Childhood adversity has long-lasting negative effects across the lifespan including increased mortality risk. The love and support individuals receive from others, also known as social support, has shown to be a protective factor against ACEs. However, little research has investigated the amplifying effects of social conflict and strain that often accompanies social relationships. Utilizing data from the Midlife Development in the U.S. (MIDUS) study, I tested whether higher levels of social support would buffer the negative effects of adverse childhood experiences on mortality risk, and whether higher levels of social strain would amplify these associations. In the first chapter I identify the impact of fentanyl exposure misinformation- namely, the erroneous belief that momentary, passive contact with the potent opioid fentanyl can be seriously harmful- on first responder behavior during overdose events, and on overall opioid-related mortality. I examine changes in opioid-related mortality following one particularly well-covered episode involving an Ohio police officer in 2017, wherein the officer appeared to experience an acute opioid overdose after touching what was believed to be fentanyl. Employing a synthetic differences-in-differences identification strategy, I find areas with greater media exposure to this misinformation exhibit marked increases in opioid overdose deaths; …

The studies described here focus on mutations in protein kinase CK2 linked to ‘Okur-Chung’s Neuropathy’ (OCN), a neurodevelopmental disorder that involves 10 distinct mutations in the catalytic subunit CK2α. It has, however, remained unresolved if these mutations impair CK2α activity or if they also impair association with the regulatory CK2β subunit, which is necessary for assembly of the α2β2 holoenzyme. I describe efforts to resolve these questions for two mutations, Y50S and D175G. The former is thought to reside at the CK2α-CK2β interface, whereas the latter is located proximal to the active site. Neural electrode technology has been around for centuries since the times of Galvani. In early electrophysiology experiments metal wires were used to induce contractions in dissected animals. The metal wire electrode has since been a standard tool to both stimulate and record neural activity. In the past two decades, a new strategy for neural stimulation has been formulated based on the emergent field of optogenetics. An excretory/secretory (E/S) product, common to many helminths, is neutrophil inhibitory factor (NIF) which negatively affects neutrophil migration and activity. Due to differences in neutrophil accumulation between parasite resistant St. Croix STC and parasite susceptible Suffolk (SUF) sheep, we hypothesized that Hc-NIF may inhibit migration of SUF and not STC neutrophils. Previous studies have demonstrated that Hc produces NIF but is not commercially available. To test our hypothesis, neutrophils were cultured from STC and SUF sheep in the presence of Ancylostoma caninum-derived NIF (Ac-NIF) and measure chemotaxis to Interleukin-8 (IL-8), HcLA or H. contortus third-stage excretory/secretory (E/S) products. The applied science of bioinformatics encompasses computational analysis of molecular biology data. Advances in genomics and DNA sequencing technology have enabled computational analysis of ribonucleic acids (RNAs), which play diverse and critical roles in most cells. To assist the study of human RNA, we trained machine learning models on RNA nucleotide sequences, devoid of domain knowledge. We built models that distinguish long non-coding lncRNA from protein-coding mRNA, and models that predict the cytoplasmic vs. nuclear preferences of lncRNAs. In Amazonian River systems, diverse fish communities depend on the floodplains for providing feeding opportunities and critical spawning habitat during the seasonal flood pulse, which annually submerges floodplain forests. In the Amazon, large-scale deforestation and hydroelectric dam construction can have profound impacts on entire ecosystems by altering river-floodplain connectivity. The neural mechanisms that control the onset of puberty are not completely understood. In livestock, the onset of puberty relies on an increase in gonadotropin releasing hormone (GnRH) and luteinizing hormone (LH). Kisspeptin, neurokinin B, and dynorphin neurons (KNDy) and their receptors, Kiss1r, NK3R, and KOR, respectively, are important regulators of puberty. Previous data from our laboratory has shown that KNDy neurons may be responding to other inputs, including the melanocortin system, that will affect the timing of puberty. Huntington’s disease (HD) is an autosomal dominant