Retroviruses exhibit high mutation rates. High mutation and recombination rates increase variation within a viral population and result in production of drug-resistant mutants and/or mutants that can escape a host immune response. Mutations are introduced into the viral genome by error-prone reverse transcriptase (RT), a virally encoded enzyme that converts single-stranded viral RNA into double-stranded DNA. In the current study we are trying to understand what structural determinants of RTs are important for fidelity, frequency of template switching, and drug-resistance. The present study examined parental influences on educational and career goals of high school students. Data on educational and career goals from the Perceived Life Chances Interview were derived from 124 rural adolescents. One hundred of their mothers and 36 of their fathers completed the Inventory of Parental Influence which consists of five subscales of Parental Involvement/Help, Psychological Support, Parental Pressure, Pressure for Intellectual Development, and Time Management/Monitoring. Responses from the Perceived Life Chances Interview were qualitatively analyzed in order to derive categories that reflected the varying responses from participants. Deep River and Muddy Creek flow across a granitic and volcanic terrain intruded by diabase dikes in the Piedmont physiographic province of central North Carolina. Geoarchaeological and sedimentological investigation of these Piedmont streams included fourteen trenches dug at four floodplain localities. Soils in these trenches were described using standard soil taxonomy and sampled for particle-size, magnetic susceptibility, and mineralogical analyses.;Increased upland erosion rates, after European settlement, left more than a meter of historic sediments in some areas of the floodplains. A comprehensive ecological survey was conducted from April 1997 to June 1999 on four turkey flocks (F5-F8) to identify the preharvest sources of Salmonella colonization. Generic E. coli and total coliforms were enumerated as indicators of fecal contamination in F7 and F8. Turkey cecal and crop contents, litter, drinkers, air, feed, feeder contents, and environmental swabs were sampled and tested for Salmonella and E. coli. Salmonella was isolated from 13% of litter, 11% of turkey ceca, 10% of drinkers, and 5% of swabs. Escherichia coli and total coliforms were detected in 45 and 53% of samples, respectively. The prediction of permeability is a critical, key step for reservoir modeling and management of oil recovery operations. Previous studies have successfully demonstrated that the new technology called Artificial Neural Network (ANN), a biologically inspired, massive parallel, distributed information processing system, is an excellent tool for permeability predictions using well log data. This technology overcomes the drawbacks caused by the inherent heterogeneity of the reservoir and lack of sufficient cores or pressure transient tests, allowing to define reservoir characterization within an acceptable accuracy while maintaining costs low. Interleukin-10 (IL-10) is an anti-inflammatory cytokine that plays an important regulatory role in inflammatory diseases. The overall goal of this research is to investigate the potential therapeutic effect of IL-10 gene therapy for the treatment of pulmonary inflammation and to develop appropriate gene delivery and expression systems for efficient gene transfer to the lung. We evaluated various liposomal and non-liposomal agents including LipofectAMINE RTM, LipofectinRTM, DOTAP, DEAE-dextran, and the DNA condensing agent protamine sulfate for their ability to promote gene transfection in macrophages. In 1994 and 1995, Cryphonectria parasitica isolates singly or doubly infected with hypoviruses in the CHV3 genus, and lacking distinctive morphological abnormalities, were recovered near plots formerly used to release hypoviruses. This study tested the hypothesis that North American hypoviruses are evolving decreased virulence, and investigated hypovirus interactions affecting virulence in double-infections. Families of single conidial progeny (SCP), differing in infection status, and derived from six recovered, three release, and three comparison isolates were tested in three field experiments. All release isolates and four recovered isolates were debilitated by their CHV3-gh2-type hypoviruses. CHV3-sr2-type hypoviruses never caused debilitation. In family SG3-2 …

Currently, there is some controversy concerning whether praise affects compliance and behavior other than compliance (i.e., general behavior) in children with disruptive behaviors. It is unclear whether it is praise or other aspects of general attention, such as descriptions and enthusiasm, that influences children's behavior. The current study examined the effects of enthusiastic praise, non-enthusiastic praise, and non-enthusiastic description on child compliance (measured by the Compliance Test) and general behavior (measured by the Marble-in-the-Hole Game) in 15 children with disruptive behavior problems and 15 typical children. The methanotrophic bacterium, Methylococcus capsulatus, is capable of the conversion of methane to methanol at ambient temperature and pressure. In this manuscript, we show that quinols can provide reducing equivalents to the membrane-bound form of methane monooxygenase (pMMO), substituting for NADH in whole cells and membrane fractions. Additionally, quinols are shown to be effective reductants for the detergent-solubilized enzyme, whereas NADH is ineffective. The decyl-analog of plastoquinol and duroquinol provide the greatest MMO activity in whole cells and membrane fractions, as well as detergent-solubilized samples. Lauryl maltoside is by far the best detergent for solubilization of catalytically-active methane monooxygenase. This …

