The placenta plays an important role in modulating xenobiotic passage from mother to fetus. Smoking is known to induce cytochrome P450 1Al (CYP1A1) expression in the placenta, however its effect on P-glycoprotein (P-gp) and ABC transporter in placenta (ABCP) levels in placenta is not known. In-vitro studies in hepatocytes have reported induction of P-gp mRNA by some polycyclic aromatic hydrocarbons. To examine whether smoking can affect placental P-gp and ABCP, their expression and activity were determined in placentas from smokers and non-smokers. Despite the wealth of research conducted on gypsy moth [(Lymantria dispar L.)(Lymantriidae: Lepidoptera)] populations dynamics, quantitative analysis of the native lepidopteran community in which gypsy moth has become naturalized is extremely limited. This study examined the population dynamics of native Lepidoptera in two gypsy moth management areas in West Virginia and Virginia. Data were collected between 1995 and 2001 on 18 plots distributed on two national forests (Monongahela National Forest, WV and George Washington National Forest, VA). Largemouth bass provide an important recreational fishery in the Ohio River. Our objectives were to determine critical over-wintering and spawning habitats of largemouth bass in the Belleville Pool of the Ohio River. We surgically implanted radio-transmitters in 39 adult largemouth bass and tracked them over a 23-month period. Our results demonstrate the importance of off-channel habitats in the life history of largemouth bass in large river systems. Sedimentation, resulting in a loss of embayment quality and surface area, is an important problem and thus merits increased attention. Restoration and protection efforts to improve largemouth bass fisheries in large river systems …

A variety of extracellular signals are transmitted into the cell interior by interactions with a superfamily of heptahelical cell surface receptors. Heterotrimeric G proteins mediate the signal transduction by coupling these receptors to intracellular effector proteins. The molecular mechanisms in G protein-receptor coupling processes are still not completely understood. In this project, two aspects of G protein-receptor coupling were examined: (1) the G protein-coupling properties of the five human muscarinic acetylcholine receptors (mAChRs); (2) a novel regulatory mechanism by a newly identified G protein signal regulator, Activator of G-protein Signaling 3 (AGS3). The most common NPS pollutants are sediment (i.e. total suspended solids, TSS) and nutrients (i.e. total nitrogen, TN). Neotyphodium lolii is a fungal endophyte of perennial ryegrass (Lolium perenne). Like related endophytes of other grasses, it increases the grass host's fitness. Endophyte-infected grasses have greater resistance to biotic and abiotic stress, and thus are highly competitive. However, the endophytes also produce toxins that cause toxicoses in grazing animals. N. lolii produces a large amount of lolitrems, which are thought to cause ryegrass staggers in mammals. The fungus also produces a smaller amount of a highly toxic ergopeptine ergovaline. The first section of this work addressed the relationship between apoptotic and necrotic cell death and their role in inflammatory lung injury induced by endotoxin. Pulmonary administration of lipopolysaccharide (LPS) into the lung of mice caused a time-dependent increase in the levels of pro-inflammatory cytokine TNF-α and inflammatory cell influx in the bronchoalveolar lavage (BAL) fluids. Control mice showed only resident alveolar macrophages with no apoptosis as determined by terminal deoxyribonucleotidyl transferase-mediated dUTP nick end-labeling (TUNEL) and histone-bound DNA fragmentation assays. The Effects Of Superimposing The Same Vs. Qualitatively Different Food On Responding Maintained By A Variable-Interval Schedule Of Reinforcement., Jessie-Sue Milo

This study examined the effects of response-independent delivery of same vs. qualitatively different food than the reinforcer on rats' lever pressing. During baselines, a variable-interval schedule delivered either pellets or milk. A fixed-time schedule then was superimposed which delivered either response-independent pellets or milk. The order of reinforcer and responseindependent food presentation was counterbalanced across subjects, such that in four different conditions each rat was exposed to each combination of reinforcer and response-independent food presentations. Similarity of the response-independent food to the reinforcer did not differentially disrupt response rates. Response-independent milk decreased response rates more than response-independent pellets. The objective of this study was to identify an effective internal parasite control for sheep managed under USDA's NOP. The case study involved the sheep flock of the WVU Organic Farming Systems Experiment over a period of 27 months, March 2001 to July 2003. A 3-day-duration/5-week-rest grazing regime was implemented to control internal parasites. The parasite burden was measured by fecal analysis (fecal egg counts). Prior to implementation of the prescribed grazing regime, parasite levels were moderate and comparable to conventionally managed flocks in the local area. Story telling is universal, and the significance of stories may lie in their ability to guide behavior. An informal analysis of story telling suggests that stories provide a model of behavior, establish motivation, and stimulate the development of self-generated rules. In psychotherapy, stories illustrate points, suggest solutions, improve client insight, increase motivation, and establish rapport (Barker, 1996; Zeig, 1980). Despite the popular use of stories across theoretical orientations and treatment approaches, it is not clear if stories facilitate behavior change. The present study was conducted to examine gender and gender role differences in cardiovascular reactivity and confidence in relation to performing gender role-oriented exercise tasks. Sixty-four participants were selected to view videos of male and female models performing two exercise tasks [baton twirling (the feminine gender role-oriented task) and martial arts (the masculine gender role-oriented task)]. Participants then completed the exercise tasks while following along with the same video models. Heart rate (HR) and systolic and diastolic blood pressure (SBP and DBP) were measured during experimental conditions. Study one investigated the ability of uric acid (UA) to scavenge reactive oxygen species (ROS) and the ability of UA to protect against ROS-mediated damage. Hydroxyl and superoxide radical ESR signals were reduced by addition of UA (P< 0.05). UA prevented lipid peroxidation in RAW 264.7 cells exposed to silica (P< 0.015). UA inhibited hydroxyl-mediated damage to λ Hind III DNA. The objective of study two was to determine the effect of changes in plasma UA (PUA) on growth and ascites development in broilers. Chicks were fed a grower diet supplemented with 0.5 moles inosine/kg feed to raise PUA, 15 mg allopurinol/kg body weight/day to lower PUA, or a control diet. Birds were housed in two environmental chambers; one had conditions to promote ascites while the second was held at standard conditions. Although dietary treatment had an effect on PUA, no effect of diet on performance or development of ascites was observed.

