Evaluation Of Survival And Recovery Characteristics Of Bifidobacteria As Indicators Of Fecal Pollution Of Water, John Elmer Ravenscroft Jr. Dec 2000

Evaluation Of Survival And Recovery Characteristics Of Bifidobacteria As Indicators Of Fecal Pollution Of Water, John Elmer Ravenscroft Jr.

Graduate Theses, Dissertations, and Problem Reports (ETD)

The examination of aquatic environments for bacterial indicator organisms has proven to be a useful and well-established practice for the purpose of monitoring microbiological water quality. An ideal indicator of fecal pollution should be present in sufficient density to allow detection, present simultaneous with pathogen(s), incapable of aftergrowth in external aquatic environments, easy to enumerate, and exclusively of fecal origin. Unfortunately, none of the indicator organisms presently in use today meet all of these requirements. Bifidobacteria show promise as indicators of fecal pollution in water with the additional ability of potentially distinguishing between fecal pollution of human and animal origin. …

Characterization Of The Airway Epithelial Bioelectric Mechanisms Associated With The Effects Of Epithelium-Derived Relaxing Factor In Guinea Pig-Isolated Trachea, Richard Alan Johnston

Graduate Theses, Dissertations, and Problem Reports (ETD)

In response to elevated serosal or mucosal tonicity, guinea-pig tracheal epithelium releases the non-nitric oxide, non-prostanoid, epithelium-derived relaxing factor (EpDRF), which subsequently diffuses to the underlying airway smooth muscle to initiate relaxation. Hypertonicity-induced smooth muscle relaxation via EpDRF involves epithelial Na+ and Cl- channels and is preceded by a depolarization of the transepithelial potential difference (Vms). By utilizing two different experimental systems, this study sought to characterize further the airway epithelial bioelectric mechanisms associated with the effects of EpDRF in guinea-pig isolated trachea.;First, this study attempted to elucidate which epithelial ion channels and/or transporters are involved in the depolarization of …

Factors Influencing Brook Trout (Salvelinus Fontinalis ) Abundance In Forested Headwater Streams With Emphasis On Fine Sediment, James Philip Hakala Dec 2000

Factors Influencing Brook Trout (Salvelinus Fontinalis ) Abundance In Forested Headwater Streams With Emphasis On Fine Sediment, James Philip Hakala

Graduate Theses, Dissertations, and Problem Reports (ETD)

The influence and relative importance of fine sediment on wild brook trout (Salvelinus fontinalis) population size was assessed. Brook trout production in headwater streams was inversely proportional to substrate permeability in one of two years, suggesting substrate composition influenced production. Results suggest the critical fine sediment size for brook trout in this study, is between 0.063 mm and 1.0 mm. Further, fine sediment (<0.063 mm) should not exceed 0.6--1.0% of spawning substrate, or negative population effects may be incurred. Under normal flow conditions, fine sediment was a principal determinant of juvenile trout abundance relative to other physical and biological factors. Adult trout abundance was principally a function of stream discharge, and is potentially further influenced by fine sediment impacts on juvenile recruitment. Spatially restricted food resources, created by drought-induced low flows in year 2 of the study, are believed to have over-powered mechanisms influencing trout abundance under normal flows.

Diversity Of Ectomycorrhizal Fungi On Two Reclaimed Surface Mines Differing In Soil Properties, Betsy S. Kurnik Dec 2000

Diversity Of Ectomycorrhizal Fungi On Two Reclaimed Surface Mines Differing In Soil Properties, Betsy S. Kurnik

Graduate Theses, Dissertations, and Problem Reports (ETD)

I analyzed community structure of ectomycorrhizal fungi on white pine (Pinus strobus) on two reclaimed surface mines in eastern Ohio. One site was acidic and metal-stressed, the other was alkaline. Completely different community composition was found on the two sites by PCR/ITS/RFLP analyses of field collected mycorrhizae. However, Shannon-Wiener index values indicated that the two communities were equally diverse. A population of isolates of one species common to both sites was obtained from trap cultures planted with white pine. The identity of this species as Wilcoxina mikolae was established through DNA sequencing of the ITS region. The W. mikolae populations …

Removal Of Uranium From Aqueous Wastes Using Electrically Charged Carbon Nanofibers, Shannon Lee Stover

