Using a four step phenomenological approach for inquiry, the researcher explored and interpreted data obtained from the lived experiences of sixteen perimenopausal women and eight health care providers living in rural areas. A purposive sampling technique was utilized. Participation was on a voluntary basis. Data collection procedures included a face-to-face semi-structured taped interview and completion of a demographic questionnaire. Following analysis of the data, twenty-two common essences were identified from intuiting of the interview data. These essences were classified into six major themes which integrated all the realities of the participants. The findings of the study revealed six major themes …

This portfolio is a two-volume set of original music compositions. The works contained herein represent a variety of media and genres, including chamber and large-ensemble pieces. The works are typically tonal, with the harmony usually combining tertian and quartal chords. In the Beginning is the major work of the portfolio. The goal of this work was to determine how polyunsaturated fatty acids (PUFAs) affect gene expression at the molecular level. Glucose-6-phosphate dehydrogenase (G6PD) is a member of the family of lipogenic enzymes known to be inhibited by dietary PUFAs. Addition of PUFAs to diet causes a 70-80% decrease in G6PD enzyme activity and mRNA accumulation in liver with no change in transcriptional activity of the gene, indicating that regulation occurs at a posttranscriptional step. Similarly, regulation of G6PD by starvation and refeeding results in a 12- to 15-fold increase in G6PD activity and mRNA accumulation but no change in transcriptional …

A Cumulative Strategies Model for drug abuse prevention employs a process by which two or more drug abuse prevention strategies are used conjointly, such that each strategy targets two or more factors that place children at high risk for drug abuse. This model was examined using an innovative multiple component program called CHOICES. CHOICES emphasizes the reduction of risk factors that are characteristic of a high risk population of children of alcoholics and predictive of drug abuse. Five clay minerals (Well Crystallized Kaolinite (KWC), Poorly Crystallized Kaolinite (KPC), Illite, Attapulgite, and Montmorillonite) with a range of surface characteristics were selected to investigate the adsorption of three oxyanions (molybdate, selenite, and selenate). The minerals were coated with five different levels of each of Fe and Al hydrous oxides. The charge characteristics were determined using potentiometric titration. Adsorption was studied by batch experiment at 11 different concentrations of each of the oxyanions at three different pH levels. This study is the first to assess the stability of radiation-induced chromosome aberrations in vivo over the life span of a rodent model. Redoxyendonuclease activity was detected in extracts of human neuroblastoma cells using a base-release assay specific for thymine glycol in DNA. The level of redoxyendonuclease activity was more than 2-fold higher in dividing cells compared to quiescent cells, suggesting that quiescent cells may have a reduced capacity to repair oxidative DNA base damages. Cells were synchronized by serum deprivation and then stimulated to enter the cell cycle by the addition of serum to determine enzyme activity at different stages of the cell cycle. The membrane bound enzyme methane monooxygenase (pMMO), is a mixed function oxygenase that initiates the oxidative metabolism of methane in methanotropic bacteria. Copper is required for catalytic activity of pMMO, illustrated by the recovery of pMMO activity in EDTA-inhibited membranes, whereas zinc and nickel had little effect or were inhibitory. Inhibition kinetics, cosubstrate dependence, and substrate protection experiments show that acetylene acts as a suicide substrate for pMMO, binding specifically to a 26-kDa peptide. The acetylene binding capability of pMMO increased as a function of copper concentration as does the specific activity, suggesting that copper activates pMMO by binding to …

Many small communities located in rural areas of the United States lack adequate domestic wastewater treatment plants. Constructed wetlands are being promoted as low-cost, low-maintenance systems for treatment of domestic wastewater in areas where standard septic tanks fail. In order to evaluate the efficacy of small constructed wetlands, 20 small-scale wetlands mesocosms (100 gal each) containing five plant treatments (no plants/control, Typha, Scirpus, and Juncus monocultures, and all three species combined) were planted into pea gravel at either 18 or 24 inches in depth. Fifteen male nursing students from four baccalaureate nursing schools in West Virginia participated in this phenomenological study. The phenomena of interest was the lived experiences of male student nurses related to caring. Demographic data was obtained in the interview process. Semistructured interview questions were used in the individual interviews, which were all audiotaped. The methodology used was based on the Swanson-Kauffman and Schonwald method, which entailed the four step process of bracketing, analyzing, intuiting, and describing. Five themes were identified. We have found that Host Factor I (HF-I), the product of the hfq gene, is required for efficient expression of the RpoS stationary phase sigma factor in Salmonella typhimurium. HF-I is an RNA-binding protein originally characterized for its role in replication of phage Q{dollar}\\beta{dollar} of E. coli. Its role in the uninfected bacterial cell has previously been unknown. Assays of {dollar}\\beta{dollar}-galactosidase in strains with rpoS-lac fusions, Western blot analysis, and pulse-labeling and immunoprecipitation of both fusion proteins and native RpoS show that an hfq mutant has a four- to seven-fold reduction in the translation of rpoS. The prison reformers and fiction writers of post-Revolutionary Philadelphia wrote prodigiously about crime and punishment. In a correlative study, I explore juridical topoi in reformist and fictive texts familiar to republican Philadelphians. My analysis assumes a public sphere of writers and readers: while late eighteenth-century prison reforms and fictions came out of separate signifying systems, there was considerable promiscuity between them. I group areas of overlap around cultural concerns (science, murder, sex) and representational "spaces" (home, prison, nation). In either discourse, crime and punishment were useful to a post-Revolutionary aesthetic of nationalism. The scientific rhetoric of penitentiary punishment targeted the …

