SERS based immunoassays for point-of-care diagnostics is a promising tool to facilitate biomarker detection for early disease diagnosis and control. The technique is based on a sandwiched system in which antigen is first captured by a selective substrate and then labeled by an extrinsic Raman label (ERL). Here, we report on the use of gold nanoparticle modified filter paper as a novel capture membrane in a vertical flow format. Gold nanoparticles (AuNPs) have been exploited in the various domains of science such as drug delivery, bio-sensing, immunoassays and environmental sensors, due to their optical properties and intriguing surface chemistry. Different scientific procedures have been used to effectively immobilize antibodies onto AuNPs. Although acceptable outcomes have been achieved in the immobilization of antibodies onto AuNPs, the sensitivity of these immobilized antibodies to target antigen or binding sites is limited due to improper orientation of the antibodies. Also, the possibility of nanoparticle aggregation when exposed to proteins limits its biomedical applicability.

Protein-gold nanoparticle (AuNP) bioconjugates have many potential applications in nanomedicine. A thorough understanding of the interaction between the protein and AuNP is critical to engineering these functional bioconjugates with desirable properties. In this work, we investigate the role of free thiols presented by the protein on the stability of the protein-AuNP conjugate. Human serum albumin (HSA) was modified with 2-iminothiolane (Traut’s reagent) to introduce additional thiols on the protein surface, and three variants of HSA were synthesized to present 1, 5, and 20 free thiols by controlling the molar excess of the chemical modifier. Widespread usage of antibiotics is a growing concern due to antibiotic resistance development in bacteria. This is due to common use of antibiotics in agricultural, livestock, and clinical usage. Antibiotic resistance is developing at a rate in which it is outpacing new drugs on the market. New strategies in drug development are necessary to combat the increasing resistance. We designed several motifs of sugar-modified nanoparticles to inhibit the biofilm formation of Pseudomonas aeruginosa.

Free ligand binding molecular dynamic simulations are a powerful tool used to probe the ligand binding process, mechanism and pathway and the insight gained can help expedite the early stages of drug discovery. Using these methods, we model the binding of two small molecule anti-cancer agents BRACO19 and CX-5461 to a variety of DNA G-quadruplexes (G4s) and a DNA Duplex. The first study focuses on the binding of BRACO19 to three different topological folds (parallel, anti-parallel and hybrid) of the human telomeric G4s. Adverse Childhood Experiences (ACEs) affect numerous outcomes in adulthood, but relatively few studies examine their implications for adolescents. Understanding the effects of ACEs is important since adolescent behaviors affect subsequent life course milestones and transitions. Deception is a tactic that is seen by individuals on a daily basis. One of the places most people endure deception is in the workplace. This thesis analyzes how leaders utilize deception in the workplace through the television show Sons of Anarchy. Throughout the show, both transformational and transactional leadership styles are present. Various story arcs portraying deceptive actions will be analyzed through the lenses of leader-member exchange theory and interpersonal deception theory. This thesis reveals that transformational leaders utilize deception more effectively, while transactional leaders use deceptive measures more frequently.

Bijels are made of non-equilibrium particle-stabilized emulsions with a bicontinuous arrangement of the constituent fluid phases. They spontaneously form through arrested spinodal decomposition in mixtures of partially miscible liquids and neutrally wetting colloidal particles. Soon after their discovery over 10 years ago, Prof. Mike Cates, Lucasian Professor of Mathematics, predicted their future use as continuously operated cross-flow reactors for chemical reactions between immiscible reactants.