Graduate Theses, Dissertations, and Problem Reports (ETD)

The presence of aqueous uranium wastes is a problem in the United States and their treatment/disposal is desirable. Treatment methods have been developed but result in concentration opposed to conversion to a disposable form. This technique involves recovery of uranium as a solid, providing an advantage over other methods. The technique utilizes carbon nanofibers as electrodes which successfully electrosorb uranium ions. Fibers with varying surface characteristics were evaluated in the removal process and all were determined to be equally and extremely effective. Various experimental parameters were evaluated including applied potential, pH, and flow rate. The critical applied potential at which …

Breastfeeding Personal Efficacy Beliefs Of Women University Students, Ann Pollard Cleveland Dec 2000

Breastfeeding Personal Efficacy Beliefs Of Women University Students, Ann Pollard Cleveland

Graduate Theses, Dissertations, and Problem Reports (ETD)

Many women do not initiate breastfeeding although breastfeeding promotes their baby's health. Three reasons why women do not breastfeed are women's mental organization, cultural beliefs about breastfeeding, and women's breastfeeding personal efficacy beliefs. The conceptual framework for personal efficacy beliefs is Bandura's social-cognitive theory (1992, 1995, 1997). This dissertation used a new research instrument to examine women university students' breastfeeding personal efficacy beliefs. The instrument was tested by mail survey techniques that resulted in a 70.6% respondent rate. University women's personal efficacy beliefs about breastfeeding factored into five factor subscales. In order of magnitude, the personal efficacy belief factors were …

Development Of A Membrane Resistance Based Modeling Framework For Comparison Of Ultrafiltration Processes, David Albert Masciola Dec 2000

Development Of A Membrane Resistance Based Modeling Framework For Comparison Of Ultrafiltration Processes, David Albert Masciola

Graduate Theses, Dissertations, and Problem Reports (ETD)

A parametric study was conducted to develop a more general modeling framework as a basis for comparison of ultrafiltration membrane modules and separation processes. Discrete experiments were performed to examine the effects of operating parameters on permeate flux behavior in the tubular and high-shear rotary ultrafiltration (HSRUF) systems using a synthetic metal working (MW) fluid as a surrogate feed stream due to its relevance in industrial process and treatment applications. The conventional thin-film model was successfully applied to permeate flux data collected at low feed oil concentrations; however, model predictions at high concentrations were physically meaningless due to the formation …

Effects Of Plant Flavonoids On The Fate Of Polynuclear Aromatic Hydrocarbons In Rhizosphere Soil, Xiujin Qui Aug 2000

Effects Of Plant Flavonoids On The Fate Of Polynuclear Aromatic Hydrocarbons In Rhizosphere Soil, Xiujin Qui

Graduate Theses, Dissertations, and Problem Reports (ETD)

High-molecular-weight polynuclear aromatic hydrocarbons (PAHs) are persistent in the environment although a wide variety of microorganisms can metabolize PAHs. In the past decades, laboratory and filed studies have shown that PAH degradation in soil is often limited by poor bioavailability and oxygen availability. Bound residue formation of PAHs with macromolecules of soil organic matter is an important fate mechanism. More recently, phytoremediation for PAH-contaminated soils is being explored. It is believed that PAH degradation may be enhanced in rhizosphere soil due to the improved aeration condition and the active soil microbial community sustained by root exudates. Whether certain root exudates …

Induction Of Myosin Cross-Reactive Antibody And Cytolytic T Cell Responses In Mice With Streptococcus Pyogenes, Cynthia Ann Cunningham Aug 2000

Induction Of Myosin Cross-Reactive Antibody And Cytolytic T Cell Responses In Mice With Streptococcus Pyogenes, Cynthia Ann Cunningham

Graduate Theses, Dissertations, and Problem Reports (ETD)

Impetigo and pharyngitis are two common suppurative infections in man elicited by Streptococcus pyogenes that lead to autoimmune complications in susceptible individuals. Although the underlying mechanisms for the onset of post-streptococcal autoimmunity have yet to be established, experimental information gained thus far suggests that the resultant pathology is a consequence of immunological cross-reactivity between streptococcal antigens, namely the anti-phagocytic factor M protein and host proteins, specifically myosin. Experimental animal model systems developed to date have predominantly relied on the immunization of adult animals with M protein emulsed in adjuvant. In the laboratory we have undertaken studies with mice to examine …

