Mass Spectrometry Of Metallothionein Adducts As Candidate Biomarkers Of Styrene Oxide And 1-Phenylpropylene Oxide, Sandra G. Tarr

Metallothioneins (MT) are cysteine-rich intracellular proteins that sequester epoxides, such as the genotoxic styrene metabolite, styrene oxide (SO). Covalent adduct formation by the ring-opening of SO and its homologue, 1-phenylpropylene oxide (trans), with MT forms stable complexes that potentially may serve as biomarkers for epoxide exposure. MT was reacted with SO and trans-phenylpropylene oxide (PPO) separately in water. Mass spectra of adducts were obtained using electrospray ionization with positive-ion detection in an ion trap mass spectrometer. MS/MS analysis established the covalent nature of the complexation. Histone acetylation is modulated through the action of histone acetyltransferases and deacetylases, which play key roles in the regulation of eukaryotic gene expression. The RPD3 group of histone deacetylases constitutes the first class of histone deacetylases (HDACs) in eukaryotes.;In yeast and mammalian cells, it was found that RPD3 histone deacetylases are involved in the ageing and development. In contrast much less is known about the function of RPD3s in plant. This research is an investigation of how remote sensing can be used to supplement traditional methods of corridor management planning. Cultural resource inventory for corridor and interpretive management practices is an important component of applications for the designation of a selected route as Scenic Historic American Byways through the National Scenic Byways Program. A case study of a section of the National Road near Wheeling, West Virginia, showed that contemporary and historic aerial photography can facilitate the identification of possible impacts on a route’s historic integrity. During the seventeenth century a vibrant group of Native Americans, the Monytons, controlled southern West Virginia and northeastern Kentucky. The effects of contact with Europeans destabilized their societies as far inland as the Ohio Valley. This began the process which eventually pushed most Monytons from the region. The Five Nations Iroquois, pressured by social changes farther north, attacked the Monytons further drawing people out of the Ohio Valley. A growing southern Indian slave trade also contributed to the decay of Monyton dominance in the region. A central point in this thesis is that Monytons formed and reformed traditions to deal …

ERFs (ethylene-responsive element binding factors) belong to a large family of transcription factors that are found exclusively in plants. A small subfamily of ERF proteins can act as transcriptional repressors. The Arabidopsis genome contains eight ERF repressors, namely AtERF3, AtERF4, AtERF7, AtERF8, AtERF9, AtERF10, AtERF11, and AtERF12. The different ERF repressors show differential expression, suggesting that they may have different function. The expression of AtERF4 can be induced by ethylene, jasmonic acid, or abscisic acid. AtERF7 can be induced by abscisic acid only. Using a transient expression system, we demonstrated that AtERF4 and AtERF7 can function as transcriptional repressors. By …

Exoenzyme S (ExoS) is a bifunctional toxin that is directly translocated into eukaryotic cells by the Pseudomonas aeruginosa (Pa) type III secretory (T3S) process. The N-terminal region of ExoS includes a GTPase activating protein (GAP) activity and the C-terminal region includes an ADP-ribosyltransferase (ADPRT) activity. The T3S-translocation (T3S-T) of ExoS allows the bacteria to directly manipulate host cell function, thereby contributing the virulence of Pa. A cell culture model of infection was developed to study the effects of ExoS on cell function, which differentiates the effects of ExoS expression from other bacterial effects. The Retinal homeobox gene, Rx, is expressed in the anterior neural plate and the optic vesicle shows Rx expression throughout development. Rx is required for the proliferation of retinal progenitor cells during eye development.;Our expression studies of the Rx gene in the adult mouse retina show Rx expression in the Photoreceptor and inner nuclear layer of the adult neural retina and ciliarybody. The cotransfection assays indicate that Rx supports the transcription of neural retinal markers, as well as the RPE-markers. Recent surveys have indicated that populations of fishes and crayfish are small in the Stony River, Grant Co., WV. The Stony River is influenced by thermal enrichment and acid mine drainage (AMD) within a 13 km stream reach, downstream of Mt. Storm Lake. In an attempt to determine the influence of each anthropogenic disturbance on these affected taxa, studies of their distribution and survival were undertaken. A crayfish trapping survey was conducted along a gradient of thermal and AMD influences in the Stony River system. This paper presents the findings of an investigation to determine whether of not there is a difference in hospitalized adolescents and non-hospitalized adolescents. The participants were 60 adolescents between the ages of 12 to 18 years. Their fears, stressors, and coping skills were evaluated by using three instruments: a 35-item self-report test on coping; a 108-item self-report test on fears; and a 15-item self-report test on stress. The results revealed that on specific measures hospitalized adolescents exhibited a statistically significantly less degree of emotional upset during hospitalization than did the community sample. The purpose of this project was to implement the West Virginia Dairy Quality Assurance Program (WVDQAP) which promotes best management practices to produce high quality milk and meat products. The genes of the endothelin system are the potential targets for regulation by prostaglandin F2α (PGF2α) and have been studied in greater detail. The effects of a luteolytic dose of PGF2α on the mRNA encoding endothelin converting enzyme-1 (ECE1), pre-pro endothelin-1 (ET-1) and the ET receptors ETA, ETB, and in bovine corpus luteum (CL) collected during the early, mid or late luteal phases were examined. Low grain prices and recurrent periods of drought have elicited interest in early weaning. Calves can efficiently utilize feed for weight gain when it is fed directly, rather than through their dams. This study evaluated calf response in three weaning systems (experiment 1) and profiles of body energy reserves in early-weaned beef cows (experiment 2). Treatment x year, and calf sex x year interaction (P < 0.05) were observed for calf weight gain. These interactions underscored the need for constant readjustments in management systems that incorporate early weaning in their programs. In experiment 2, body weight and condition score of cows were affected marginally (P = 0.06) by early weaning, whereas ribfat and rumpfat differed (P < 0.05) between earlyweaned and control cows. These findings indicated that subjective methods (body weight and condition score) should be used in combination with more objective measurements (ribfat and/or rumpfat) for accurate evaluation of energy status of beef cows. Cows of various ages responded differently to early weaning. Three to five-year-old cows exhibited greater response to early weaning as opposed to 2-year-olds and cows > 5 years of age, indicating that profit potential of the cow-calf operation can be improved by altered management of middle-aged cows.

