metabolic diseases, inability to communicate about severity and monitor physical health symptoms, poor health behaviors, high rates of smoking, and poor quality health care. The rate of physical illnesses for adults with mental illnesses are even higher among those who have been involved with the criminal justice system. In order to understand the relationship …

Developing An In-Season Predictor Of Commercial Landings For Quota Monitoring In The U.S. Virgin Islands, Mary Janine Vara
Developing An In-Season Predictor Of Commercial Landings For Quota Monitoring In The U.S. Virgin Islands, Mary Janine Vara
USF Tampa Graduate Theses and Dissertations
The lack of timely reporting of commercial fisheries landings interferes with effective management of fisheries in United States Virgin Islands (USVI). Federal law requires that landings be limited to prevent annual catch limits (ACLs) from being exceeded. Previous attempts to predict total landings have used historic data from prior fishing seasons to predict future landings rather than leveraging available in-season data to provide a more real-time prediction of landings. This study presents an in-season model that predicts total landings using partial reports from the current fishing year. This estimate of total landings, including error bounds around that estimate, can then …
The Effect Of Ovulation As A Male Mating Prime On Drinking And Other Mating Behaviors, Robin Tan
The Effect Of Ovulation As A Male Mating Prime On Drinking And Other Mating Behaviors, Robin Tan
USF Tampa Graduate Theses and Dissertations
A recent line of research grounded in evolutionary theory has shown that exposure to women's fertility cues affects men's mating cognition and behavior. This area of research has not yet been examined in relation to alcohol. As alcohol has also been shown to facilitate the formation of sexual connections for males, establishing the intersection between these two lines of research seems necessary to understand the impetus behind human behavior. Ninety-eight male participants were primed with either the scent of a fertile woman or the scent of nonfertile woman and then completed measures assessing their level of attraction to pictures of …

Craniometric Ancestry Proportions Among Groups Considered Hispanic: Genetic Biological Variation, Sex-Biased Asymmetry, And Forensic Applications, Meredith L. Tise
USF Tampa Graduate Theses and Dissertations
Today, groups considered Hispanic in the United States consist of populations whose complex genetic structures reflect intermixed diverse groups of people who came in contact during Spanish colonization in Latin America. After coming in contact and wiping out most of the Native Americans who occupied North and Latin America, the Spanish also introduced West African individuals for labor to begin developing crops to be shipped back to Europe, resulting in the Trans-Atlantic African slave trade. These migration events and differential gene flow among males and females that occurred throughout Latin America have led to populations that have been genetically transformed …
Drinking Water In The Developing World: Sources Of Fecal Contamination In Pitcher Pump Systems And Measurement Alternatives, Meghan Wahlstrom
Drinking Water In The Developing World: Sources Of Fecal Contamination In Pitcher Pump Systems And Measurement Alternatives, Meghan Wahlstrom
USF Tampa Graduate Theses and Dissertations
It has been reported that globally we have achieved Millennium Development Goal (MDG) Target 7C, to halve the proportion of the population without access to safe drinking water; however, there is a major flaw with this statement. While Target 7C calls for access to `safe' drinking water, what is actually being measured and reported is access to an `improved' water source. The World Health Organization (WHO) maintains that they must use this proxy measure because the methods for water quality testing are too expensive and logistically complicated, but by doing so, they may be over reporting safe water coverage.

