The objectives of this study were to compare the performance of two feed treatments on a population of rainbow trout, Oncorhynchus mykiss, reared in a linear raceway system, which utilized mine water from a mine water treatment facility (MWTF); to measure and compare waste outputs; to monitor the water quality of the system; and to identify the impacts, if any, of the water quality on the fish population. In a two-year study, eight vegetable crops were grown as part of a certified organic four-year crop rotation. High input plots received off-farm composted cow manure and cover crops, and low input plots were managed with the use of cover crops only. High input plots produced higher yields than low input plots in all crops, except peas, which yielded higher in low input plots. Soil organic matter levels were significantly higher in the high input than the low input plots and increased from 2003 to 2004 in both treatments. Available soil phosphorus levels in the high input plots were excessive, …

Three experiments were conducted to test hypotheses relative to the ability of gonadotropin releasing hormone (GnRH) and melatonin to improve responses of anestrous ewes to rams. Treatment with GnRH two days after treatment with progesterone at introduction of rams did not increase ovulation, pregnancy or lambing rates. Treatment with GnRH on days two, seven, or both after introduction of rams, resulted in ovulation, pregnancy, and lambing rates that did not differ. In another trial, GnRH four days before, or four days before and one day after introduction of rams did not improve a consistently high ovulatory response to introduction of …

Ailanthus altissima (Mill.) Swingle (tree of heaven) is an exotic species from China that was intentionally introduced as an urban ornamental species but has since become naturalized throughout much of the U.S. This pioneer species appears poised to invade rural areas as current rates of human-induced forest disturbance lower dispersal barriers and increase available habitat, especially in eastern forests, where it will compete with pioneer native trees such as Liriodendron tulipifera (L.) The current experiments were conducted to determine if an orally active progestin, melengestrol acetate (MGA), would be an effective method to induce molting that addresses hen well-being during and after molt. MGA can easily be administered to a large population because it is orally active and can be included in a balanced layer diet. Hens could be fed throughout the molting process, which would address the concerns brought about by other methods in that there is a nutrient deficiency that results in hen weight loss. Noninvasive functional magnetic resonance imaging (fMRI) methods may provide new ways to detect and track renal hemodynamic changes in-vivo. In this thesis, two fMRI methods were correlated with simultaneous invasive hemodynamic measurements. A particular goal of this thesis was to measure the relative contributions of oxygenation and perfusion changes to changes in the relaxation rate, T2*. Also, an MRI compatible motion detector was built and along with the invasive probes was interfaced to a data acquisition system for use during scanning. Spirochetes are unique bacteria that cause a variety of diseases. Motility, provided by periplasmic flagella (PFs), is a virulence factor for spirochetes. This work focuses on PFs from Borrelia burgdorferi and Brachyspira hyodysenteriae. PFs consist of three main parts: basal body, hook, and filament. Monoclonal antibodies were developed to attempt identification of individual FlaB filament proteins in B. hyodysenteriae .;The PF filament of B. burgdorferi consists of a major flagellin, FlaB, and a minor flagellin, FlaA. Sprawl can be defined as dispersed, automobile-dependent development typically found along roadways outside of urban downtown areas.;Because of the problems associated with sprawl, planning and design guidelines have been developed for certain major land uses that offer guidance on more sustainable development practices. The purpose of this study was to identify characteristics of farmers' market vendors in West Virginia, including products, season extension methods, promotional strategies, and sales levels, as well as to identify the obstacles and educational needs relevant to farmers' market vendors. While the majority of products produced by vendors were vegetables, it was found that products such as processed products and live plants were used to extend marketing seasons. Meat and dairy were the least represented categories. A study to evaluate lamb production under organic management was carried out in 2003 and 2004 at the West Virginia University Organic Research Farm. About 0.93 ha of tall fescue-orchard grass-red clover grassland was assigned to three blocks based on soil type, slope and previous treatment. This paper discusses three research studies, each investigating a different treatment window for controlling tree-of-heaven using effective and environmentally sound herbicide applications. In the first study, efficacy of the herbicides triclopyr and imazapyr was tested using injection and basal bark treatment methods. In the second study, efficacy of triclopyr with different levels of imazapyr was tested using the basal bark treatment method. In the third study, weekly timing of chemical controls was investigated. In our research, the quality of a number of calcium supplements was evaluated by their physical properties, disintegration and dissolution profiles. Physical tests and disintegration did show a pH sensitivity. Disintegration confirmed the good uniformity of the products. In the USP required dissolution test, most of the products did not achieve acceptable drug release in the specified time period. While formulation factors and the salt form of calcium supplements can to some extent affect their dissolution pattern, it was ultimately considered that the failure may be related to the poor solubility of the calcium salts.;Based on the results and the …

