Genes specifically expressed in oocytes are important for the development of oocytes and early embryos. By analyzing ESTs from a rainbow trout oocyte cDNA library, we identified a novel EST sequence that does not show homology to any sequences in the GenBank. Analysis of tissue distribution by RT-PCR revealed that this gene was only expressed in unfertilized oocytes. Sequencing of the EST clone identified a cDNA of 3163 bp. Northern blot analysis showed the novel gene has a single transcript of 3.4 kb. Additional 5' sequence was obtained by 5' RACE, extending the novel cDNA to 3333 bp. Analysis of …

The DNA damage response is a complex network of signals that coordinate to protect cells from accumulating mutations that lead to the development of cancer. Upon the introduction of DNA damage from either environmental or endogenous sources, the DNA damage response coordinates the control of cell cycle with DNA repair mechanisms to ensure genomic integrity within the cell. Mutations in proteins in this pathway lead to genomic instability and early onset of cancer. This thesis investigates issues related to the processing of multispectral and color infrared images of the iris. When utilizing the color bands of the electromagnetic spectrum, the eye color and the components of texture (luminosity and chromaticity) must be considered. This work examines the effect of eye color on texture-based iris recognition in both the near-IR and visible bands. A novel score level fusion algorithm for multispectral iris recognition is presented in this regard. Cathedral State Park (CSP) is a 54-ha, old-growth, hemlock-hardwood forest located in the central Appalachian Mountains of West Virginia. Trees larger than 100 cm diameter at breast height (dbh) and 35 m tall are common. Hemlock woolly adelgid (HWA) (Adelges tsugae Annand) is an exotic insect that currently threatens eastern hemlock (Tsuga canadensis (L.) Carr.) throughout its native range, and was found in CSP in 2002. In 2006, field plots established in 2000 were remeasured to assess adelgid-induced changes to forest structure and species composition. The amount of sediment delivered to stream channels and the determination of the topographical attributes responsible for the origination and transfer of sediment were investigated in a central Appalachian mixed hardwood forest from 2002 through 2005. Two study watersheds were chosen within the Indian Run watershed in north central West Virginia. One remained undisturbed (control) and the other underwent road construction and tree harvesting operations (treatment). Sediment samples were collected from silt fences which were installed along both sides of the stream channels in both watersheds to ensure all sediment material delivered from adjacent hill-slopes was captured and collected. Season extension methods have been used in many climates and crops for centuries on all scales of vegetable production. Our research compared the effect of six season extension methods on soil and ambient temperatures and yields of warm and cool season vegetables in organic production. The methods under investigation in field plots were row cover, row cover with water tubes, low tunnel, low tunnel with water tubes, and a control for comparison, which were all replicated three times in field plots. Despite its abundance and nutritional value, krill has not been widely utilized for human consumption due to the lack of proper technology for protein recovery. The study objectives were to isolate krill protein concentrate (KPC) and to determine the nutritional value, health benefits, and safety of KPC for human consumption. Proximate analysis indicated KPC on a dry basis is composed of ∼78% protein and ∼8% fat of which ∼27% are omega-3 polyunsaturated fatty acids (o-3 PUFAs). The amino acid composition of KPC indicated that all nine essential amino acids were present in amounts that met the requirements for adult humans …

An intersex condition, defined as the presence of oocytes in the testes of male gonochoristic fish, has been observed in smallmouth bass in the South Branch of the Potomac River, West Virginia, which indicates exposure to exogenous estrogens. Endocrine disrupting chemicals (EDC's) are generally hydrophobic and would tend to be found within the sediment of aquatic environments. Few studies have attempted to show the effects of exposure to EDC's on fish using sediment chemical extracts. Acid mine drainage (AMD) and acid rain are important sources of impairment to streams in the Tygart Valley and Cheat River basins in north central West Virginia, USA. Due to a network of abandoned mined lands and bond forfeiture sites in this coal-mining region, AMD represents severe, but rather localized impacts to water quality. AMD is a consequence of the chemical oxidation of reduced