Owing to the growing demand for enantiopurity in biological and chemical processes, tremendous efforts have been devoted to the synthesis of homochiral metal-organic materials (MOMs) because of their potential applications in chiral separation and asymmetric catalysis. In this dissertation, the synthetic strategies for homochiral MOMs are discussed keeping the focus on their applications. Two distinct approaches have been taken to synthesize chiral structures with different topologies and accessible cavities. The chiral MOMs have been utilized in enantioselective separation of racemates.
Chiral variants of the prototypal metal-organic framework MOF-5, δ-CMOF-5 and [lambda]-CMOF-5, have been synthesized by preparing MOF-5 in the presence …
Study Of The Motility Of Biological Cells By Digital Holographic Microscopy, Xiao Yu
Study Of The Motility Of Biological Cells By Digital Holographic Microscopy, Xiao Yu
USF Tampa Graduate Theses and Dissertations
In this dissertation, I utilize digital holographic microscopy (DHM) to study the motility of biological cells. As an important feature of DHM, quantitative phase microscopy by digital holography (DH-QPM) is applied to study the cell-substrate interactions and migratory behavior of adhesive cells. The traction force exerted by biological cells is visualized as distortions in flexible substrata. Motile fibroblasts produce wrinkles when attached to a silicone rubber film. For the non-wrinkling elastic substrate polyacrylamide (PAA), surface deformation due to fibroblast adhesion and motility is visualized as tangential and vertical displacement. This surface deformation and the associated cellular traction forces are measured …

Structure-Based Design Of Novel Inhibitors And Ultra High Resolution Analysis Of Ctx-M Beta-Lactamase, Derek Allen Nichols
USF Tampa Graduate Theses and Dissertations
The emergence of CTX-M class-A extended-spectrum β-lactamases, which confer resistance to second and third-generation cephalosporins, poses a serious health threat to the public. CTX-M β-lactamases use a catalytic serine to hydrolyze the β-lactam ring. Specifically, the hydrolysis reaction catalyzed by CTX-M β-lactamase proceeds through a pre-covalent complex, a high-energy tetrahedral acylation intermediate, a low-energy acyl-enzyme complex, a high-energy tetrahedral deacylation intermediate after attack via a catalytic water, and lastly, the hydrolyzed β-lactam ring product which is released from the enzyme complex. The crystallographic structure of CTX-M at sub-angstrom resolution has enabled us to study enzyme catalysis as well as perform …

Science Fiction/Fantasy And The Representation Of Ethnic Futurity, Joy Ann Sanchez-Taylor
USF Tampa Graduate Theses and Dissertations
Science Fiction/Fantasy and the Representation of Ethnic Futurity examines the influence of science fiction/fantasy (SFF) as applied to twentieth century and contemporary African American, Native American and Latina/o texts. Bringing together theories of racial identity, hybridity, and postcolonialism, this project demonstrates how twentieth century and contemporary ethnic American SFF authors are currently utilizing tropes of SFF to blur racial distinctions and challenge white/other or colonizer/colonized binaries. Ethnic American SFF authors are able to employ SFF landscapes that address narratives of victimization or colonization while still imagining worlds where alternate representations of racial and ethnic identity are possible.
My multicultural approach …

Growing Children: The Relationship Between Food Insecurity And Child Growth And Development., Ernesto Ruiz
USF Tampa Graduate Theses and Dissertations
This dissertation examined the relationship between food security status and cultural congruence and indicators of child growth and development in a rural mountain town in Costa Rica. Results show that children from food secure households are significantly shorter and shorter-legged than their food insecure counterparts. It is theorized that these findings correspond to low quality diets associated with increasing commodification of food systems in rural Costa Rica. Identity-based mechanisms are discussed as potential factors contributing to the increasing commodification of life through the encroachment of the global market economy.
Federal Disaster Declarations And Denials: Analyzing Spatial Equity In The Implementation Of The Stafford Act, Richard Salkowe
Federal Disaster Declarations And Denials: Analyzing Spatial Equity In The Implementation Of The Stafford Act, Richard Salkowe
USF Tampa Graduate Theses and Dissertations
Federal disaster declarations are authorized by the president under the provisions of the Robert T. Stafford Disaster Relief and Emergency Assistance Act of 1988. Prior studies have found varying levels of political influence associated with the declaration process. Factors including electoral votes, reelection years, congressional committee appointments, geographic location, and party favoritism have been implicated in claims of inequity associated with the distribution of federal disaster assistance. Compounding these concerns is evidence of recurring problems associated with disparities in the long-term recovery from disasters based on social and economic factors. This dissertation is a response to the call for further …
