In the first part of this study, two novel c&barbelow;old s&barbelow;hock domain p&barbelow;roteins from rice (OsCSP) were cloned and subsequently characterized their roles during stress conditions and development. OsCSP1 and OsCSP2 ( Oryza sativa CSD protein) encode putative proteins consisting of an N-terminal CSD and glycine-rich regions that are interspersed by 4 and 2 CX2CX4HX4C (CCHC) retroviral-like zinc fingers, respectively. Using an in vitro DNA binding assay, I demonstrate that OsCSPs exhibit conserved ssDNA binding activity. In vivo functional complementation in a cold-sensitive bacterial strain, that lacks four cold inducible cold shock domain proteins revealed that OsCSPs function as RNA …

Prostaglandin F2 alpha (PGF2alpha) brings about regression of the bovine corpus luteum (CL). This luteolytic property of PGF 2alpha is used in beef and dairy cattle to synchronize estrus. A limitation of this protocol is an insensitivity of the early CL to luteolytic actions of PGF2alpha. The mechanisms underlying this differential luteal sensitivity are poorly understood. Therefore the main objective of the current study is to understand the cellular mechanism of luteal insensitivity. Cadmium (Cd) is both an environmental pollutant as well as a component of cigarette smoke. Recent data suggests increased cancer risks and increased mortality in environmentally exposed populations. Although evidence demonstrates that adult exposure to Cd causes changes in the immune system, there are limited reports in the literature of immunomodulatory effects of prenatal exposure to Cd. The sonic hedgehog (Shh) and Wnt/ss-catenin pathways are required for thymocyte maturation. Several studies have demonstrated that Cd exposure affects these pathways in different organ systems. Between the Wheels examines three Carolina Piedmont streetcar strikes in 1919-22. These years were marked by the aspirations of workers for industrial democracy, corporate anti-labor backlash and by the first Red Scare. Inevitably, these trends swept through the Carolina Piedmont, long viewed as isolated and resistant toward progress. But scholars should now re-examine the New South in light of broader American context. Three case studies in Spartanburg and Columbia, South Carolina, and Charlotte, North Carolina, highlight the struggles of New South labor reformers against union-busting monopolists, exemplified by James B. Protein wasted by the disposal of fish processing byproducts may be recovered using isoelectric solubilization and precipitation. The protein is dissolved by extreme pH shifts and recovered via precipitation and centrifugation. Microbial safety throughout this process had not yet been evaluated; therefore, the purpose of this study was to determine if Listeria innocua and Escherichia coli would survive extreme pH shifts during the protein recovery process. The main objective of this study is to develop a novel network-based methodology to identify prognostic signatures of genes that can predict recurrence in cancer. Feature selection algorithms were used widely for the identification of gene signatures in genome-wide association studies. But most of them do not discover the causal relationships between the features and need to compromise between accuracy and complexity. Mindfulness is a trained or dispositional state of "being" that has been linked to positive mental and physical health effects. Although preliminary findings on cortical activation have shown mindfulness to be associated with increased frontal activation and reduced limbic activity, very little is known regarding the influence of mindfulness on autonomic nervous system activity. Differences between younger (N=78) and older (N=78) adults' frequency of experience of interpersonal problems were examined. Previous research has shown age-related declines in contact frequency and negative interactions (Akiyama, et al., 2003; Carstensen, 1992). Aging has also been associated with concurrent increases in emotional closeness and relationship satisfaction with social partners (Birditt, et al., 2005; Lang & Carstensen, 2002), and social self-efficacy and problem-solving effectiveness (Artistico, Cervone, and Pezzuti, 2003; Blanchard-Fields, Mienaltowski, & Seay, 2007; Lachman, 1986). Health promoting behaviors are typically categorized as behaviors that move individuals towards optimal health while concurrently decreasing one's susceptibility to disease and illness (Becker & Arnold, 2004; Breslow, 1999). When compared to other developmental age periods, less is known about health promotion in midlife (Merrill & Verbrugge, 1999). Thus, the present study examined the relations among demographic variables, future time perspective, internal health locus of control, and middle-aged adults' engagement in health promotion. In October 2008 West Virginia University Division of Theatre and Dance presented Christopher Cartmill‟s The Legend of Sleepy Hollow in the Lyell B. Clay Concert Theatre in Morgantown, WV. This thesis details the scenic design process including script analysis, research, planning, construction and an analysis of the final design. A series of models and discussions took the first impressions of the script to the final version which incorporated some of the initial concepts but left others behind for better or for worse.

