In my findings, I have established a set series of protocols to recombinantly produce, purify and apply various fluorescent probes in vitro for the fluorescent labeling and study of the human immunodeficiency virus type 1 (HIV-1) envelope (Env) protein during HIV viral assembly. There remains insufficient knowledge about the molecular dynamics and interactions of HIV-1 Env protein with its counterpart, Gag, on the inner host cell surface during assembly of a mature virus particle. Criminalization measures, such as Denver’s Urban Camping Ban, are an attempt to mask systemic causes of houselessness and assign blame to the personal failures of those living on the streets. Unaccompanied houseless women utilize street families to create community, survive houselessness, and fight criminalization measures. Since liberation and solidarity can become forces of oppression, this dissertation considers cultural evidence for the desire and means of liberation. Street families and the praxis of kinship form the basis of a liberative ethic that critiques systemic causes of houselessness and provides a model of relatedness that works toward social justice. I estimate the effect of poor child health on maternal labor force participation. Mothers of health-impaired children may decide not to work and stay at home to take care of their children. Alternatively, mothers may choose to enter the labor force to pay for these children’s additional resources. Which action dominates is the empirical question I answer in this paper. I control for the potential endogeneity of a child’s health status by using an instrumental variables approach. I find that if mothers have a child in poor health, the probability that the mother works is decreased by thirteen percentage points, …

Force generation in epithelial tissues is often pulsatile, with actomyosin networks generating high-tension contractile forces at the cell cortex before cyclically disassembling. This pulsed nature of cytoskeletal forces implies that there must be cellular processes to extract unidirectional changes that drive processive transformations in cell shape. During Drosophila melanogastergastrulation, the invagination of the prospective mesoderm is driven by the pulsed constriction of apical surfaces. Here, we address the mechanisms by which the irreversibility of pulsed events is achieved while also permitting uniform epithelial behaviors to emerge. Fragile X Syndrome (FXS) is a neurodevelopmental disorder caused by the disruption of Fragile X Mental Retardation Protein (FMRP) function in neurons, affecting nearly 1 in 7,500 individuals. Although FXS typically occurs from a complete loss of FMRP expression due to a CGG trinucleotide expansion within the 5’UTR of the FMR1 gene, single nucleotide polymorphisms (SNPs) within the KH domains of FMRP have been shown to severely disrupt FMRP function. FMRP is an RNA-binding translation repressor that interacts with ~4% of the neuronal transcriptome. Vps54 is a subunit of the Golgi-associated retrograde protein (GARP) complex, which is involved in tethering endosome-derived vesicles to the trans-Golgi network (TGN). The “wobbler” mouse is the phenotypic result of a destabilizing point mutation in Vps54. This mutation causes neurodegeneration and is subsequently used as a model for human motor neuron disease. Presently, it is unclear how disruption of GARP complex function leads to motor neuron degeneration. The ubiquity of vortices nearly rivals that of the innumerable fluids and spaces in which they live. Not only do they exist in systems such as superfluids, superconductors, optical fields, or cold atomic gases, for example, but they also exist in our atmospheres, oceans, and even in our veins. This makes understanding and accurately predicting the dynamics of vortices in various systems a relevant and meaningful endeavor.

The purpose of this study has been to investigate the impact of a well-being intervention, using a well-being curriculum, known as A. B.A.L.A.N.C.E.. The method of educational criticism and connoisseurship was used as the qualitative method. Educational criticism is rooted in the process of expressing the specific qualities of one’s observations, while educational connoisseurship is rooted in the process of appreciation. The intervention will be conducted by an interventionist, myself, who gave the 10-lesson intervention to six 6th grade (11 – 12 years old) participants over a two-week period. This thesis examines the way gender expands and nuances W.E.B. DuBois’s double consciousness theory, which depicts the African American identity as a doubleness that is both American and Negro. Black feminist criticism’s nuanced formulation of DuBois’s formulation of Black identity allows the African American literary tradition to be seen through three lenses: an American, a Negro, and an African American’s gender identity. Crude oil and motor oil spills on land have severe environmental consequences that pose threats to both humans and natural resources. It is particularly important to know the flow behavior of oils to protect terrestrial ecosystems from this kind of damage. The spreading kinetics, contact angle (CA), and droplet baseline studies of crude oil and motor oils were investigated over several realistic soil-based matrices composed of topsoil (silt-dominant), sand, clay, and moisture. Three studies were undertaken to examine strategies to mitigate environmental stress faced by Northern Plains cattle feeders. In the first study, 46 feedlot managers from SD (n = 21), MN (n = 6), and NE (n = 19) were surveyed on use and perceptions of heat stress (HS) mitigation strategies. All respondents avoided cattle handling during heat stress events and incorporated some method of HS mitigation. Buildings or shades were perceived as the most successful strategy (60.9%) and were most commonly named as strategies managers would like to employ (63% for shades and 17.4% for buildings). Pollen cross-contamination has been a major problem for maize breeders. Various mechanical methods applied to avoid the contaminations are ineffective. The genetic factors related to maize fertilization can be used to develop an effective method to prevent pollen contamination. Pollen rejection ability controlled by Teosinte crossing barrier 1 (Tcb1) is such a genetic system. Silks possessing dominant Tcb1-s reject pollen possessing the recessive allele (tcb1). Successful fertilization occurs when Tcb1-s pollen falls upon tcb1 silks. The efficacy of dominant Tcb1-s was, however, reduced when repeatedly backcross with maize inbred lines, which suggests that there are modifiers to Tcb1-s. To find …

This Master thesis evaluates the impact of general health care and eating habits (Study 1), vision care (Study 2), and mental health status (Study 3) on Latino dairy farmworkers lifestyle and work performance in South Dakota. The current efficiency of conventional nitrogenous fertilizers (e.g. urea, ammonium sulfate, etc.) in agricultural practices is low, fluctuating from 30 to 40%. Approximately 60% of nitrogenous fertilizers were wasted due to vaporization into the air, runoff or leaching into water systems. Soybean leaf-associated gemycircularvirus-1 (SlaGemV-1) is a novel mycovirus discovered through the metagenomic sequencing of soybean leaves which is capable of inducing hypovirulence in the highly pathogenic fungus Sclerotinia sclerotiorum. RNASeq analysis techniques were used to determine the transcriptional changes caused by the infection of virus in S. sclerotiorum, as well as the transcriptional changes in Glycine max caused by the colonization of hypovirulent, SlaGemV-1-infected S. sclerotiorum. RNASeq results indicate that viral infection leading to hypovirulence may attenuate expression of genes relating to cell wall synthesis, microtubule formation, and metabolism of steroids and natural antibiotics. Metal 3D printing has been reserved for aerospace and high-end automotive industries because of its cost. A gas metal arc welder (GMAW) on a rugged 3D printer frame could make metal additive manufacturing an option for more industries and consumers. 3D welded aluminum has not been examined in depth as an option for additive manufacturing (AM). Extensive tests are necessary to determine the correct settings to use a metal inert gas (MIG) welder for AM. Volatile organic compounds (VOCs) are abundant in nature and can occur in various forms in a wide variety of sources. Petroleum is one of the major sources of VOCs. Petroleum being the most widely used energy resources of the world, often released to the environment due to emission and leakage or accidental spillage during exploration, transportation and manufacturing. Once released to the environment, petroleum poses a serious threat to the environment due to some of its toxic organic components.
