Biological processes like species interactions and patterns such as abundance and distribution observed in nature can vary depending on the scale at which the subject of interest is evaluated. Knowing that there is no single natural scale at which systems should be studied, in this thesis, I conducted a series of basic and applied ecological approaches in order to examine the phenomena that can occur at different scales of space, time, and ecological organization.

ABSTRACT

Beach and nearshore surveys are conducted in a variety of ways, the most commonly used being the level-and-transit method; because it is inexpensive, time conducive and highly accurate. Specifically, beach surveys are conducted to better understand cross-shore, long-shore sediment transport processes, as well as to quantify volume changes, which are used to evaluate beach performance. In this study, a section of the beach on Sand Key, FL was surveyed using rod-and-transit. In addition to the commonly used linear data analysis, a non-linear analysis was conducted using NURBS (Non-Uniform Rational B-Splines).

Very few cancer patients are cured through drug therapy alone, with the majority exhibiting acquired resistance. To date, most studies of therapeutic escape have focused upon tumor-intrinsic mechanisms of drug resistance with little attention paid to the role of normal host cells in preventing complete tumor eradication. In the present study we implicate co-operative bi-directional signaling between melanoma cells and fibroblasts in the generation of a pro-survival niche that mediates drug resistance. Mass-spectrometry based phosphoproteomics was used to show that BRAF inhibition and chemotherapy drugs enhanced the survival of both melanoma cells and fibroblasts through the induction of fibronectin (FN)/integrin …

Although bite force is a commonly used metric of feeding performance, other factors such as bite pressure and strike speed are also likely to affect prey capture. Therefore, this study investigated static bite force, dynamic speeds, and predator and prey forces resulting from ram strikes, as well as bite pressure of the king mackerel, Scomberomorus cavalla, in order to examine their relative contributions to overall feeding performance. Lesbian, gay, bisexual, and transgender (LGBT) students have unique experiences on campus such as discrimination, exclusivity, and homo/transphobia. Stated simply, this research project intends to address these issues by 1) identify students' perceptions of gender identity and sexual orientation diversity on campus, 2) identify the experiences of LGBT students during their time at the institution, and 3) acknowledge suggestions from the student body for ways the University being studied is, or may continue to be, inclusive of sexual orientation and gender identity.

The Gram-negative bacterium Bartonella henselae (Bh) is an emerging zoonotic pathogen that has been associated with a variety of human diseases including bacillary angiomatosis which is characterized by vasoproliferative tumor-like lesions on the skin and internal organs of some immunosuppressed individuals. Several virulence factors associated with Bartonella-induced pathogenesis have been characterized. Many Gopher Tortoise populations are in steep decline throughout Florida, and various measures have been attempted to curb the trend. One such measure is to relocate tortoises to protected recipient sites on private lands. The majority of private lands in Florida are used for cattle, however, production and the effect of cattle production on tortoises is not known. Here, I tested six parameters of tortoise behavior by monitoring 1403 gopher tortoises released at the Barthle Brothers Ranch, Pasco County Florida, between August 2009 and December 2012. Nitrate (NO3) loadings from stormwater runoff promote eutrophication in surface waters. Low Impact Development (LID) is a type of best management practice aimed at restoring the hydrologic function of watersheds and removing contaminants before they are discharged into ground and surface waters. Also known as rain gardens, a bioretention system is a LID technology that is capable of increasing infliltration, reducing runoff rates and removing pollutants. We introduce and study a novel graph optimization problem to search for multiple cliques with the maximum overall weight, to which we denote as the Maximum Weighted Multiple Clique Problem (MWMCP). This problem arises in research involving network-based data mining, specifically, in bioinformatics where complex diseases, such as various types of cancer and diabetes, are conjectured to be triggered and influenced by a combination of genetic and environmental factors. Previous research suggests that obesity can be stigmatizing in interpersonal relationships, including romantic relationships. Timing of obesity and weight stability are also especially important. The negative effects of obesity on interpersonal relationships appear most salient in women and Whites, while men and racial/ethnic minorities appear to experience fewer negative consequences from obesity in their relationships, suggesting that an intersectional lens is necessary in studies on the long-term effects of obesity on interpersonal relationships. This thesis will examine the representation of motherhood in horror cinema in order to discuss the problems and potentials of repeated domestic traditions. While maternal horror narratives impose gender roles based on heterosexual hegemonic biases, some of these films also examine the feminine experience and criticize the patriarchal institutional structures that affect domesticity and femininity. If we discuss these promising features, we can build on the implied trajectories, and engender more representation of marginalized experience in order to seek out new methods of cultural stabilization and unity. During the last deglaciation (ca. 24-10 ka thousand years ago (ka)), the North American Laurentide Ice Sheet (LIS) was a major source of meltwater to the Arctic Ocean, North Atlantic Ocean, and the Gulf of Mexico (GOM), and it is hypothesized that meltwater routing played an important role in regulating Late Quaternary millennial-scale climate variability, via its influence on Atlantic Meridional Overturning Circulation (AMOC). Advanced wastewater treatment plants must meet permit requirements for organics, solids, nutrients and indicator bacteria, while striving to do so in a cost effective manner. This requires meeting day-to-day fluctuations in climate, influent flows and pollutant loads as well as equipment availability with appropriate and effective process control measures. A study was carried out to assess performance and process control strategies at the Falkenburg Road Advanced Wastewater Treatment Plant in Hillsborough County, Florida.