Towards this goal, work in this thesis focuses on designing bijels via Solvent Transfer Induced Phase Separation (STrIPS) for microfluidic transport applications. Artificial neural networks (ANNs) are used in many types of computing applications. Traditionally, ANNs have been implemented in software, executing on CPUs and even GPUs, which capitalize on the parallelizable nature of ANNs. More recently, FPGAs have become a target platform for ANN implementations due to their relatively low cost, low power, and flexibility. Some safety-critical applications could benefit from ANNs, but these applications require a certain level of reliability. This study investigated the experiences of clients who completed a brief heart rate variability biofeedback protocol. The purposes of this study were to (1) learn about client experiences of biofeedback because almost no previous research has done so and (2) explore the potential role of common factors in biofeedback. Fifteen clients were interviewed and their data analyzed according to the methods of Consensual Qualitative Research (CQR; Hill, 2012). CQR relies on the use of group consensus to construct representations of participant experiences and categorize themes within the data. Individuals with Intellectual and Developmental Disabilities (IDD) are at an increased risk of experiencing sexual abuse. While there are evidence-based prevention programs for typically developing children, research addressing the IDD population is lacking. Research is also lacking in parent's attitudes towards sexual abuse prevention in the IDD population. Study 1 used a mixed-methods design to measure parent's perceptions of a sexual abuse prevention program. A survey method was used to gather information from parents (n=79). This study investigated the strategies academically successful college student athletes use to do well in their class work, where academic success was defined as a 3.5 GPA for 12 or more credits for the two semesters preceding the study. Data were transcriptions of individual interviews with five male and five female athletes in seven sports at an NCAA Division I university in the western United States. Independent coders analyzed the data and agreed upon themes related to challenges to academic success and strategies to meet those challenges. In this dissertation, I consider the problem of model reduction in both oscillatory and networked systems. Previously, the Manifold Boundary Approximation Method (MBAM) has been demonstrated as a data-driven tool for reducing the parametric complexity of so-called sloppy models. To be effective, MBAM requires the model manifold to have low curvature. I show that oscillatory models are characterized by model manifolds with high curvature in one or more directions. Chenopodium quinoa, a South American pseudocereal, has valuable agricultural traits such as salt tolerance and drought tolerance, and it has beneficial nutritional properties such as high protein content and a complete amino acid profile. However, problems including disease susceptibility, low harvest index, lodging, seed shattering, low heat tolerance, and saponin content plague quinoa. Genetic resources for quinoa are needed to fix these problems and make quinoa more available throughout the world. Without evolutionary theory, biology becomes a somewhat tangential assemblage of facts about living organisms, which is precisely how it is viewed by many students. Art, process, and materials offer me an escape from the sometimes-crushing realities of my personal struggle with chronic mental illness. Escape is often my primary motivator for making art. However, personal meaning and understanding sometimes come while I'm working in the studio. Sometimes the meanings of the work only become clear years later. Personal experience and experimenting with materials and processes have led me to an appreciation and awareness of entropy--the second law of thermodynamics. This cross-sectional study examines if there is a relationship between student ethnic identity and their motivation towards physical activity (PA), with a particular interest with students who self-identify as Hawaiian. Results indicate that there is a small but positive correlation between motivational indices and measures of ethnic identity. This study surveyed 301 sixth, seventh, and eighth grade students from one of the Hawaiian Islands. In this research, one of the widely used polymers, poly(vinylidene fluoride)(PVDF), was used to develop thin-film polymer energy harvesters. Dimethylformamide (DMF) and methyl ethyl ketone (MEK) were used to dissolve the PVDF powder. Four different (0%, 3%, 5%, and 7%) ZnO nanoparticle (NP) concentrations were used to enhance the electrical output of the thin-film energy harvesters. A sonication bath and additional MEK were used in dispersing the ZnO NPs to obtained uniform PVDF/ZnO NP solution. Over the past decade, policymakers have promoted marriage as a pathway to improve child outcomes in single-parent households. However, previous research on single mothers who later married in the United States has failed to examine how the structural advantages and disadvantages of race influence post-birth marriages and the advantage they may confer. I investigate how white advantage—the human- and social-capital benefits that come from being a white individual—acts as a resource distributed differently across three couple configurations. Microfluidic devices have been fabricated from a wide variety of materials and methods for many applications.
