The onset of the 2020 COVID-19 pandemic, caused by the novel coronavirus SARS-CoV-2, issued a call to action in the fields of biomedical science and public health to implement strategies to contain and protect against the viral threat. In record times, pharmaceutical companies produced antiviral therapies and novel mRNA vaccines that were approved by the FDA for human use. With time, these vaccines were able to assist in ending the pandemic. However, time also showed the limits of mRNA vaccine induced immunity. Black bass Micropterus spp. are popular sport fish in West Virginia’s navigable rivers. In recent years, anglers have expressed concerns about a potential decline in fishery quality. To investigate these claims, we evaluated whether fishery quality has declined compared to earlier periods and whether population trends correspond to angler catch rates. Pregnancy loss is a common and intense personal event that has been found to be a risk factor for increased symptoms of mood, trauma, obsessive-compulsive and anxiety disorders in subsequent pregnancies. Using a self-report online survey, the current study hypothesizes that levels of anxiety, trauma, and obsessive-compulsive symptoms will be higher in pregnant individuals who had a pregnancy loss in their most recent previous pregnancy (n= 61) when compared to pregnant individuals whose most recent previous pregnancy resulted in a live birth (n= 62). Osteosarcoma (OS) is a rare pediatric bone malignancy. Those who develop advanced disease, often in the form of metastasis to the lung, experience poor five-year survival rates near approximately 25%. Considering minimal-to-no therapeutic breakthroughs have occurred for treatment of this disease since the 1980s, efforts to develop new therapeutics are underway. The goal of this dissertation was to initiate the preclinical efforts necessary to reinvigorate the use of immunostimulatory cytokine therapy for the treatment of OS and other solid tumors. This study employed design-based research to explore the efficacy of professional learning materials and approaches in developing social justice advocacy with rural elementary teachers in Appalachia. Bronfenbrenner’s (2005) bioecological theory and Boler’s (1999) pedagogy of discomfort formed the theoretical framework for the design, implementation, and analysis. Mid-way through the study, Allport’s (1954) contact hypothesis was included to inform the revision of intervention approaches. Four elementary teachers attended seven in-person informational discussion meetings over five months. During this time, these teachers also read two books and completed journal responses and surveys. Phosphodiesterase-6 (PDE6) serves as a pivotal component in the phototransduction pathways of both cone and rod photoreceptors. In cones, PDE6 consists of tetrameric subunits: inhibitory (γ') and catalytic (α'). The catalytic subunit, PDE6α', contains a C-terminal prenylation motif. Deletion of this motif is associated with achromatopsia (ACHM), a form of color blindness. The mechanisms underlying the disease and the roles of PDE6 lipidation in vision remain elusive. Meanwhile, rod PDE6 is composed of α and β catalytic subunits and γ inhibitory subunits, with alterations in the C-terminal "prenylation motif" of PDE6β linked to retinitis pigmentosa (RP) pathology. Although considered a rare form of nonmelanoma skin cancer, Merkel Cell Carcinoma incidence rate has increased to 0.74 per 100,000 and projections expect this number to steadily rise over the next decades and to reach an average of 3,285 cases per year in 2025. MCC incidence increases significantly with age. Among those aged 40-44, it is 0.44, it rises to 1.0/100,000 in the 60-64 age group and soars to 9.8/100,000 among those ≥85 years and above. Antisocial personality disorder (ASPD) is a destructive and severe mental illness that costs the U.S. trillions of dollars annually. Although ASPD is significantly correlated with acts of aggression, violence, and imprisonment, not all individuals with ASPD are aggressive. In samples without ASPD, increased aggression has been linked to an error in social information processing, which is referred to as hostile attribution bias (HAB).

The objectives of this study were to 1) formulate a novel energy bar containing significant macronutrients and calories per serving with favorable ratings from consumers; 2) analyze responses to a consumer preference survey and sensory evaluation of novel energy bars compared to a commercially available energy bar. For the first study, fourteen iterations of an energy bar were synthesized with differing protein sources (whey, soy, pea, hemp), and characteristic variations (nougat, chocolate truffle, peanut butter, double-date caramel). Exploring the routes of exposure and biological fate of 4,4’-methylene diphenyl diisocyanate (MDI) has been a challenge for many years. Occupational exposure to MDI can result in the development of occupational asthma (MDI-OA) which results in loss of productivity, large economic costs, and costs to the individual worker. Initially, it was thought that respiratory exposure to MDI was the most likely route leading to MDI-OA. This dissertation discusses three instances of temporal prediction, applied to population dynamics and deep learning.