geological minerals (sulfides) usually associated with coal during mining operations. This dissertation proposes a new algorithm for performing simultaneous haplotype resolution and block partitioning. The algorithm is based on genetic algorithm approach and the parsimonious principle. The multiloculs LD measure (Normalized Entropy Difference) is used as a block identification criterion. The proposed algorithm incorporates missing data is a part of the model and allows blocks of arbitrary length. In addition, the algorithm provides scores for the block boundaries which represent measures of strength of the boundaries at specific positions. The objective of the work was to conduct water electrolysis at room temperature with reduced energy costs for hydrogen production. The electrochemical gasification of carbons consumes only 9.6 kcal/mol H2O compared to 56.7 kcal/molH 2O for conventional water electrolysis. In this work, carbon-assisted hydrogen production and the reaction energetics/kinetics at applied potentials |E0| between 0.1 and 1.8 V are studied. The carbon promoted water electrolysis could be performed at applied potentials as low as |E0|=0.21 V as opposed to conventional water electrolysis which requires |E0|>1.25 V. The Fivemile and Eads Mill members of the upper part of the Hinton Formation are two significant marine zones that occur within the Mauch Chunk Group in southwestern Virginia and southern West Virginia.;Multivariate analyses of binary fossil data for the Fivemile Member were performed in order to determine any underlying paleoecologic signals. Analyses of the data sets indicate that this member is dominated by a euryhaline fauna in four of the five locations studied. Euryhaline fauna characteristic of the Fivemile Member include the brachiopod Lingula, the bivalves Modiolus and Septimyalina, and ostracods. We developed a novel in vivo model utilizing acute stretch to investigate endothelial cell (EC) proliferation as a marker of vascular growth in healing SKH1 mouse skin. We also used human umbilical vein endothelial cells (HUVECs) as an in vitro model system to validate postulated tissue insulin-mediated signal transduction pathway(s) using paradigms that would prove lethal in the animal model.;Dorsal distally based flaps of skin were stretched for 3 min using linear (skin hook) plus hemispherical load cycling (inflated subcutaneous silicone catheter). This dissertation research examines differential flood vulnerability in the Thulamela municipality of the Limpopo Province of South Africa. The study employs a triangulation of methods within a Participatory GIS methodology. Political ecology is the conceptual framework. The main premise of this research is that household flood vulnerability is socially constructed in that it is mediated by locally contingent political, socio-economic and environmental factors. As a result, conventional solutions that focus mainly on the physical event are ineffective and costly because they ignore local knowledge about flood hazards and flood coping mechanisms that have been disrupted or destroyed. Culture-dependent denaturing gradient gel electrophoresis (CD DGGE) fingerprinting of the 16S rRNA gene was developed to characterize mixed soil bacterial communities recovered on agar plates. CD DGGE analyses resulted in clear banding patterns of sufficient complexity (16--32 major bands) and reproducibility to investigate differences in bacterial communities in silt loam soils. Replicate CD DGGE profiles from plates inoculated with less-dilute samples (10 -3) had a higher band count and were more similar (72--77%) than profiles from more-dilute samples (51--61%). The performance of saw-tooth gradient for comprehensive proteome analysis is evaluated using model proteins and a low molecular weight human serum by reverse-phase high performance liquid chromatography. The peak splitting and number of peptides detected is used to characterize the effective peak capacity of the saw-tooth gradient as a function of solvent strength change (Delta&phis;). Peak splitting demonstrated in experimental result as one single peptide or protein detected in more than one saw-tooth step. No peak splitting is observed directly from chromatograms for two model proteins by decreasing the solvent strength change (Delta&phis;) from 5.0%, 1.0%, to 0.1%. The numerical …

