Investigations Into Indole Formation: Base-Mediated Formation Of N-Hydroxy- And N-Alkoxyindoles, Ancillary Ligand Development For Watanabe-Cenini-Söderberg Cyclization, And Total Synthesis Of Racemosin B, Blaine Thomas Mcclay

Graduate Theses, Dissertations, and Problem Reports (ETD)

A novel, base-mediated cyclization was developed to form N-hydroxy- and N-alkoxyindoles from 2-(2-nitrophenyl)butenoates. Potassium tert-butoxide or sodium tert-pentoxide, with or without the in-situ addition of an electrophile, formed a multitude of N-hydroxy- and N-alkoxyindoles. The formation of these butanoate precursors was improved via a Barluenga coupling reaction. Novel 4,5,6,7-tetramethoxy-1,10-phenanthroline was utilized as an ancillary ligand within the Watanabe-Cenini-Söderberg reductive cyclization reaction conditions. These new reaction conditions produced high yields for sterically hindered 2-nitrostyrenes and the first synthesis of dilemmaone A using this reductive cyclization. Racemosin B was synthesized in a three-step process utilizing a …

Learning Representations For Novelty And Anomaly Detection, Ranya Almohsen Jan 2023

Learning Representations For Novelty And Anomaly Detection, Ranya Almohsen

Graduate Theses, Dissertations, and Problem Reports (ETD)

The problem of novelty or anomaly detection refers to the ability to automatically

identify data samples that differ from a notion of normality. Techniques

that address this problem are necessary in many applications, like in medical

diagnosis, autonomous driving, fraud detection, or cyber-attack detection, just to

mention a few. The problem is inherently challenging because of the openness of

the space of distributions that characterize novelty or outlier data points. This is

often matched with the inability to adequately represent such distributions due

to the lack of representative data.

In this dissertation we address the challenge above by making several …

Burkholderia Collagen-Like Protein 8, Bucl8, Is An Outer-Membrane Component Of A Novel Efflux Pump And Immunogenic Vaccine Candidate, Megan Elizabeth Grund

Graduate Theses, Dissertations, and Problem Reports (ETD)

Burkholderia spp. are Gram‐negative intrinsically resistant bacteria that are responsible for environmental and nosocomial infections of clinical and biodefense concern. Our laboratory previously identified in silico Burkholderia collagen-like protein 8 (Bucl8) in the BSL-3 pathogens Burkholderia pseudomallei and Burkholderia mallei (Figure 1). The protein sequence of Bucl8 contains tandem outer-membrane efflux protein domains (OEP) typical of efflux systems and a unique collagen-like domain (CL) consisting of a repeating sequence of Glycine-Alanine-Serine (GAS). Because efflux pumps are often a mechanism of pathogenicity and antibiotic resistance, we investigated Bucl8 further. Here, we determined that the locus of bucl8 contains genes …

Graduate Theses, Dissertations, and Problem Reports (ETD)

Early embryonic loss is a significant factor in livestock species' infertility, resulting in an economic deficit. In cattle, the in vivo fertilization rate is ~90%, with an average calving rate of about 55%, indicating an embryonic-fetal mortality rate of roughly 35%. Further, 70-80% of total embryonic loss in cattle occurs during the first three weeks after insemination, particularly between days 7-16. Growing evidence indicates that the oocyte plays an active role in regulating critical aspects of the reproductive process required for successful fertilization, embryo development, and pregnancy. However, defining oocyte quality remains enigmatic. Recently, many have abandoned the notion that …

Structural Health Monitoring Using Machine Learning And Synthetic Data, Michail Tzimas Jan 2023

Structural Health Monitoring Using Machine Learning And Synthetic Data, Michail Tzimas

Graduate Theses, Dissertations, and Problem Reports (ETD)

Structural health monitoring spans many decades of research across multiple engineering fields. However, typical monitoring processes for damage detection of complex structures usually prohibit real-time or fast detection of debilitating damage to the structure. One of the major issues of real-time detection of damage is the enormity of data that needs to be processed, which is worsened by the relative inability of fast relaying of data to structural engineers. With the rapid advancement of Machine Learning, both issues can be overcome, and detection of failure is achieved with non-invasive techniques. This dissertation explores the applicability of Machine Learning as a …

Graduate Theses, Dissertations, and Problem Reports (ETD)

The objective of this dissertation is to investigate how class and stigma influence spatial inequalities in health across the US, from the structural to the individual level. Class, stigma, and subsequent access to capital resources are not equally distributed across the US. Women, poor, and minority populations continue to have unequal access to capital resources across the country, though this is spatially determined. Similarly, while there are health inequalities along the same social cleavages at the national level, they differ significantly across localities. Research has not paid enough attention to the fundamental social causes of inequities, resulting in the inability …

The Effect Of Silver Carp On Large River Community Size Spectra, Bradley R. Novak

Graduate Theses, Dissertations, and Problem Reports (ETD)

The introduction of silver carp (Hypophthalmichthys molitrix) has had dramatic effects on native fish communities. Silver carp are large bodied planktivores and compete with native planktivores which can cause cascading effects throughout the food web. Our study strives to determine the threshold of abundance beyond which silver carp alter fish community structure. We used a community size spectra approach to evaluate fish community structure and compare size spectra across spatial and temporal gradients of silver carp abundances. Community size spectra (CSS) is an ataxic approach quantifying the predictable decrease in fish abundance with increasing body size that reacts …
