Heart failure (HF) is increasingly recognized as a multisystem disease often linked to gut dysbiosis; however, its specific effects on gut microbial composition remain poorly understood. This study examined long-term changes in the gut microbiome in a murine HF model induced by transverse aortic constriction (TAC) and evaluated the effects of NMEG-CGRP. TAC mimics pressure overload-induced cardiac dysfunction, replicating key features of HF. CGRP, a neuropeptide with vasodilatory and cardioprotective effects, shows potential as a therapy for HF but is limited by rapid degradation. However, the energy density limits of traditional graphite anodes require the investigation of alternative materials. Silicon (Si), with its high theoretical capacity, is a promising anode material. This dissertation uses a qualitative, constructionist approach to examine the sociotechnical and cultural dynamics that shape digital sexual health-seeking practices of young adults. Guided by information practices and affordance theory, the research draws on semi-structured interviews and digital walkthroughs with 20 young adults (ages 18–26) from a convenience sample of young adults residing in South Carolina. Participants described how they use platforms like TikTok, Instagram, and Reddit not only to locate information but to assess its trustworthiness, inclusivity, and personal relevance in real time. The chemokine CCL8 (monocyte chemoattractant protein-2) is a versatile signaling molecule involved in tumor progression, immune cell recruitment, and therapeutic resistance. Through activation of receptors including CCR1, CCR2B, and CCR5, CCL8 promotes leukocyte trafficking, epithelial–mesenchymal transition, and the establishment of immunosuppressive tumor microenvironments. In breast cancer and inflammatory diseases, elevated CCL8 levels correlate with poor clinical outcomes, making it a promising therapeutic target.

Fibroblast activation protein (FAP) is abundantly expressed in cancer associated fibroblasts and contributes to extracellular matrix (ECM) remodeling. Recent studies using imaging techniques have identified increased levels of FAP in the left ventricle (LV) of patients identified as at risk for developing heart failure. However, the role of FAP in regulated LV myocardial structure and function remains unclear. This project tested two hypotheses: First, genetic ablation of FAP would modify LV structure and function and be more pronounced with aging. Second, genetic ablation of FAP would modify LV structure and function with LV pressure overload (LVPO) accompanied by ECM remodeling.

Unmanned aerial vehicles (UAVs) are increasingly used in precision agriculture, where extended autonomous operation is required for monitoring, intervention, and field management. However, achieving long-term autonomy remains challenging due to battery constraints, environmental uncertainty, and the need to balance exploration with event-driven tasks. To address these challenges, a multi-layer decision-making framework inspired by Dual Process Theory (DPT) is developed. The mitotic kinesin EG5 is essential for spindle bipolarity and accurate chromosome segregation during cell division, making it a validated anti-cancer target. Unlike conventional microtubule-targeting agents, EG5 inhibitors offer mitosis-specific action, reducing neurotoxicity. Although inhibitors targeting EG5’s allosteric site-I have been extensively studied, mechanistic insights into inhibition at site-II, and their combined effects, remain underexplored. This thesis systematically investigates the structural and dynamic mechanisms underlying EG5 function and its inhibition using an integrative computational approach.

Agrobacterium tumefaciens is a soil-dwelling plant pathogen known for transferring DNA to host plants through a type IV secretion system. Central to its virulence and cellular regulation is the LysR-type transcriptional regulator, VtlR, which controls processes including biofilm formation, motility, tumorigensis, and small RNA expression. Although VtlR’s regulatory role is established, the specific functions of its conserved cysteine residues have only recently begun to be explored. Public opinion on punitiveness is an important topic of study because it can influence political elites’ decision making when developing legislation. This study focused on the role gender ideology and anti-transgender attitudes on punitiveness while controlling for religious and political conservatism. The 2020 American National Election Study (ANES) was used to examine the association between conservatism, traditional gender roles, and anti-transgender attitudes on their influence of punitiveness. Three ordinal logistic regressions were run for each type of punitiveness (i.e., death penalty support, crime spending, and felon disenfranchisement). Cyclic diguanylate monophosphate (c-di-GMP) is a second messenger that is responsible for attachment and motility in many prokaryotic systems and controls the signaling pathway for the motile to sessile transition in many bacteria. Many c-di-GMP-metabolizing proteins are diguanylate cyclases (DGCs), responsible for the synthesis of c-di-GMP and characterized by a c-di-GMP synthesizing domain, e.g., GGDEF domain. Agrobacterium tumefaciens is a gram-negative alphaproteobacterium that is one of the causative agents of crown gall tumors on plants. Its ability to transform plants using its T-DNA has been studied to improve plant-mediated transformation. sRNAs are small regulatory RNAs that play a role in gene expression in a variety of organisms and bind to target proteins or mRNAs. A. tumefaciens contains two sibling sRNAs, called AbcR1 and AbcR2, which are conserved among alphaproteobacteria. Hypersonic vehicle design requires understanding complex aerodynamic phenomena across the full flight regime. This study presents a novel MF surrogate modeling methodology that enables the prediction the full field response across a vehicle’s surface. A Space-Filling Curve (SFC) is used to convert unstructured data into 1D vectors. The a Convolutional Autoencoder is used with transfer learning to reduce the dimensionality of the data. An Emulator-Embedded Neural Network (E2NN) combines multi-fidelity data for fast, accurate predictions. A benchmark analytical example and hypersonic application are used to evaluate the methodology. Acute Lymphoblastic Leukemia (ALL) is a hematologic malignancy that primarily affects children and young adults. While advancements in treatment—including chemotherapy and cranial irradiation—have significantly improved 5-year survival rates in children to nearly 90%, radiation-induced cognitive impairment (RICI) remains a persistent and debilitating side effect. The brain, particularly the hippocampus, is highly susceptible to radiation-induced oxidative stress and inflammation. Fusing a variant of the sterile alpha motif domain of the human translocation ETS leukemia protein (TELSAM) to a protein of interest significantly enhances crystallization and diffraction quality. TELSAM is a pH-dependent, polymer-forming crystallization chaperone that, when covalently fused to a protein, forms a well-ordered crystal lattice. However, the success of this method depends on two factors: 1) the design of the linker connecting TELSAM to the target protein, which influences both crystal packing and target orientation, and 2) the prevention of TELSAM polymer flipping, a phenomenon that disrupts lattice periodicity and impairs structure determination. Reservoir sediment deposition poses a challenge to sustainable water supplies around the world. Sediment management has proven to be an expensive endeavor. Two models have been developed to guide economic analysis for reservoir sediment management, namely the Reservoir Sediment Economics Model (RSEM) and the Reservoir Conservation Model 2 (RESCON 2). While these are helpful tools for benefit-cost prefeasibility studies, they rely heavily on sometimes elusive estimations of the benefits of water supply. Satellite-Based Identification Of Mesoscale And Submesoscale Eddies And Fronts In The Gulf Of Mexico, Ethan Cruz

