A novel core-shell silica based antimicrobial nanoparticle was synthesized. The Stöber silica shell has been engineered to accommodate copper. Synthesis of the core-shell Cu-silica nanoparticle (C-S CuSiNP) involves preparation of base-hydrolyzed Stöber silica “seed” particles first, followed by the acid-catalyzed seeded growth of the Cu-silica shell layer around the core. Scanning electron microscopy and transmission electron microscopy showed monodispersed, spherical shaped nanoparticles with smooth surface morphology. Characterization of particle size distribution in solution by the Dynamic Light Scattering (DLS) technique was fairly consistent with the electron microscopy results. We report a mechanism based screening technique to rapidly identify eukaryotic topoisomerase I targeting agents. The method is based on genetic tagging of topoisomerase I to immobilize the enzyme on a solid surface in a microtiter well format. DNA is added to the wells and retained DNA is detected by Picogreen fluorescence. Compounds that result in an increase in Picogreen staining represent potential topoisomerase interfacial poisons while those that reduce fluorescence report catalytic inhibitors; therefore, the solid phase assay represents a „bimodal‟ readout that reveals mechanisms of action. The gut microbiota is essential for mammalian health and metabolism. Thus identifying factors that influence the host microbiota is key to understanding the dynamic interplay between the host and its microbiota. Recent studies have shown the effect of chronic exposures of trivalent arsenic [As(III)] in environmentally relevant concentrations on host physiology, however, little is known of how it impacts the gut microbiota.

Hydrazine (HZ) and monomethylhydrazine (MMH) are used extensively as hypergolic propellants at Kennedy Space Center. These highly reactive fuels are considered highly toxic, and potentially carcinogenic. Consequently, the transport, handling, and disposal of hydrazines is strictly regulated to protect personnel and the environment. Currently, KSC generates large volumes of hydrazine-laden wastewater for disposal. This waste is contained and shipped on public highways for subsequent disposal by incineration presenting a potentially catastrophic threat to the environment and the general public in the event of an accidental release. Since the 1970s, recessions have become more frequent and severe, causing major changes to the labor market and the importance of education. This study focuses on the problems for employees with master's degrees and MBAs, and asks three questions: 1) Do employees with graduate degrees have higher earnings than bachelor's degree recipients, 2) Do they experience greater job security as a result of their additional education, and 3) Is graduate school a way to avoid unemployment during a recession? Ordinal logistic regression analysis is conducted and finds that master's degrees and MBAs experience very different outcomes, with much higher returns …