Surface-Enhanced Raman Spectroscopic Studies Of The Orientation Of Organonitriles On Metal Colloids, Ramaa Nagapattinam Ramakrishnan

Graduate Theses, Dissertations, and Problem Reports (ETD)

The orientation of adsorbates on metal colloids was studied by Surface enhanced Raman spectroscopy. We studied the orientation of cyanopyridines, aminomethylpyridines and 4-biphenylcarbonitrile on gold and copper colloids as a function of concentration of the adsorbates. It was found that the orientation of the adsorbate did not change as a function of concentration when adsorbed on gold colloids and was end-on for all concentrations. The orientation of 4-biphenylcarbonitrile was flat-on on gold colloid. The orientation of the adsorbate changed with change in concentration when adsorbed on copper colloids. The orientation was flat-on for higher concentrations and end-on for lower concentrations …

Scale, Ecological Fallacy, And The River Continuum Concept, Hope M. Childers Aug 2000

Scale, Ecological Fallacy, And The River Continuum Concept, Hope M. Childers

Graduate Theses, Dissertations, and Problem Reports (ETD)

Concern over scale is not new, and it is not restricted to geographers. Spatial and temporal scaling is a conceptual and methodological problem for all sciences using geographic information. This paper teases out issues of scale and ecological fallacy from the literature and discusses how these issues influence the applicability of an influential theoretical framework in stream ecology, the River Continuum Concept (RCC). Investigators are faced with decisions regarding scale during sampling location selection, field data capture, and subsequent data interpretation. A thorough understanding of the heterogeneity of stream habitats and the life histories of the organisms being studied could …

Dynamical Analysis Of Infant Social Referencing, Melissa Sue Atkins Aug 2000

Dynamical Analysis Of Infant Social Referencing, Melissa Sue Atkins

Graduate Theses, Dissertations, and Problem Reports (ETD)

The purpose of the present study was to examine relations between joint attention, language, maternal sensitivity, and temperament and the emergence of social referencing in infants and to closely observe variability in social referencing. Five infants, 7 months of age, were observed on a weekly basis for a period of four months. All five infants were engaging in social referencing behavior at the beginning of the study, which is much earlier than the literature indicates; therefore, the onset of the behavior was missed. However, results did indicate variability in social referencing behavior, in that infants referenced more in the laboratory …

Graduate Theses, Dissertations, and Problem Reports (ETD)

Applications of the electrochemical method, AC Voltammetry (ACV), and its modifications based upon monitoring ac current as a function of ac potential modulation are described. The first chapter involves investigating the effect of a short peptide, melittin, on both hybrid bilayer membrane (HBM) dielectric properties and HBM resistance to heterogeneous electron transfer. These effects are discussed in terms of three possible mechanisms by which melittin may affect HBMs. In the second chapter, a modification of ACV, second harmonic ACV, is considered. A data processing method is suggested to analyze data collected at high perturbation frequencies at which the effect of …

Structural Determinants Of Murine Leukemia Virus (Mlv) Reverse Transcriptase (Rt) Important For Fidelity And Drug Resistance In Vivo, Elias Konstantine Halvas Aug 2000

Structural Determinants Of Murine Leukemia Virus (Mlv) Reverse Transcriptase (Rt) Important For Fidelity And Drug Resistance In Vivo, Elias Konstantine Halvas

Graduate Theses, Dissertations, and Problem Reports (ETD)

Error-prone DNA synthesis by retroviral reverse transcriptases (RTs) is a major contributor to variation in retroviral populations generating mutants that exhibit altered host tropisms, resistance to antiviral drugs, and the ability to escape the host's immune system. To identify structural elements of murine leukemia virus (MLV) RT important for fidelity and drug-resistance in vivo, we developed a D17-based encapsidating cell line (ANGIE P) which is designed to express the amphotropic MLV envelope. ANGIE P cell line also contains an MLV-based retroviral vector (GA-1), which encodes a wild-type bacterial beta-galactosidase gene (lacZ) and a neomycin phosphotransferase gene. Transfection of ANGIE P …
