Rod photoreceptors are specialized neurons located in the retina, the neural tissue that lines the back of the eye. Rod photoreceptors are necessary for dim light and peripheral vision through the photoactivation rhodopsin (Rho). Rho is synthesized in the inner segment of rods before being unidirectionally trafficked to the outer segment. In diseases such as retinitis pigmentosa (RP), Rho trafficking can be disrupted, leading to progressive rod cell death. In many cases of RP, Rho is mislocalized to the rod photoreceptor synapse (“spherule”). The impact of Rho mislocalization on spherule structure and protein organization is poorly understood. The propensity to attribute incentive salience to reward-related cues, or sign-tracking (ST) has been linked to addiction in animal models and some initial human data. The current study explores the role of sign-tracking (ST), as a potential risk factor for problematic slot machine gambling, which abundantly features audiovisual reward-related cues, henceforth sensory features. A sample of healthy participants without gambling problems (n = 30) completed a Pavlovian-to-Instrumental Transfer (PIT) paradigm to assess ST propensity and played on a realistic slot machine simulator consisting of two versions: with sensory features (SF+) and without (SF-). This thesis investigates the West Virginia Human Rights Commission and its action surrounding the Civil Rights Movement. The Commission represented the liberal ideals that led white moderates to be potent allies of the Civil Rights Movement. Throughout the years following its inception, the Commission investigated and documented many instances of Jim Crow that were crushing Black communities in West Virginia. However, the limits of its liberal approach prevented it from creating solutions that would address the power structures that supported discrimination in West Virginia. Situated in the Appalachian Mountains, one of the oldest mountain ranges on Earth, West Virginia waters boast rich ichthyofauna including native sportfish species walleye (Sander vitreus), largemouth bass (Micropterus nigricans), and muskellunge (Esox masquinongy). These three species are all native to the contemporary Ohio River watershed and play a major recreational and ecological role in local fisheries. Storytelling is a primary form of communication between grandparents and grandchildren that has been shown to strengthen relational bonds and enhance grandchildren’s well-being. However, the specific narrative features and interactional processes involved in grandparent-grandchild storytelling remain underexplored, particularly regarding how they influence well-being and perceptions of generational differences. Guided by Communicated Narrative Sensemaking (CNSM) theory, this study examined the content (story framing) and process (interactional sensemaking behaviors) of grandparent-grandchild storytelling episodes as reported by young adult grandchildren (N = 110). By combining cultural, memory, and material culture studies, this dissertation examines the ways objects associated with George Washington became instruments within the proxy war over whether the United States or the Confederacy best represented the American Revolutionary era’s principal values of liberty and equality. Exploring monuments, statues, historic homes, relics, and mementoes, as well as antebellum speeches and schoolbooks, letters, diaries, military orders, newspapers, and memoirs, this work demonstrates how the relics linked to Washington and the nation’s founding shaped how the Civil War generation thought about America’s past.

Tsetse flies (Diptera: Glossinidae) are the vector of trypanosomes (Trypanosoma spp.), which are responsible for African trypanosomiasis, also known as sleeping sickness. African trypanosomiasis is a significant health and economic burden yet remains a neglected tropical disease. As an obligate blood feeder, tsetse flies are reliant on B vitamin supplementation from intracellular Gammaproteobacteria, Wigglesworthia glossinidia. Folate (B9) is essential for tsetse development and reproduction, as well as DNA methylation in animals. This research presents the development of a Mixed Reality (MR) robotic framework that integrates real and virtual environments for experimental robotics, education, and visualization. The proposed system combines a motion-capture system, a simulator, and a projector to create a spatially aligned, interactive workspace where physical robots coexist with projected virtual objects. The ability for beef cattle to utilize feed efficiently is a critical determinant for both economic and environmental sustainability in livestock production. However, despite extensive research, the complex microbial ecosystem within the rumen and its relationship to feed efficiency remain incompletely understood. Virtual flight test aircraft provide good opportunities to test new parameter estimation methods. However, good estimates for the parameters in VIRTTAC Castor, a virtual test aircraft, are unavailable. This thesis uses two well-established time domain methods, the Output Error Method and the Filter Error Method, to estimate the parameters of VIRTTAC Castor. Three standard maneuvers were used: Elevator 3-2-1-1, Rudder Doublet, and Aileron Multi-Step. The parameters were estimated across three different flight conditions: weight, altitude and air speed, which resulted in 27 tests for each maneuver. All parameters, and standard deviations estimated are included in the appendix as a table.

The primary focus of this research is the modeling and validation of crankcase methane emissions stemming from natural gas combustion engines commonly deployed in the natural gas compression industry. The models employed rely on equations predicting the flow rate and composition of losses, typically generated within reciprocating piston-cylinder systems. Since crankcase emissions originate in a piston–cylinder arrangement, multiple variables were considered in this study, including displaced volume, pressure, temperature, flow, and viscosity (with corrections applied to account for the thermal and compositional effects on the mixture of fluids). Endodontic treatment aims to prevent or address apical periodontitis by reducing intracanal bacterial populations to promote tissue healing, requiring thorough debridement and disinfection of the canal space. This study aims to examine the presence of trace metal elements and chemical compounds in over-the-counter (OTC) and medical-grade sodium hypochlorite (NaOCl) solutions.

Method

Over-the-counter NaOCl brands (Clorox® and Great Value®) and medical grade 6% NaOCl solutions (CanalPro® and Chlor-Xtra®) were purchased, stored in amber glass bottles, and tested in triplicate. Building on the foundations of intergroup contact theory (Allport, 1954), the parasocial contact hypothesis (Schiappa et al., 2005), and Harwood’s Contact Space (2010), this dissertation advances the existing literature on how intergenerational contact with media characters can improve young adults’ willingness to engage with older adults. Specifically, this study examines the effects of intergroup contact quality on the parasocial contact process. The family Nectriaceae includes numerous phytopathogenic fungi that cause canker diseases on both angiosperm and conifer hosts worldwide. Numerous Neonectria spp. cause cankers on hardwoods and conifers in North America, but their roles in contributing to tree decline and mortality outside of beech bark disease are largely understudied. One such pathogen, Neonectria magnoliae, causes perennial cankers on two native hosts in central Appalachia: Fraser magnolia (Magnolia fraseri) and tulip-poplar (Liriodendron tulipifera). Resurgence is a type of relapse that consists of the recurrence of a previously eliminated response following worsening reinforcement conditions for an alternative response. Resurgence can occur following a history of positive or negative reinforcement; however, no previous evaluations have directly compared resurgence following these processes. In the present set of experiments, college students responded on a computer program to earn points (positive reinforcement) and to avoid losing points (negative reinforcement). A transition away from fossil fuel use is necessary for the future of energy production, and biofuels are a renewable energy option that can mitigate the impacts of fossil fuels through lowered carbon emissions and possible carbon sequestration. In order to successfully compete with fossil fuels, the production of biofuels must be sustainable and cost effective. Populus species are a biofuel crop used for bioenergy conversion from lignocellulosic biomass that have been proposed for use in feedstock plantations on marginal lands, which would avoid competition with agricultural production.