Subsurface defects in wooden logs such as knots, decays, splits, embedded metallic nails and bullets are of major concern to timber saw mills. Presence of these defects decreases the value of the sawn lumber boards. Also, the factory down time and operation cost increases significantly whenever the saw blade is damaged by encountering embedded metals during the sawing process. This study has been conducted to assess the possibility of detecting subsurface defects in logs using Ground Penetrating Radar (GPR) before the sawing process. An effective use of Nd:YAG lasers in remote, in-situ sensing and other applications require compact size and efficient operation. Our prior work demonstrated that a functioning Nd:YAG laser resonator could be reduced to the size of approximately 1cm2 (10mmx12mm) and 2mm thickness with an output power of 2.2W and an efficiency of 6%. Enhancing the efficiency and output energy and further reducing the size of these slab lasers will make them attractive candidates for compact laser sources in applications such as Laser Induced Breakdown Spectroscopy (LIBS), laser-based spark plug systems, and Raman excitation-based integrated sensor systems. Carboniferous plant fossils have been reported from the Appalachian Basin since the middle part of the nineteenth century. The lack of easy access to European literature and comparative specimens resulted in a body of work characterized by overly diagrammatic figures and confusing lists of junior synonyms with limited figures. This report provides 41 photographic plates of Carboniferous plant fossils from the Appalachian Basin. Comparative range charts are used to summarize the similarity of the fossil macrofloras of the Appalachian region with those of other parts of Euramerica, although each paleoregion has a small number of endemic species. With the reduction of solid oxide fuel cell (SOFC) operation temperature from 1000°C to 800°C, it is possible to use stainless steel as interconnect. Chromia forming ferritic stainless steels are the most acceptable materials for interconnects application because of their cost effectiveness and good oxidation resistance, as compared to other candidates. However, excessive scale growth and chromium evaporation will degrade the cell performance rapidly. Highly conductive coatings that aide oxidation resistance and prevent chromium evaporation may prevent the problems. Social anxiety is experienced across the lifespan, with many individuals reporting symptoms of anxiety and shyness from a very early age in childhood. A number of measures have been designed to assess social anxiety in children, adolescents, and adults, yet little attention has been focused on children below 8 years of age. To better understand the American experience of the Great War, one must look at it through the lens of the American Civil War. The surge of voluntarism seen in the Preparedness Movement, especially the formation of elite volunteer units, could trace its roots to the mid-nineteenth century. The ubiquitin/26S proteasome system (UPS) is conserved in all eukaryotic species. The 26S proteasome serves as the protease in this pathway and is responsible for the degradation of abnormal and short-lived proteins after their modification with a polyubiquitin tag. It consists of one or two ATP-dependent, 19S regulatory particles and an ATP-independent 20S core protease. PAG1 serves as one of at least 14 different subunits of the 20S core protease. Study I examined the effects of aging and reactive oxygen species on flow-mediated vasodilation in coronary arterioles. Isolated coronary arterioles of young (4 mo) and aged (24 mo) male Fisher 344 rats were subjected to stepwise increases in flow (0–50 nL/sec) in the presence and absence of superoxide dismutase (SOD) mimetic Tempol (100 µM), hydrogen peroxide (H2 O2) scavenger catalase (100 U/mL), Tempol + catalase, or Tempol + deferoxamine (100 µM), an iron chelator and inhibitor of hydroxyl radical formation. Fire is an inherently spatial process that is tightly coupled to spatial patterns in climate and topography. This is particularly true in the dry western United States, where confounded by the legacy of fire suppression and the trend in global warming, large high severity fires are increasingly frequent. Recent work has integrated space and time analyses of fire regimes through the application of geographic information science (GIS). GIS was developed for spatial studies, and it can be effective in multi-dimensional interpretations of fire ecology in many vegetated ecosystems. Computer simulation is an appealing approach for the reliability analysis of structure-based software systems as it can accommodate complexities present in realistic systems. When the system is complex, a screening experiment to quickly identify important factors (components) can significantly improve efficiency of the analysis. The challenge is to guarantee the correctness of the screening results with stochastic simulation responses. Control Sequential Bifurcation (CSB) is a new method for factors screening using simulation experiments, when only main effects models are considered. West Virginia University (WVU) currently has a Transportable Emissions Laboratory capable of measuring the exhaust constituents produced by a heavy duty diesel engine. The laboratory has the capability of measuring the exhaust constituents from a partial exhaust stream of a locomotive diesel engine. As the field of locomotive engine emissions testing broadens, it has become desirable for WVU to develop a laboratory that is capable of sampling the gaseous exhaust constituents produced by a locomotive engine from a raw, undiluted gas stream, in tandem with a partial flow dilution tunnel system for particulate matter. Due to the rising power costs and lack of nonrenewable energy sources, the cooling of houses is becoming more expensive. Looking for alternative methods applicable to this process is becoming not only an option, but also a necessity. Changes in the roof structure of buildings can be applied in order to achieve a more favorable thermal transmission behavior. The utilization of a tilted roof, composed of two separated surfaces, generates natural convection currents in the channel between them.