More nontraditional students are attending college, and many of these students are enrolled with GED certification. The validity using GED certification as postsecondary admission criteria has been questioned. Participants included 251 students enrolled with GED certification as admission criteria to a small Mid-Atlantic college in the Spring 1997 semester. The influence of ACT, GED, literacy scores, learning and study skills, and the number of successfully completed developmental classes on grade point averages was researched. The chicken very low density apolipoprotein II (apoVLDLII) gene encodes an egg yolk protein that is synthesized in the liver in response to estrogen. Regulation of this gene is mediated by the estrogen receptor via two estrogen response elements (EREs) located in the promoter. Following the administration of estrogen, there is a four hour lag period prior to the detection of apoVLDLII mRNA. To examine the events that occur during this lag period, in vivo footprinting and run-on transcription assays were performed. Whether growth of large, ovulatory follicles differed after different modes of stimulation with FSH and whether growth was enhanced by exogenous LH were examined in prepubertal gilts. Control is often a central concept in the explanation and treatment of physical spouse abuse. This study examined the usefulness of W. J. Doherty's and N. Colangelo's Family FIRO Model in explaining issues of control in physically aggressive couple relationships. The model is an adaptation of W. Schutz's Fundamental Interpersonal Relations Orientation theory, which predicts the interpersonal fit in relationships from the satisfaction of the partners' intrapersonal inclusion, control, and affection needs. This was an instrumental case study that examined the extent to which the politics of education in Tennessee that resulted in the passage of the Education Improvement Act of 1992 and related tax increases conformed to the assumptions of the Advocacy Coalition Framework as articulated by Sabatier. The framework viewed education policy change as the result of the interaction of (a) competing advocacy coalitions within a policy subsystem over a decade or more, (b) changes that are external to the policy subsystem, and the (c) effects of stable system parameters. This study was designed to examine the relationship between the self-perceived preferred brain hemispheric processing of West Virginia public school superintendents and their self-perceived leadership styles. Respondents completed the Hermann Brain Dominance Profile, the Leader Behavior Description Questionnaire, and a demographic sheet. Data were analyzed using the General Linear Model of the Statistical Analysis System (SAS). Two hypotheses guided the study. The present investigation was based on operant theory. A prospective six-week self-monitoring design was utilized to examine naturally-occurring reinforcing and punishing effects of cardiovascular exercise in a college-aged sample of healthy women. The sample consisted of exercise initiators ({dollar}{dollar}6 months of regular exercise). Seven rewarding exercise effects emerged from factor analysis of the self-monitoring form (i.e., mood improvement, goal achievement, physical performance enhancement, exercise "high", praise from others, social affiliation, and appearance changes). The objective of this research was to determine if endothelin is a mediator of hexachloro-1,3-butadiene (HCBD)-induced acute renal failure. The nephrotoxicity associated with HCBD is acute tubular necrosis (ATN). HCBD-induced ATN is characterized by a decrease in renal blood flow (RBF), mediating a decline in the glomerular filtration rate (GFR), polyuria, glucosuria, proteinuria and natriuresis. These effects on the kidney are mimicked by the endogenous peptide endothelin. Endothelin is produced by many different cell types and is the most potent endogenous vasoconstrictor peptide identified to date. The renal vasculature is particularly sensitive to the vasoconstrictive action of endothelin. Endothelin causes …

A background questionnaire, one test, and four controlled language instruments were administered to each subject. The test, Accuracy Level Test (ALT) by Ronald P. Carver, measured the Spanish subjects English reading ability by grade level. This paper examines retiree recruitment as a strategy in state economic development policy. The study employs a theoretical framework of policy adoption and lesson-drawing to answer the questions (1) why would a state adopt the policy of retiree recruitment and (2) how is the policy adopted. Three states in different stages of retiree recruitment policy adoption were selected as case studies. Myelin associated glycoprotein (MAG) plays an important role in myelinogenesis, nerve plasticity and nerve regeneration. Hence, knowledge of how this gene is regulated may be essential for developing clinical strategies, e.g., to promote regeneration in the injured spinal cord. We employed the primer extension technique for mapping MAG gene transcription start sites. The analysis revealed 13 transcription start sites spanning 72 bp. No canonical TATA box and no known transcription initiator (Inr) sequences that position transcription complex in other TATA-less promoters were found. The determination of selenium(IV), arsenic(III), vanadium(V), and molybdenum(VI) at sub-nanomolar to picomolar levels has been investigated with square wave cathodic stripping voltammetry (SWCSV) and square wave adsorptive stripping voltammetry (SWAdSV). Se(IV) and As(III) form intermetallic compounds with copper and are deposited at a hanging mercury drop electrode (HMDE). V(V) and Mo(V) form dihydroxynaphthalene (DHN) complexes which can be adsorbed at an HMDE. The reduction currents for the complexes are greatly enhanced by coupling a catalytic effect of bromate, and reaction mechanisms are reported. Several military attack scenarios involve the detonation of explosive in the soil backfill surrounding a target structure. In this study, experiments and numerical simulations were performed to determine if the enhanced mechanical properties of geogrid-reinforced soil backfills could mitigate blast effects from buried explosions. The dynamic response of an explosively-loaded, geogrid-reinforced sand was successfully characterized, identifying and quantifying certain blast mitigating effects. Six high explosive experiments were conducted, each involving a 0.91 kg nitromethane explosive charge placed at 76.2 cm depth within a test pit of compacted fill. The concept of using prefabricated vertical drains (PVDs) for remediation of contaminated sites with fine-grained soils is examined. The PVD system is used to shorten the drainage path of the groundwater flow and promote subsurface liquid movement expediting the soil flushing process. The use of PVDs in the current state of practice has been limited to soil improvement. The use of PVDs under vacuum conditions is investigated using study soils consisting of 100% sand and 80% sand/20% kaolinite mixture.