Little is known about nest tree use of the endangered Virginia northern flying squirrel. I compared characteristics of nest trees used by Virginia northern flying squirrels with randomly selected trees during summer and fall of 2000 and 2001. I tracked 13 squirrels to 59 nest trees. Sixty-nine percent of the nests were in cavities and 31% were leaf nests. Yellow birch, black birch and American beech were selected for nest trees more than expected. Nest trees were in larger and taller trees than trees in the surrounding area. There were a significant number of trees located near skidder and hiking …

The goal of this study was to evaluate the relative success of mitigation wetlands in West Virginia in supporting vegetation, invertebrate, and wildlife communities. Eleven mitigation wetlands were compared to 4 naturally occurring reference wetlands. For all vegetation species sampled, species richness (no. species/quadrat; P = 0.035), evenness (P = 0.033), and diversity (P = 0.025) were higher in mitigation than reference wetlands. Mean weighted averages per quadrat were similar between mitigation and reference wetlands (P = 0.242). Differences in vegetation composition between wetland types were reflected through ordination using Detrended Correspondence Analysis (DCA). Both mitigation and natural wetlands met …

This analysis uses economic modeling techniques to analyze direct farm marketing sales, profits, and public policy in West Virginia. To investigate factors that affect county direct farm marketing, the impact of consumer demographic, land, and spatial variables upon county direct farm marketing sales is tested. The results reveal that income, education, land characteristics, and adjacent metropolitan areas influence direct farm marketing sales. An Input-Output (I-O) model is used to analyze a direct farm marketing public policy in the Mountain State. Neotyphodium spp. are members of the fungal family Clavicipitaceae that grow endophytically within several important forage grasses, imparting benefits to the host grass. Several species produce toxic ergopeptines that are thought to adversely affect mammalian health. Inclusion of 1% agar in endophyte-infected seed diets was found to ameliorate some of the consequences of feeding endophyte-infected seeds to mice. Based upon feeding preferences of rabbits and mice, it seems that ergot alkaloids are not critical factors for deterring feeding on endophyte-infected plants by these animals, but the presence of the fungus appeared to deter rabbits. Inputs of leaf litter from riparian zones are the primary energy source for secondary production in these streams and inputs of large woody debris (LWD) help shape stream habitat. This thesis examines the interaction between the Innu of Labrador, an indigenous people in Canada, and the state since the middle twentieth century. The changing spatiality of this interaction can be understood through the analysis of the politics of scale. The scale of Innu social practices has historically been wide-ranging and fluid. Since the 1950s, the state has been increasingly involved in development projects on the lands the Innu have traditionally occupied. Surface mining disturbs thousands of hectares of forest every year in the Appalachian region resulting in the creation of forest edges. In this study, variability in air temperature and moisture, soil temperature and moisture, photosynthetically active radiation and wind velocity are quantified for open, edge and interior environments of reclaimed surface mines in north central West Virginia. Tree species composition, soil pH and compaction were also examined. Pulmonary fibrosis due to acute or chronic occupational exposures is the leading cause of work-related illness in the U.S. according to the 2002 report by the American Lung Association. Asbestosis and silicosis are well described lung diseases that result from asbestos fiber or crystalline silica inhalation, respectively. This project examines the sexual content in top-rated shows amongst teenage audiences. Cultivation theory and social learning theory (SLT) are the basis of the study to help determine what impact sexual portrayals on television have on this young audience. Scripts from the top shows from one television season were examined to determine first, how much sexual content existed in the scripts, and second, what type of sexual content it was in the form of gender-based dyadic communication. Analysis of responses from (N = 64) undergraduate students enrolled in Learning Strategies and Orientation courses on the Career Decision Making Self-Efficacy scale and the Occupational Self-Efficacy Scale, revealed that participants who selected male-dominated careers scored higher on the goal selection subscale compared to participants in the female-dominated group. Similar results were observed for career decision-making, self-efficacy and major status. As predicted, the scores for males on the OSES indicated that males reported equally higher scores for male and female-dominated occupations, whereas females reported higher self-efficacy for female-dominated occupations compared to male-dominated occupations.

Dopaminergic agonists such as L-dopa may improve visual sensitivity in subjects with abnormal vision, and possibly in subjects with normal vision. The aim of this study was to evaluate the effects of L-dopa on the contrast sensitivity function (CSF) psychophysically and physiologically in 14 healthy subjects. Subjects received either 250mg L-dopa/25mg Carbidopa or placebo. Functional magnetic resonance imaging (fMRI) and perceptual performance data was collected while subjects viewed stimuli over a range of spatial frequencies, at threshold (barely visible) contrast.