neurodegenerative disorder resulting from an abnormally long CAG repeat in the huntingtin gene. This results in an expanded polyglutamine (polyQ) domain in the huntingtin protein (htt) that directly leads to self-assembly of huntingtin (htt) into a variety of aggregate species that underlie cellular toxicity. Interaction of protein aggregates with a variety of lipid membranes has been linked to disease pathology. Big game hunting is an important source of revenue and recreation in the United States. Of big game species, white-tailed deer (Odocileus virginianus) are the most heavily pursued. White-tailed deer are a widespread generalist species, capable of existing at high densities. The project first traces the earliest origins of natural history writing, surveying how natural history writing from antiquity presented and categorized the living world, and then how the rise of print culture and growing popularity of natural history writing would facilitate the rise of European empires. Massive galaxy mergers are a fundamental consequence of the dynamic evolution of the Universe and play a central role in the evolution of galaxies. Because all massive galaxies harbor a central supermassive black hole (SMBH; M ≥ 106 M⊙), studying these merging systems with electromagnetic (EM) techniques constrains galaxy evolution models and provides a systematic means to examine the astrophysical mechanisms that facilitate the growth of SMBHs. Long-term internal states drive many behaviors in animals. Zebrafish is a common vertebrate model conducive to finding the neural coding to these behaviors. In the Horstick lab, the larval zebrafish light search behavior is central to our research. This behavior, which is the locomotor response to the loss of light in larval zebrafish, is controlled by multiple neural populations that drive different internal states. The sensory neurons initially responsible for this behavior are photosensitive due to the expression of light-absorbing proteins called opsins. In female sheep, the release of gonadotropin-releasing hormone (GnRH) from the hypothalamus triggers luteinizing hormone (LH) to be released from the pituitary. Luteinizing hormone, in turn, acts on the ovaries to elicit estradiol (E2) release. E2 acts in a negative feedback manner to control the amount of LH released. The increase in LH release that drives puberty onset results from a decrease in sensitivity to E2 as the animal ages. Child abuse is a significant public health concern that impacts children worldwide. Efforts to connect at-risk parents with prevention and intervention programs require the use of high-quality measures that evaluate child abuse potential. Researchers frequently use the Child Abuse Potential Inventory (Milner, 1986) and the Brief Child Abuse Potential Inventory (BCAP; Ondersma et al., 2005) which contain Validity indices that evaluate response patterns such as social desirability responding. Traumatic brain injury (TBI) and alcohol use disorder (AUD) exhibit a bidirectional relationship: AUD increases the risk of incurring a TBI and, inversely, a history of TBI increases the risk of future AUD. The second portion of this relationship has not been as thoroughly investigated. Our lab previously demonstrated that a juvenile TBI significantly increases alcohol self-administration in females compared to uninjured females and the injured females developed a conditioned place preference (CPP) for ethanol. Synaptic zinc signaling modulates synaptic activity and is present in specific populations of cortical neurons, suggesting that synaptic zinc contributes to the diversity of intracortical synaptic microcircuits and their auditory tuning properties. Stimulus-specific adaptation is a hallmark of sensory processing in which a repeated stimulus results in diminished successive neuronal responses, but a deviant stimulus will still elicit robust responses from the same neurons. Recent work has established that synaptically released zinc is an endogenous mechanism that shapes neuronal responses to sounds in the auditory cortex. Prescribed burning has become a commonly used tool in the mitigation of wildfire, though monitoring the way it changes ecosystems has historically been a time-intensive process. Rapid change across ecosystems has necessitated advancements in remote sensing technologies to quantify the changes taking place. Single-scan terrestrial LiDAR scanning is one such method of monitoring these changes through the quantification of ecosystem structural characteristics. Acute disturbance events such as fire can transform the structure of an ecosystem, and by extension, change the way that ecosystem functions.