The knowledge of dispersal biology and species diversity of eriophyoids is very limited, because their tiny body size makes observation and study difficult.;The current method of using sticky glass slides or plates to sample airborne eriophyoids is inefficient and inappropriate. The purpose of this study was to determine the impact of prenatal care utilization and maternal dietary intake on pregnancy outcome among West Virginia WIC participants. The sample size (102--155) used in this study varied, due to the number of subjects that provided complete information for specific variables. Data were obtained from survey questionnaires, participants' medical records and food frequency questionnaires. An allergic reaction is related to corrosion of the alloys and subsequent leaching of nickel ions into the oral cavity. Regional declines in Allegheny woodrat (Neotoma magister ) populations suggest a better understanding of the ecology of this species is needed for proper management. I described seasonal food habits of the Allegheny woodrat in the Ridge and Valley and Allegheny Plateau physiographic provinces of West Virginia and Virginia using microhistological techniques. In the Allegheny Plateau, the top 5 food items were fungi, blueberries ( Vaccinium spp.), acorns (Quercus spp.), ferns ( Dryopteris spp.), and lichens. I examined productivity, chick mortality, and factors influencing ruffed grouse (Bonasa umbellus) chick survival in West Virginia during 1998--1999. The proportion of hens that attempted to nest, hen success, average clutch size, and hatch dates were similar between age classes, between sites, and between years. Hatch dates were similar between age classes and between sites. Depredation was the major cause of nest failure. I examined 2 types of radio transmitter attachment methods (glue-on and collar type) to determine mortality and survival in ≤3-day-old grouse chicks. Glue-on transmitters performed poorly, whereas collar types had excellent retention time. Chick survival was low …

The primary objective of the present study was to examine associations among several family relationships and adolescent social anxiety utilizing a family systems perspective. The second objective was to investigate specific family relationships and their association with adolescent adjustment outcomes such as depression, general anxiety, global self-worth, and social acceptance self-worth. Mathematics education has long been in need of improved methods of instruction, particularly in the area of problem-solving skills. This study compared three methods of training rules about laws of exponents and order of operations. All three training methods used the same mastery criterion for training each rule and included the same number of practice trials during review sessions that preceded each test. We placed leaf packs containing different proportions of sugar maple and red oak leaves into a headwater stream before and after a winter spate to determine the effects of leaf species and flood disturbance on leaf pack processing and macroivertebrate colonization patterns. We also collected natural leaf packs from three streams to examine how leaf pack quality influences macroinvertebrate colonization. Red oak leaves processed faster in association with maple leaves. Invertebrate densities were higher on leaf packs containing more fast-processing leaves. The purpose of this study was to investigate the antimicrobial effect of calcium hydroxide paste and calcium hydroxide containing gutta percha ìPlusî points. Confocal scanning microscopy created a more sensitive model for assessing endodontic antimicrobial activity. The model consisted of anterior teeth with canals prepared to a #60 K Flex file. Target organisms (A) Enterococcus faecalis or a combination of (A) Enterococcus faecalis and (B) Bacteroides fragilis were inoculated into the prepared site. Despite a deluge of national attention to equity issues, public school administration is still the domain of men. Research attention to women in leadership has done little to close the gender gap in superintendents' and secondary school principals' offices, perhaps because of its focus on successful men rather than on the women who hold these positions. Qualitative interviews examine seven central Appalachian women school leaders in light of feminist standpoint theory, focusing on the way the women enter leadership, articulate a personal construction of the leadership role, and negotiate the borders between their own constructions and the prevailing ones. Although anxiety-related responding and skill deficits have historically been associated with performance-based problems such as high mathematics anxiety, the relative contributions of these variables to substandard performance remains poorly understood. Utilizing a 7% carbon dioxide gas mixture to induce fear, the present study was designed to determine the impact of fear-related responding to cognitive task performance. The purpose of this study was to examine subordinates’ perceptions of superiors’ humor orientation, assertiveness, responsiveness, avoidance strategies, approach strategies, and overall superior satisfaction. Participants were individuals over thirty that work at least 30 hours a week. 158 participants completed surveys. Results of the study show that humor orientation was positively and significantly correlated with responsiveness. Likewise, humor orientation was positively and significantly correlated with superior satisfaction. Humor orientation was also positively and significantly correlated with approach strategies. However, humor orientation was negatively associated with avoidance strategies. Furthermore, a post hoc analysis revealed that females perceived their supervisors as being …

This study examined the organizational personality types of "upward mobile," "ambivalent," and "indifferent" with reference to their relationships with worker sociocommunicative orientation, communication apprehension, immediacy, and job satisfaction. The study examined self-report data from the measures of organizational personality scales, job satisfaction, communication apprehension, immediacy, and assertiveness/responsiveness. The study found that each of the three personality variables have unique relationships (some linear, some non-linear) with communication apprehension, immediacy, assertiveness, responsiveness, and job satisfaction.

One hundred and fifty-two volunteer male participants from Carnegie Mellon, Duquesne and Fairmont State took part in the study. The sample consisted of three National Collegiate Athletic Association (NCAA) football divisions (i.e., IAA, II, III). To understand tissue participation in lysine oxidation, chicken lysine α-ketoglutarate reductase (LKR) activity and lysine oxidation (LOX) were measured in vitro. LKR activity was measured using a spectrophotometer and LOX was measured by collection of 14CO2 from [U-14C] L-lysine. LKR and LOX were measured in homogenates prepared from liver, kidney, pancreas, heart, brain, lungs, spleen, muscle, and intestine. LKR activities were 1000-fold LOX. LKR activity was highest in liver and intestine each approximately 63 µmoles/(hr*g tissue). Activity in other tissues was significantly greater than zero except for muscle. The overall goal of this project was to investigate the feasibility of a new strategy to inhibit the NF-κB signal transduction pathway. Peptide P1 that resembled IκBα , the intracellular inhibitor’s kinase binding domain was designed to competitively inhibit IκBα’s degradation. Preliminary results in stably transfected cells indicated that P1 produced a two-fold inhibition in NF-κB dependent luciferase activity in Lipopolysaccharide (LPS) stimulated RAW cell line, but not in BEAS-2B cell line. A transient transfection protocol was optimized in the BEAS-2B cell line and the effect of P1 was compared to SN50 a known inhibitor of NF-κB pathway. Our results …