A vast amount of the current research in medical image analysis has aimed to develop improved techniques of image segmentation. Of the existing approaches, active contour methods have proven effective by incorporating edge or region information from the image into a level set formulation. However, complications arise in images containing regions of low-contrast due to noise, occlusions, or partial volume effects, which are often unavoidable in practical applications. Assessment in higher education has become increasingly important throughout the past 20 years. Institutions of higher education engage in student outcomes assessment for a plethora of purposes.;The researcher conducted this study to investigate current assessment practices at the undergraduate program level. The researcher determined the effectiveness of assessment plans within specific disciplines at institutions that have implemented and sustained their assessment plans. The researcher also identified the degree to which related institutional commitments and resources were provided to support assessment the assessment process. During the post-fledging period, some juvenile songbirds appear to require early successional habitats at some level, suggesting that disturbance (whether natural or anthropogenic) in the landscape is important. Previous studies of juvenile Wood Thrushes (Hylocichla mustelina) have found them in early successional areas in which adults do not nest. My objective was to determine how juvenile Wood Thrushes used areas harvested through various silvicultural techniques within a forested landscape. In this study we first conducted a three pass removal study to estimate electrofishing capture probabilities for species and size classes of fish common in the Cheat River and Tygart River watersheds of West Virginia. We then calculated models of capture probabilities for 12 species that were divided into two size classes based on site-specific physico-chemical variables. We took these capture probability models and applied them to a watershed scale study in the Cheat River and Tygart Valley River watersheds of West Virginia. Recirculating aquaculture systems enable increased fish production per volume of water. Unfortunately, recirculation systems also provide conditions for bacterial growth. Aeromonas, a genus of bacteria ubiquitous in the environment, contains fish pathogens, human pathogens, and/or spoilage organisms. Under mild conditions, a derivative of 11H-benzo[ b]fluorine 76, was successfully synthesized through tandem cyclization of the corresponding enediynyl propargylic alcohol, 69. Condensation of the substituted indanone 67 with the lithium acetylide 23 furnished benzannulated enediynyl propargylic alcohol 69 in 1:1 diastereomeric ratio. Reduction of propargylic alcohol 69 with triethylsilane in the presence of trifluoroacetic acid gave benzannulated enediynes 70. Treatment of 70 with potassium tert-butoxide in refluxing toluene furnished alkene 74. Hydroboration of alkene 74 followed by iodination in the presence of base produced alkyl iodide 75. The purpose of this investigation was to determine college students' perceptions of their own dialects and 4 specific dialects common to the English language. To determine this, the investigator developed a 3-part survey (Appendices H, I, J). On Part I, respondents completed identifying information. On Part II, they rated 9 general Likert Scale items related to dialect. On Part III, they completed the same semantic differential scale for speakers who represented 4 different dialects: Appalachian-American, Eastern-American, General-American, and African-American. Two-age harvesting has been used more frequently in the management of Central Appalachian hardwood stands as an alternative to clearcutting, but long-term responses of avifauna to these harvests have not been investigated during the breeding season. No studies have examined post-breeding bird communities in these harvests; recent research concerning post-breeding use of early-successional habitats has only examined clearcuts. Because of recent changes in the USDA Forest Service (USFS) travel management regulations, all National Forests are required to create a travel management atlas that includes a motor vehicle use map by 2009. The purpose of this study was to determine the perceptions of OHV use, issues, and management among USFS District Rangers on National Forests in the Appalachian region, and to determine the differences in perceptions of those District Rangers based on the level of OHV recreation opportunities provided on their Districts. Hexavalent chromium [Cr(VI)] compounds are redox cycling environmental carcinogens that induce apoptosis as the primary mode of cell death. Apoptosis is abnormally regulated in various disorders including cancer. Therefore, to understand the etiologies of these diseases, it is important to delineate the biochemical and molecular pathways involved in the regulation of apoptosis. The main objective of this study was to characterize the molecular mechanisms involved in Cr(VI)-induced apoptosis and malignant transformation. We found that both death receptor and mitochondrial pathways of apoptosis are involved in Cr(VI)-induced apoptosis, with the latter being more dominant.
