Construction And Characterization Of Lac Containing Plasmids And The Transformation Of Escherichia Coli And Saccharomyces Cerevisiar With These Plasmids, Bradley J. Kleinsasser

Electronic Theses and Dissertations

The science of genetic engineering or direct genetic manipulation required the availability of four basic procedures. These procedures are: i) a way to cut and join DNA molecules, ii) a carrier or vector for genes that can replicate both itself and the foreign DNA it harbors, iii) a way of gaining entrance for this carrier and its foreign DNA into a host cell, and iv) a way of selecting the cells which had received the chimera DNA from among a large cell population which had not. The lac operon of E. coli is a segment of DNA on the chromosome …

An Economic Analysis Of The South Dakota Personal Property Tax Replacement Formula, Ann Louise Kuyper Jan 1984

An Economic Analysis Of The South Dakota Personal Property Tax Replacement Formula, Ann Louise Kuyper

Electronic Theses and Dissertations

In 1978, after years of considerable pressure for tax relief, the South Dakota Legislature repealed the personal property tax. The repeal took place in two stages. In 1979 the tax on household goods, personal effects, appliances, sporting and hobby goods, pets, grain and seed, and honey and sugar beets was repealed. The remainder was repealed in 1980. Personal property tax collections in 1977 accounted for approximately 20.2% of total property tax collections in South Dakota. Because the personal property tax was a significant portion of local government revenues, there was no question at the time that the repeal of the …

Ability Of Klebsiella Pneumoniae To Fix Nitrogen Utilizing Selected Energy Sources, Theresa Helen Lemme Jan 1984

Ability Of Klebsiella Pneumoniae To Fix Nitrogen Utilizing Selected Energy Sources, Theresa Helen Lemme

Electronic Theses and Dissertations

There are many nutrients in a farming operation which can limit production. Nitrogen deficiencies are found more often than any other. Crop yield increases needed to meet world population growth result in a greater demand for nitrogen. The commercial chemical manufacturer of fertilizer nitrogen by the Haber-Basch method is very energy intensive. Biological nitrogen fixation as an alternative source of nitrogen is now being extensively studied. The legume-rhizobium symbiosis has been well documented. This symbiosis has occurred for centuries yet the knowledge of its process and how it helps to decrease fertilizer nitrogen requirements was welcomed by the agricultural community. …

A Study Of The Effects Of Gender And Judging Experience In Lincoln-Douglas Debate, Charles Alan Lubbers Jan 1984

A Study Of The Effects Of Gender And Judging Experience In Lincoln-Douglas Debate, Charles Alan Lubbers

Electronic Theses and Dissertations

Many people still recall the years when contestants in debate and individual events were divided into a "men's" and "women's" division. It is just in the last decade that some high school forensic tournaments in South Dakota have abolished this practice of segregation and have combined the two divisions into a single all-encompassing division. The original rationale for such a practice of separation is not immediately evident. Apparently, the tournament directors believed that one sex had an inherent advantage over the other in equal competition. One event that had not yet been initiated during this period of segregation was the …

A Morphometric Analysis Of Ultrastructural Changes In The Shoot Apex Of Glycine Max During The Morphogenetic Shift From Quiescence To Germination, Saba Mebrahtu Jan 1984

A Morphometric Analysis Of Ultrastructural Changes In The Shoot Apex Of Glycine Max During The Morphogenetic Shift From Quiescence To Germination, Saba Mebrahtu

Electronic Theses and Dissertations

This research is a study of a morphogenetic shift of the soybean shoot apex from quiescence to germination. The minute changes in the organelles were recorded with the use of stereological methods. The ultrastructural data was statistically analyzed. The ultimate goal was the recognition of relationships between the cell's functional units and its structural units, with the identification of morphogenetic regulation mechanisms. Morphometric image analysis of cellular organelles indicated that storage reserves were transformed from lipid bodies to plastid starch. Proplastids and promitochondria, so difficult to differentiate in early stages of germination became obvious by 96 hours post germination. Significant …

Inheritance Of Resistance In Corn To Leaf Freckles And Wilt Incited By Corynebacterium Nebraskense, Edward N. Ngong-Nassah Jan 1984

Inheritance Of Resistance In Corn To Leaf Freckles And Wilt Incited By Corynebacterium Nebraskense, Edward N. Ngong-Nassah

Electronic Theses and Dissertations

Resistance enables crop plants to defend themselves against their pathogens or against a level of disease deleterious to the crop Frequently, the relative success of other control measures is determined by the precision of their use. Resistance, however, is not so subject to external variation. Concerns for the quality of the environment when chemicals are used to control disease illustrates the importance of genetic control of plant diseases. Whereas the protection of crops of high unit value (fruit orchards, vegetables, ornamentals) by the use of fungicides is considered profitable, hereditary control remains the most economically feasible strategy to protect crops …