In population modeling, dynamic processes are frequently represented by systems of differential equations, allowing for the analysis of various phenomena. The first application explores modeling cloned hematopoiesis in chronic myeloid leukemia (CML) via a nonlinear system of differential equations. By tracking the evolution of different cell compartments, including cycling and quiescent stem cells, progenitor cells, differentiated cells, and terminally differentiated cells, the model captures the transition from normal hematopoiesis to the chronic and accelerated-acute phases of CML. Lung cancer remains the most frequently diagnosed malignant neoplasm shared in men and women worldwide, second only to the sexually dimorphic prostate cancer in men and breast cancer in women. Overall, lung cancer was responsible for 12.5% of new cancer cases globally in the year 2022. This dissertation explores predicting the physical properties of solids using first-principles methods, with a focus on Density Functional Theory (DFT). DFT uses the electronic density within a material to predict its properties, simplifying the treatment of electron-electron interactions and allowing the study of realistic systems with a balanced treatment between accuracy and computational efficiency. Nearly all neurodegenerative diseases (NDs) and some eye diseases are characterized by the presence of misfolded and aggregated proteins. The ubiquitin proteasome system (UPS) is responsible for degrading the majority of proteins including misfolded and aggregated proteins. However, in NDs the performance of the UPS is impaired, and this impairment partially comes from these protein aggregates inhibiting the proteasome. This inhibition leads to an accumulation of protein aggregates, more proteasomal inhibition, and fewer chaperones for newly synthesized proteins, triggering a collapse in proteostasis, which is thought to contribute to the pathogenesis of NDs. In Chapter 2, a study was designed to evaluate the effects of varying dietary branched-chain amino acids (BCAA) ratio in corn and soybean meal-based turkey starter diets. The BCAA leucine, isoleucine, and valine are considered indispensable amino acids needed for energy and stimulation of muscle protein synthesis via the mammalian target of rapamycin (mTOR) pathway. Imbalanced BCAA ratios can lead to antagonism and degradation of limiting BCAA, particularly in diets with excess leucine. This research presents the development of a data-driven machine learning-assisted approach to simulate and optimize the design of composite materials subjected to low-velocity impacts. It focuses on a specific type of hybrid composite comprised of fiberglass and Kevlar fabrics stitched with Kevlar threads. The study begins by creating a multiscale finite element simulation for the composite under a low-velocity impact, which operates across three scales: microscale, mesoscale, and macroscale. This simulation accepts various inputs, such as different layer configurations and orientations, and provides impact outputs including maximum load and energy absorption capacity and displacement at failure.

Investigation Of L-Serine Synthesis And Transport As Stabilizing Feedback Mechanisms Within The Tsetse-Wigglesworthia Symbiosis, Ryan Phillips

Tsetse flies (Glossina morsitans), which are the obligate vectors of Old-World trypanosomes (Trypanosoma spp.) that cause African trypanosomiasis, have a bacteriome that houses the intracellular endosymbiont Wigglesworthia glossinidia. The ancient coevolution of these species has led to the integration of Wigglesworthia metabolism into tsetse physiology and a subsequent reliance on nutrient exchange for maintaining the fitness of both partners. The establishment of this obligate mutualism has facilitated the emergence of feedback mechanisms that stabilize the symbiosis, such as that which involves the exchange of L-serine and B vitamins. Metal-organic frameworks (MOFs) are organic crystalline hybrids that have been used for a variety of applications from catalysis to gas storage and drug delivery. Studies have shown that generally, while such frameworks are more biocompatible than similar aspect ratio nanostructures, they are also more prone to transformation and disintegration due to their flexible and fragile nature as resulted from the structure assembly and implemented structure-function relationships.
