Streptococcus sobrinus (SSO) is a pathogenic microbe native to the human oral microbiome. SSO is historically implicated in carie formation, and may be involved in neurodegenerative disease progression. The study of SSO is challenged by a lack of access to a serotyping assay. This study aims to identify sero-specific candidates from available SSO genomes using sequence alignment and phylogenetic analysis. Three candidate genes were found that may be sero-specific: putative quinol monoxygenase, a hypothetical protein, and elongation factor Tu. Phages, as viruses that specifically target and eliminate bacteria, present as a promising treatment strategy against antibiotic-resistant bacteria. Giant phages, however, with longer genomes and a higher level of virion complexity, remain understudied. This research aims to evaluate the applicability of data-dependent acquisition mass spectrometry (DDA-MS) versus data-independent acquisition mass spectrometry (DIA-MS) to characterize and quantify the copy numbers of virion proteins of the giant Salmonella phage SPN3US.  Accurate copy number estimations require confirmation of protein sequences in the mature virion, as previous analyses showed nine SPN3US head proteins were shorter than their predicted lengths, as they had been cleaved …

The increasing incidence of screen time of all ages has generated a greater interest in the effect of screens on the human psyche. This paper looks at behavioural data of 2, 000 individuals aged 13-64 to investigate the predictability of a composite mental- health score by various modalities of screen-time, lifestyle and psychological charac- teristics. This thesis investigates the potential of moisture pathways and cracks in heritage buildings, long regarded as structural weaknesses, as advantageous passive design elements. The study addresses the issue of improving the energy efficiency of heritage buildings, particularly in tropical monsoon climates with high humidity and fluctuating temperatures. With an empirical experiment in an environmental chamber using a prototype wall, the study assesses the effect of artificially created cracks on indoor temperature and humidity control. Using rigorous data preparation methods including imputation, feature transformation, and exploratory statistical analysis the study applies multiple regression models (Decision Tree, Random Forest, Gradient Boosting, and MLP) to replicate the GTMI structure and identify its primary drivers. This work reframes Gavi’s funding decisions as a multi-objective portfolio optimization problem that jointly considers health impact, equity, cost-effectiveness, sustainability, and epidemiological interdependence.