Roads and culverts have many effects on stream salamanders. We examined 120 culvert crossings in the Shavers Fork and Dry Fork watersheds in eastern West Virginia. Complete barriers to salamander passage occurred at 55.0% of the sites visited and partial barriers at 34.2%. Analyses showed that 20.6% of the total stream length in the Dry Fork watershed and 18.4% in the Shavers Fork watershed were isolated by barriers. We conducted salamander sampling at 16 streams and used Akaikeís Information Criterion to select the best a priori models explaining salamander diversity, richness, and abundance. I investigated whether seedlings of non-agricultural plants exposed to varying levels of UV-B exposure had altered growth, physiological, and tissue chemistry responses under changing environmental conditions. In a field experiment, using a modulated UV-B lamp bank system, elevated UV-B did not significantly alter the total biomass of the 6 plant species that were studied, and only Quercus rubra showed a trend towards some sensitivity to UV-B. In two species—Platanus occidentalis and Setaria faberii—I found an increase in the production of UV-B absorbing compounds under elevated UV-B while two grasses—S. faberii and Elymus virginicus—had significantly decreased C:N ratios under elevated UV-B. …

Cytochromes P450 comprise a ubiquitous superfamily of heme-thiolate enzymes, considered among the most versatile biological catalysts known. Structure-function relationships of P450s are of particular interest to those attempting to elucidate such aspects of xenobiotic metabolism as substrate selectivity and regiospecific oxidations. We studied two members of the P450 1A subfamily, P450 1A1 and 1A2, found in highest abundance in lung and liver tissues, respectively. Rx is a paired-like homeodomain transcription factor that is expressed in the developing eyes, ventral forebrain, hypothalamus, posterior pituitary, and the pineal gland. In adults, Rx is primarily expressed in the neural retina, ciliary bodies, pineal gland, hypothalamus, and posterior pituitary. Rx mutations in multiple species are associated with anophthalmia. During the course if this work, we identified a human patient with anophthalmia and sclerocornea that carries mutations in the RX gene. CYP1A1 metabolizes polycyclic aromatic hydrocarbons (PAHs), such as those in cigarettes, to reactive intermediates which interact with DNA and lead to cancer. Silica is a lung carcinogen. Epidemiology studies of silica and lung cancer are not all positive. A possible explanation for these inconsistencies may be that silica is a modifier of PAH metabolism and, thus, cigarette smoke carcinogenesis. We hypothesize that crystalline silica exposure alters CYP1A1 expression, thereby modifying lung cancer risk. Prechondrocytes exhibit a change in their actin cytoskeletal arrangement when subjected to loading. This study looks at the implications of the altered solid-fluid interactions due to this change. A 2-D Finite element model of a cell was developed. A solid-fluid interaction solution procedure was validated and used to determine fluid flow/pressure in a cell when subject to deformation. These values were then used to calculate fluid flow induced stresses on individual actin fibers. The results suggest that prechondrocytes alter their cytoskeletal structure to reduce stresses as a result of fluid flow due to deformation of the cell.

Nutrition and reproduction are linked in that the nutritional status of an individual influences its reproductive performance, i.e. an individual in negative energy balance will have decreased ability to reproduce. The mechanisms in which nutrition influences reproduction are not understood. My data suggest that reductions in lutenizing hormone secretion resulting from photoperiod-induced increases in sensitivity to testosterone negative feedback may be mediated, at least in part, by elevated neuropeptide Y (NPY), a hypothalamic neuropeptide shown to alter gonadotropin releasing hormone release and potently affect food intake. Historically, hypovirulent strains of Cryphonectria parasitica have been introduced into cankers principally by inoculating wounds made to the margin of cankers. Twelve hypovirus introduction methods were tested. Seventy-two trees were inoculated in June 2002 with an orange-pigmented virulent strain of C. parasitica (two inoculations per tree). After 11 weeks, the resulting cankers were treated with a brown-pigmented hypovirulent C. parasitica isolate using one of the following methods: (1) a non-invasive treatment where the inoculum was painted onto the canker surface; (2) an invasive treatment where the canker face was wounded with a sharp blade prior to the painting application of …

Natural Gas Exploration Associated With Fracture Systems In Alleghenian Thrust Faults In The Greenbrier Formation, Southern West Virginia, Craig A. Edmonds

A hydrocarbon play has been identified in southern West Virginia targeting the intersection of thrust faults with specific Mississippian reservoirs. Late Paleozoic sandstone and limestone reservoirs are traditional pays of the study area, often yielding economic natural gas production, with initial production rates ranging between 100 to 200 Mcf per day per well. In contrast, production rates from wells that encounter faults often exceed 500 Mcf per day. We believe this prolific production results from secondary fracture porosity and enhanced permeability from fracture zones associated with thrust faults, providing a conduit to the Union Oolite reservoir member of the Greenbrier …