This dissertation follows a collection of agentive objects around and through the networks of humans and nonhumans in four disparate works of English literature: the Anglo-Saxon poem The Dream of the Rood, William Shakespeare's narrative poem The Rape of Lucrece, Thomas Hardy's novel The Woodlanders, and Philip Pullman's trilogy His Dark Materials. While the etiology of ovarian cancer (OC) is not completely understood, evidence suggests that chronic inflammation may promote malignant transformation. However, familial history remains the strongest risk factor for developing OC and is associated with germline BRCA1 mutations, such as the 185delAG mutation. Normal human ovarian surface epithelial cells expressing the 185delAG mutant, BRAT, exhibit molecular and pathological changes that may contribute to OC oncogenesis. The physical tumor microenvironment contributes significantly to carcinogenesis, cancer progression and metastatic dissemination. Two main components of the tumor microenvironment, hypoxia and acidosis, are present in nearly every solid tumor and act as powerful selection forces against the tumor. Hypoxia and acidosis promote tumor heterogeneity and contribute to chemotherapy and radiotherapy resistance. This dissertation interrogates methods to target the tumor microenvironment including two novel studies describing mechanisms of buffer therapy resistance and targeting tumor hypoxia with vasodilators to enhance the efficacy of a hypoxia activated prodrug, TH-302.

Inlets and channels are dredged often to maintain navigation safety. It is beneficial to reintroduce the dredged material back into the littoral system, in the form of beach or nearshore nourishments. Nourishment in the nearshore is becoming an increasingly utilized method, particularly for dredged material that contains more fine sediment than the native beach. This research examines the morphologic evolution of two different nearshore nourishments. A nearshore berm was constructed at Fort Myers Beach, Florida using mixed-sized sediment dredged from a nearby channel. Through a two-phase process an instrument was created to measure the cognitive domain as proposed by King and Baxter Magolda in their Developmental Model of Intercultural Maturity (2005). The first phase involved expert panels who identified the competencies which exist in the cognitive domain, identified situations which might exist between individuals from different cultures, validated scenarios created from the identified situations, and created responses which corresponded to the three developmental levels (Initial, Intermediate, and Mature) defined in the Developmental Theory of Intercultural Maturity.

Rna Detection Technology For Applications In Marine Science: Microbes To Fish, Robert Michael Ulrich

The accurate identification of taxa from mixed assemblages using genetic analysis remains an important field of molecular biology research. The common principle behind the development of numerous documented genetic detection technologies is to exploit specific nucleotide sequences inherent to each taxon. This body of work focuses on practical applications of real-time nucleic acid sequence-based amplification (RT-NASBA) in marine science, and is presented in four case studies. Each study represents novel work in the genetic identification of respective taxa of interest using RT-NASBA.