The Effects Of Sex And Delayed Chill On The Biophysical And Organoleptic Properties Of Intact And Restructured Beef Steaks, Bruce Colin Paterson Jan 1984

The Effects Of Sex And Delayed Chill On The Biophysical And Organoleptic Properties Of Intact And Restructured Beef Steaks, Bruce Colin Paterson

Electronic Theses and Dissertations

The meat industry presently utilizes large, inefficient storage coolers for cooling carcasses rather than more efficient systems for cooling edible boneless meat. When these facilities are combined with the practice of chilling, reheating and rechilling tons of product each day, the need for efficient processing systems becomes evident. It is this inefficiency which has led to hot processing. Hot processing is the removal of bone and trim prior to chilling and it has several possible economic benefits: (1) removal of bone and trim prior to chilling which reduces the energy required to chill; (2) more rapid chilling; (3) decreased refrigeration …

S.I. Hayakawa And The Use Of Danger Indicators Of General Semantics, Janine Rae Rudnick Jan 1984

S.I. Hayakawa And The Use Of Danger Indicators Of General Semantics, Janine Rae Rudnick

Electronic Theses and Dissertations

Many books and articles have been written about general semantics and specific phrases included in general semantics study. The books generally have a theme of the meaning of words. Very few books or articles, however, have stressed the importance of J. Samuel Bois' danger indicators as part of the meaning of words. "Ego statements" 11 and "either or" 12 indicators have been touched upon in books and articles, but no one has dedicated significant study to the specific danger indicators as presented by Bois. It is this writer's belief that the danger indicators of general semantics should be studied more …

Patient Demand For Nursing Home Care, Kenneth Ray Spear Jan 1984

Patient Demand For Nursing Home Care, Kenneth Ray Spear

Electronic Theses and Dissertations

A great deal of attention has been devoted in recent years to the rising costs of health care and to the resulting pressures upon private and public budgets. Likewise, much attention has been focused on the economic well-being of the increasing number of older citizens. Surprisingly, the tools of economics have been infrequently applied to the nursing home industry, which is a growing segment of the health care sector that now provides services to over one million elderly persons annually (U.S., Department of Health and Human Services, 1981). Since economics is concerned with scarce resources and their efficient allocation among …

Engineering And Economic Analysis Of A Solar Energy Intensifier-Thermal-Energy Storage System For Agriculture, John Van Zweden

Electronic Theses and Dissertations

World fossil fuel depletion and the unpredictability of foreign sources have increased the desirability of energy independence in the United States. Rising fuel prices affect every segment of the economy. Agriculture consumes only 2.5 percent of the total national energy consumption, but an uninterrupted supply of inexpensive fuel is vital to the success of this important industry. Solar energy, because it is one of the only energy incomes of the earth, holds great promise as a future energy source. For solar energy to become widely adopted as a source of alternate energy, solar systems must be developed that are both …

Intensive Culture Of Largemouth Bass And Walleye Fry In Experimental Systems, Gerald A. Wickstrom

Electronic Theses and Dissertations

Six genera of invertebrates were made available to largemouth bass (Micropterus salmoides) fry for 25 days. Brachionus spp. was selected for during both day and night (1100 and 2300 h) for the first seven days. Cyclops vemalis was selected on days 1 - 14; it was selected for more during the day for days 1 - 7. Daphnia pulex and D. magna were selected for during days 15 - 25. As the fry grew during the study they ate significantly (P < 0.05) larger D. pulex and D. magna. Moina brachiata was not selected for; it was eaten throughout the study in approximately the same proportion as available. Diaptomus spp. And Ceanestheriella setosa were not eaten. The fry increased in mean length from 6.5 mm on day 1 to 29.8 mm on day 25 of the study. Mean daily length increase was 0.93 mm. Newly hatched walleye (Stizostedion vitreum vitreum) fry were intensively cultured on a diet of live zooplankton under three conditions for 48 days. The fry were reared in tanks under constant illumination (375 lux), in tanks under reduced illumination (105 lux), and in tanks which contained visual interceptors (400 lux). Mean survival of walleyes was 14.0% in tanks under constant illumination, 11.6% in tanks under reduced illumination, and 4.7% in tanks which contained visual interceptors. Mean unaccountable mortality of walleyes was 21.8% in constant illumination tanks, 35.7% in reduced illumination tanks, and 27.4% in tanks with visual interceptors. The fry increased in mean length from 8.6 mm at the start of the study to 36.3 mm (0.76 mm/day) in constant illumination tanks, 36.6 mm (0.76 mm/day) in reduced illumination tanks, and 33.7 mm (0.70 mm/day) in tanks with visual interceptors after 48 days of intensive culture.