Upland pastures in West Virginia receive low-input management. The effect of manure addition to upland pasture soils was measured with a variety of biological indicators such as soil microbial biomass carbon (SMBC), hot water extractable carbon (HWC), potentially mineralizable nitrogen (PMN) and soil CO2 efflux. The focus of this work was on the design and implementation of a web based forensic information management system for the West Virginia State Police Forensics Laboratory (WVSPFL). The software tool enables law enforcement agents to submit crime information to the Forensic Laboratory via a secure Internet connection. The tool also enables each of the nine units of WVSPFL to process the information electronically. Online electronic forms were created to mirror the existing paper based forms, making the transition easier. The relationship between land cover characteristics and water pollutant concentrations in surface water was evaluated for 10 sub-basins in the Potomac Headwaters Region of West Virginia. Using water quality data collected between 1998 and 2004 by the West Virginia Department of Agriculture (WVDA), mean and median in-stream concentrations of fecal coliform, total phosphorus, and nitrate nitrogen were determined for specific periods of time during the sampling period. Land use categories were evaluated on a basin-wide and a floodplain-specific scale. The percentages of forest, agriculture, cropland and urban land were determined. Numerous studies that have examined the physical health consequences of individual variables have focused on negative traits such as hostility, anger, and stress. These studies have demonstrated that such variables are often related to deleterious health outcomes as well as exaggerated cardiovascular reactions to stress. Fewer studies have examined the health benefits of positive variables like forgiveness. Forgiveness and hostility are thought to have an inverse relation, and it is not surprising that forgiveness has been associated with reduced cardiovascular reactivity to stress. The dissemination of European (Euro-7) and North American (GH2) hypovirulent (H) isolates of Cryphonectria parasitica to American chestnut saplings was investigated. Nine 0.01 ha plots were established in a clearcut. Each of 24 trees per plot received 4 punch- and 1 branch wounds in October, 1985; January, April, and July, 1986. Control plots received a virulent (V) strain of the fungus. H inocula were placed in three plots per treatment, respectively, at 2–4 month intervals. Cankers were sampled over a 2.5-year period. In 1784, the residents of the upper east Tennessee Valley declared themselves independent from their parent state of North Carolina. The political and economic leadership of the newly formed state of Franklin utilized the ideology, symbolism, and rhetoric of the American Revolution to garner local, regional and national support for the movement. After the state's collapse in 1788, the state of Franklin continued to attract supporters and admirers, who considered the movement to be a noble extension of the revolution. Heat shock proteins (HSP's) are expressed in response to various stressors, such as physical exercise, therefore this study investigated the role of these proteins in skeletal muscle remodeling in response to resistance training. First, we determined the time course of HSP response as a result of a single bout of damaging contractions characterized by non-recoverable force deficit. Ethanol and 6-hydroxydopamine (6-OHDA), a Parkinson's disease (PD) mimetic, are two toxins to the cerebellum. This study investigated the molecular mechanisms underlying their neurotoxicities. The work in chapter 2 of this thesis focused on 6-OHDA-induced neurodegeneration. Previous studies indicate that impaired function of the endoplasmic reticulum (ER), commonly referred to as ER stress, is an important factor in the neuropathology of a wide variety of neurological disorders. This study demonstrated that 6-OHDA evoked ER-stress and led to apoptosis in SH-SY5Y neuronal cell line and cultured rat cerebellar granule neurons (CGNs). Furthermore, GSK3beta played a crucial role in mediating 6-OHDA's neurotoxicity. …

We have examined the effects of the cytokines tumor necrosis factoralpha (TNF), transforming growth factorbeta (TGFbeta) and the effect of cyclooxygenase inhibitor ibuprofen on prostaglandin and 15-HETE biosynthesis by normal adult human osteoblast-like (hOB) cells. In the present studies, conditioned media harvested from hOB cell cultures were assayed by eicosanoid specific EIAs to determine quantitatively the effects of cytokines on PGE2, 6-keto-PGF1alpha, PGF2alpha, PGD2, thromboxane (TXB2) and on 15-HETE biosynthesis. Heat shock protein 70 (Hsp70) expression has been reported to be a sensitive biomarker of cell stress. Arsenic (As), cadmium (Cd), mercury (Hg), chromium (Cr), nickel (Ni), vanadium (V), and manganese (Mn) are widely used and exposure to these metals is associated with the development of pulmonary disease. Our data showed that exposure to these metals resulted in a significant increase in cytotoxicity, apoptosis, and generation of reactive oxygen species (ROS) in a human bronchial cell line, BEAS-2B. CK2 is a highly conserved Ser/Thr protein kinase composed of catalytic (alpha) and regulatory (beta) subunits. The enzyme targets proteins that are involved in a variety of cellular processes such as DNA replication, transcription, translation, cell cycle progression, and development. However, very little information is available on either the regulation of this enzyme or its role during development. These two aspects of CK2 have been addressed using the fruit fly, Drosophila melanogaster, as a model organism. Evidence from investigations of genetic differences among human beings shows that genetic diseases are often the results of genetic mutations. The most common form of these mutations is single nucleotide polymorphism (SNP). A complete map of all SNPs in the human genome will be extremely valuable for studying the relationships between specific haplotypes and specific genetic diseases. Some recent discoveries show that the DNA sequence of human beings can be partitioned into long blocks where genetic recombination has been rare. Because of the species diversity, varied site conditions and growth rates, it is really challenging to manage the central Appalachian hardwoods. Examining the harvesting techniques and interactions among stand, harvest, and machines is becoming a concern to the researchers in the region. A simulation system was developed to aid these efforts by estimating the productivity, cost, and traffic intensity of different harvesting configurations under a variety of harvesting prescriptions and stand conditions.;Stands used in the simulation were generated by using the stand generator that was validated by comparing the generated stands with the actual mapped stands statistically. Experiments were established in fall 2003 at West Virginia University to evaluate the effects of incorporation of composted poultry litter (CPL) on subsoil physical and chemical properties, Kentucky bluegrass ( Poa Pratensis L.) establishment, root depth, and weed pressure. Twenty cm of topsoil was removed and CPL was incorporated to a depth of 12.7 cm at 10%, 20%, and 40% v/v prior to seeding or sodding. Composted plots were compared to N-fertilized (454 Kg ha-1) and control plots. Positive linear increases in total water content (thetaT), organic matter, pH, and CEC were observed following compost incorporation with a decrease in …