Mesoscale and submesoscale processes in the Gulf of Mexico (GoM) are fundamental in shaping the region’s physical and biogeochemical ocean dynamics. Advances in high-resolution satellite observations now allow detailed detection of submesoscale (< 25 km radius) and mesoscale eddies and ocean fronts, which drive heat, nutrient, and carbon fluxes. This dissertation integrates two complementary investigations: (1) a comparison of eddy-tracking algorithms: Contour Tracing (CT) and Temperature Thresholding (TT), applied to Optimum Interpolation Sea Surface Temperature (OISST, 1/4°) and Operational Sea Surface Temperature and Ice Analysis (OSTIA, 1/20°) products; and (2) a comparative evaluation of three ocean front detection algorithms: Canny Edge Detection (Canny), the Cayula–Cornillon Algorithm (CCA), and the Belkin–O’Reilly Algorithm (BOA), across multiple satellite-derived parameters, including sea surface temperature (SST), sea surface salinity (SSS), chlorophyll-a (Chl-a), and altimetry-derived absolute dynamic topography (ADT). The eddy-tracking analysis shows that the higher-resolution OSTIA consistently detected more eddies than OISST, with TT performing best during winter when SST gradients were strongest. The front detection analysis, incorporating datasets from the Surface Water and Ocean Topography (SWOT) and Plankton, Aerosol, Cloud, Ocean Ecosystem (PACE) missions, was evaluated under diverse conditions, including flood and drought periods, Loop Current phase changes, and extreme weather events. A machine learning–based Gaussian Mixture Modeling (GMM) approach was also developed, providing a novel framework for dynamic front detection. By benchmarking traditional and machine learning methods, this integrated study identifies optimal algorithm–dataset combinations for resolving submesoscale and mesoscale features in the GoM, with applicability to other regions.

Multicultural literature was used to conduct interactive read-alouds to first-grade students. A look across all data sources suggested three main themes that occur throughout the research study. These themes included student engagement and questioning, making connections by experiencing culture through an authentic lens, and developing cultural awareness and empathy for those who are different. A highly selective Magnetic Molecularly Imprinted Polymer (MMIP) was developed for the detection and purification of Quercetin. The Density Functional Theory was utilized for the selection of suitable monomers based on binding energy interaction between quercetin and monomers (Acrylamide, Acrylonitrile, Methacrylic Acid). From computational studies, Acrylamide was selected as a suitable monomer for the synthesis of MMIP. Under ultrasound irradiation both quercetin and naringin imprinted materials MMIP-Q and MMIP-N were fabricated.

Long segmental defects in peripheral nerves impair sensory and motor function by disrupting neural impulse transmission. End-to-end autologous grafting with proper fascicular complementation has demonstrated functional recovery per the Medical Research Council Classification (MRCC). However, obtaining donor nerve tissue exceeding 30 mm remains a significant challenge. Artificial hollow conduits serve as an alternative but are limited to defects smaller than 30 mm due to inadequate innervation across the lumen.

Tissue-engineered three-dimensional (3D) myocardial patches offer superior and innovative treatment strategy for myocardial infarction (MI). Among the various scaffold fabrication techniques, electrospinning and 3D bioprinting have received great attention due to their ability to recreate native cardiac tissue mimetic characteristics. Despite several advantages, electrospun nanofibers have limitations in cell delivery due to random cell seeding, while bioprinting faces challenges in creating hydrogel constructs with ideal mechanical properties capable of supporting cardiac contraction/relaxation cycle. Hence in this work, hybrid myocardial patches were developed by converging electrospinning and bioprinting techniques.