The Effects Of Limited Energy Supplementation On Pasture And Level Of Dietary Energy In The Feedlot On The Finishing Performance And Carcass Traits Of Beef Cattle, Mark J. Goetz Jan 1984

The Effects Of Limited Energy Supplementation On Pasture And Level Of Dietary Energy In The Feedlot On The Finishing Performance And Carcass Traits Of Beef Cattle, Mark J. Goetz

Electronic Theses and Dissertations

Traditionally, pasture and range have been the major source of nutrients for grazing animals. The manner by which ruminants survive and thrive on range with no additional nutrients other than those supplied by forage is unique in the animal kingdom. Nevertheless, western man has cultivated a taste for grain-finished beef. To meet that demand, modern beef production involves both the grazing of forages and the feeding of rations high in concentrates. Major disruptive changes, either physical (as in prolonged drought) or economical (fluctuation in market prices) encourage the reexamination of traditional systems. Within the last 10 years, extreme fluctuations in …

Price Relationships Of Selected South Dakota Cash Corn Markets With Destination And Futures Markets, Steven P. Gunn Jan 1984

Price Relationships Of Selected South Dakota Cash Corn Markets With Destination And Futures Markets, Steven P. Gunn

Electronic Theses and Dissertations

The United States grain marketing system often is considered to be an excellent example of a competitive market. Tomek and Robinson have argued that the basic assumptions of a competitive market are perceived to exist in the grain markets. Grain commodities of a specific grade are homogeneous in characteristics. In many markets the total number of buyers and sellers is large enough to prohibit a single firm from influencing grain prices over an extended period of time. Although government actions restrict price movements in the grain markets, prices are allowed to fluctuate free of government intervention within a specified range. …

A Market Research Study For A South Dakota Weekly Newspaper The Mobridge Tribune, Jan Laughlin Jan 1984

A Market Research Study For A South Dakota Weekly Newspaper The Mobridge Tribune, Jan Laughlin

Electronic Theses and Dissertations

This thesis attempts to solve the dilemma by presenting a model for an in-house market research study for a weekly newspaper that cannot afford an outside research firm. The following study was developed for one weekly newspaper, the Mobridge Tribune, Mobridge, South Dakota. One area in which newspaper market research can gather information is media competition. This is particularly important with the diversity in media apparent today. Where once the newspaper was the only business on main street, there are now other print competitors like marriage mail, direct mail, shoppers, circulars and suburban editions. The traditional electronic media are additional …

Effects Of Dietary Dehydroepiandrosterone (Dhea) On Body Weight, Food Consumption, And Glucose-6-Phosphate Dehydrogenase (G6phd) Activity In Lethal Yellow (Ay/Aw) And White-Bellied Agouti (Aw/Aw) Mice (Strain 129/Sv), Lisa Deette Staber Jan 1984

Effects Of Dietary Dehydroepiandrosterone (Dhea) On Body Weight, Food Consumption, And Glucose-6-Phosphate Dehydrogenase (G6phd) Activity In Lethal Yellow (Ay/Aw) And White-Bellied Agouti (Aw/Aw) Mice (Strain 129/Sv), Lisa Deette Staber

Electronic Theses and Dissertations

Dehydroepiandrosterone (DHEA) is a non-competitive inhibitor in vitro of glucose-6-phosphate dehydrogenase (G6PDH), the rate controlling enzyme in the pentose-phosphate shunt. This pathway produces NADPH, a necessary co-factor for the synthesis of fatty acids and ribo- and deoxyribonucleotides. Some researchers believe that by decreasing G6PDH activity, DHEA produces an anti-obesity and anticarcinogenic effect in laboratory mice and rats genetically inclined toward obesity and cancer. It is DHEA's anti-obesity effect which lends it to the study of the yellow obese (AY/-) mouse with its peculiar pleiotropic syndrome. Yellow obese AY/AW mice were found to be more susceptible to dietary DHEA's effects than …

An Economic Valuation Of South Dakota Wetlands As A Recreation Resource For Resident Hunters, Craig W. Johnson Jan 1984

An Economic Valuation Of South Dakota Wetlands As A Recreation Resource For Resident Hunters, Craig W. Johnson

Electronic Theses and Dissertations

In spite of efforts to conserve wetlands, intensified agriculture, water projects, and urban development annually diminish the quantity and quality of wetland resources (Weller 1981). Leitch and Danielson (1979) noted that when the discounted value of the returns to drainage exceed drainage costs there is an economic incentive to drain. If present drainage rates continue, Weller (1981) estimated that most wetlands will disappear by the year 2140. Research has only recently focused attention on the need to estimate the public value of wetland benefits and the social costs of drainage (Leitch and Danielson 1979). The disparity between private and social …