The goal of this study is to better understand the genetic basis of Reading Disability (RD) and Attention Deficit Hyperactivity Disorder (ADHD) by examining molecular G x E interactions with parental education for each disorder. Research indicates that despite sharing genetic risk factors, RD and ADHD are influenced by different types of G x E interactions with parental education - a diathesis stress interaction in the case of ADHD and a bioecological interaction in RD. Family systems providing informal care and support to their aging relatives, who were residing in assisted living systems and receiving formal care were central to this study. A broader understanding of family involvement, with respect to exploring both care-receiver and caregiver roles and relationships, within the regulatory and environmental context of assisted living was sought through a qualitative research process. Today's graphical processing units have hundreds of individual processing cores that can be used for general purpose computation of mathematical and scientific problems. Due to their hardware architecture, these devices are especially effective when solving problems that exhibit a high degree of spatial locality. Cellular automata use small, local neighborhoods to determine successive states of individual elements and therefore, provide an excellent opportunity for the application of general purpose GPU computing. There is longstanding tension in the study of religion between those who believe religion can be reduced to general psychosocial processes and those who think that religion is somehow unique. One way to test these two possibilities is to compare religious versions of mechanisms to nonreligious versions. If religion is somehow unique, then the religious versions should explain variance in outcomes that the nonreligious versions do not. Three studies confirmed religion's independent predictive power. Convergent extension is a highly conserved process among mammals, in which the tissue narrows in one axis, and extends across another. Tissue elongation is directed by the regulation of cell interface behaviors, which guides cell intercalation and rosette formation. Rosette formation occurs through the contraction of vertically oriented cell interfaces, and the subsequent elongation of new horizontal interfaces. It has been shown that actomyosin-generated tension functions to direct rosette formation. Using the General Theory of Family Communication Patterns (FCPT) and Emotion Regulation Theory (ERT) of Meta-emotion, Parenting, and Child Outcomes, this dissertation sought to investigate adults' emotion socialization in their family of origin through an examination of multiple family schemata. Furthermore, the interactions of parental meta-emotions, family communication patterns, and family communication environments were investigated. Two surveys were used in this study. First, 228 adult-children across two universities in the western United States responded to items retrospectively describing the family communication patterns and family communication environments within their home of origin during their adolescence. Fragile X syndrome (FXS) is the most common form of inherited mental retardation in humans. FXS is caused by loss of the Fragile X Mental Retardation Protein (FMRP), an important regulator of neuronal mRNA translation. Patients with FXS display cognitive deficits including memory problems. Protein synthesis-dependent long-term changes in synaptic plasticity are involved in the establishment and maintenance of long-term memory. One prevalent theory of FXS pathology predicts that FMRP is required to negatively regulate the translation of important mRNAs at the synapse. Writing at a time when women had few property rights, were given scarce educational opportunities, and were viewed as incorrigibly irrational, the largely autodidactic English intellectual Margaret Cavendish is fascinated by knowledge and how to secure for herself a place in the micro- as well as macrocosmic community of letters. In particular, Cavendish holds an abiding interest in what we now call "consciousness" which she attributes to every piece of matter. Throughout the universe, the three aspects of matter--inanimate, sensate, and rational--are omnipresent. This thesis presents work done simulating a type of organic neuromorphic architecture, modeled after Artificial Neural Network, and termed Synthetic Neural Network, or SNN. The first major contribution of this thesis is development of a single-transistor-single-organic-bistable-device-per-input circuit that approximates behavior of an artificial neuron. The efficacy of this design is validated by comparing the behavior of a single synthetic neuron to that of an artificial neuron as well as two examples involving a network of synthetic neurons. Shyness is associated with several emotional, social, and academic problems. While there are multiple difficulties that often accompany shyness, there appear to be some factors that can moderate negative effects of shyness. Research has demonstrated that certain parenting factors affect the adjustment of shy children in early childhood, but there is minimal research illuminating the effect of parenting factors in older age groups. Acute lymphoblastic leukemia (ALL) is the most common childhood cancer and accounts for 30 - 35% of all cancers in children. Significant improvement in the treatment of pediatric ALL has been achieved in recent years. Only 50 years ago, the disease was uniformly fatal with an Overall Survival (OS) rate < 5%. Modern-day, multi-drug chemotherapy is associated with an overall survival rate over 80%. Standard-risk ALL comprises the majority of ALL with an overall survival approaching 90%. Despite this success, children who relapse from this disease accounts for the majority of cancer-related deaths in children. The backbone of treatment protocols have incorporated somatic but not host genetic features in the treatment regimens. The current study examined 12 genetic polymorphisms affecting the pharmacodynamics of antileukemic drugs in an attempt to identify biologic markers related to the risk of disease relapse.
In the current research program, 125 standard-risk ALL patients who were treated at The Children's Hospital were enrolled in a retrospective study. The United States currently has a process but no plan in implementing effective programs for rehabilitating youth offenders. The evolution of the melanocortin receptor (MCR) gene family has been dictated by two genome duplication events (2R hypothesis). The gnathostomes are thus predicted to possess a minimum of four MCR genes. Previous studies on cartilaginous fish have shown evidence for the presence of only three MCRs. The purpose of this thesis is to functionally express the MC5R from the spiny dogfish (Squalus acanthias) and the putative MC2R and MC3R from the elephant shark (Callorhinchus milii); these receptors have not yet been fully characterized. Cambodia, a least developed country (LDC) with a recent history of civil war and genocide, has been widely considered to be a success story when it comes to the fight against HIV/AIDS. CXCL16 is an interferon-inducible chemokine of the CXC-subfamily and functions as an adhesion molecule, when membrane bound, and a chemoattractant when soluble. Upregulation of cell associated CXCL16 (cCXCL16) in colorectal cancer is associated with increased tumor infiltrating lymphocytes and good prognosis. ADAM10 and ADAM17 are metalloproteinases responsible for cleaving CXCL16, releasing soluble CXCL16 (sCXCL16) and contributing to proliferation and migration of mesangial cells, in kidney inflammatory disease. We hypothesize that cCXCL16 is a substrate for ADAM10 and ADAM17 cleavage in colorectal cancer, releasing sCXCL16, which mediates cell proliferation. In the last decade, the immigrant population in Allegheny County has doubled and currently accounts for approximately seven percent of the general population. This number, despite being low, especially compared to other counties and states, is very diverse from an ethnic point view and includes immigrant and refugees from Iraq, Bhutan, Burma, Burundi, Nepal, Somalia, numerous Latin American and other countries. Such an ethnically diverse population presents new opportunities for Allegheny County, the population of which has been declining and aging, at the same time posing various challenges to local human service organizations.

Landau Kleffner Syndrome, or acquired epileptic aphasia, is an epileptic syndrome involving a neurological impairment related to the appearance of paroxysmal (i.e., sudden intense) electroencephalograph (EEG) activity (Pearl, Carrazana & Holmes, 2001). Landau Kleffner syndrome results from an epileptogenic lesion arising in the speech cortex during a critical period of development, which may interfere with the establishment of satisfactory and functional circuits for normal language function (Morrell et al., 1995). LKS is a complex and severe syndrome that affects all aspects of a child's life, including communication, socialization, and the everyday ability to function within the environment.
